The paper studies the effect of additional administration of ultrafine particles on the cattle rumen microbiome composition. The in vitro method was used using the ANKOM Daisy II incubator according to a specialized method. Microflora analysis was performed using MiSeq (Illumina, USA) by a new generation sequencing method with a MiSeq reagent kit. After a detailed analysis of the structure and composition of the microbial community in the contents of the rumen sampled for different diets, it was found that no significant differences were observed in the bacterial communities, with the exception of a slight shift in the Bacteroidetes/Firmicutes ratio. However, we observed numerical differences in the abundance of some representatives, namely, with additional inclusion of Fe and Cr2O3, decrease in the abundance of the methane-forming species Methanobrevibacter, Methanobacterium, Methanosphaera, and Methnaomicrobium was noted regarding the control.

Molecular phylogenetic analysis of the marker sequences ITS and trnL–trnF of the Phragmites australis,P. altissimus, some putative hybrids between them and the sample of P. australis s. l. from Republic of Tuva was performed.ITS1 sequences of the sample from Tuva were studied by the New-generation sequencing method (NGS) revealingintragenomic polymorphism of the ITS sequences in allopolyploid species. We showed good differences betweenP. australis and P. altissimus in the ITS1-5.8S rDNA-ITS2 sequences by many single-nucleotide substitutions anddeletions, in particular, there is a 12 b.p. deletion. P. australis s. l. sample from Tuva (Hemchik river) demonstrates aseparate position on the phylogenetic tree. All reed species studied by us do not have significant differences between eachother according to the chloroplast sequences trnL–trnF.

Phytoplasmas are phytopathogenic prokaryotes of great importance because they have been linked to numerous diseases around the world. The aim of these research was to present the general characteristics, the size and composition of the genome and the genes and/or regions used as molecular markers for the identification and characterization of phytoplasmas. Among its main hosts are fruit and wood trees, vegetables, cut flowers and weeds, which generate alterations in the hormonal balance producing symptoms such as philodias and virescence. Because its isolation in vitro has not been possible, the detection and characterization has been carried out, mainly, with molecular techniques. In addition, the use of New Generation Sequencing (NGS) has allowed to know the complete genome of some phytoplasmas, as well as the regions and genes that constitute it. In the present bibliographic review, the information generated from the molecular techniques and NGS sequencing is compiled, as well as the primers reported to identify some groups of phytoplasmas.

Целью исследования было определение таксономической структуры микробиома кишечника крупного рогатого скота породы Абердин-Ангус с помощью технологии секвенирования нового поколения. 16S метагеномный анализ, позволил определить микробный состав содержимого кишечника, минуя стадию культивирования на питательных средах. Проведена генетическая идентификация и получен таксономический профиль всех присутствующих бактерий, в том числе и некультивируемых форм. The aim of the study was to determine the taxonomic structure of the intestinal microbiome of Aberdeen Angus cattle using a new generation sequencing technology. 16S metagenomic analysis made it possible to determine the microbial composition of the intestinal contents bypassing the stage of cultivation on nutrient media. Genetic identification was carried out and a taxonomic profile of all bacteria present, including non-cultivated forms, was obtained. This chapter illustrates how genomics and other -omics approaches coupled with new-generation sequencing (NGS) platforms have been deployed to dissect the genetic make-up of RSA traits and better understand their functions, particularly under environmentally constrained conditions that commonly occur in most farmed soils. The major emphasis is devoted to studies during the past two decades in crops and only occasional reference is provided to the vast literature from RSA studies conducted in Arabidopsis and other model plants. The chapter also provides examples on how, in some cases, this knowledge is already benefiting farmers and how it can help in reducing the environmental impact of agriculture worldwide.

We report the complete genome sequence of the first Mexican human coronavirus (HCoV) OC43, obtained by new-generation sequencing and a metagenomic approach, isolated from a child hospitalized with pneumonia. The genome is closely related to the other OC43 genome sequences available, ranging from 99.8% to 98.2% nucleotide sequence identity.
