AbstractThe Pantanal wetland harbors a rich flora with uncharted pharmacological potential. This study evaluated 20 Brazilian Pantanal plants against Candida albicans, C. parapsilosis, C. tropicalis, and C. krusei. Fungal susceptibility was determined by agar diffusion and broth microdilution; active compounds were identified by bioautography and HPLC-DAD-MS/MS. Sesbania virgata, Polygala molluginifolia, and Cantinoa mutabilis extracts and their chloroform and ethyl acetate (EtOAc) fractions exhibited the best activity against all Candida species tested. The EtOAc fraction of P. molluginifolia proved to be more efficient in inhibiting C. parapsilosis and C. krusei growth (Minimum inhibitory concentration of 125 and 62.5 μg/mL, respectively). Bioautography of this fraction revealed two active bands, characterized by HPLC-DAD-MS/MS as a mixture of podophyllotoxin derivatives blumenol, besides some flavonoids. This work demonstrated antifungal potential of P. molluginifolia podophyllotoxin derivatives and the versatility of bioautography with HPLC-DAD-MS/MS to identify the bioactive compounds.

This is a review of current studies in diversity of the flora and main vegetation types in the Brazilian Pantanal. The flora of this wetland, nearly 2,000 species, constitutes a pool of elements of wide distribution and from more or less adjacent phytogeographic provinces, such as Cerrado, dry seasonal forests, Chaco, Amazonia and Atlantic Forest. The most numerous group includes wide-distribution species, mainly herbs, while the second contingent comes from the Cerrado. Endemic plants are rare, numbering only seven. The vegetation of the sedimentary floodplain is a mosaic of aquatics, floodable grasslands, riparian forests, savannas (cerrados), cerrado woodlands, dry forests, and a large area of mono-dominant savannas, and pioneer woodlands. The main vegetation types are briefly described with their characteristic species, and their estimated areas are given according to the latest mapping.

A library of six novel phenylhydrazones were synthesized and evaluated for their in vitro antimicrobial and resistance modulating activity against a panel of Gram-positive, Gram-negative, and fungal species. The compounds were produced in good yields of 60–92% w/w and characterized using melting point, UV-visible spectroscopy, infrared, and nuclear magnetic resonance (1H, 13C, and DEPT-Q) techniques. Mass spectroscopy was used to confirm the identity of one of the most active compounds, 5 [SA5]. The phenylhydrazones showed activity against all the six selected microorganisms with minimum inhibitory concentration (MIC) values of the most active compounds, 1 [BP1] and 5 [SA5], at 138 µM (Klebsiella pneumoniae) and 165 µM (Streptococcus pneumoniae), respectively. Compound 1 [BP1] further demonstrated a high resistance modulatory activity at 1.078 µM against Streptococcus pneumoniae and Klebsiella pneumoniae.

The Brazilian Pantanal wetland undergoes landscape alterations that can cause impacts on hydrological processes, changing the flood pulse. The objective of this work is to analyse the vegetation cover of the Pantanal in the period of 2000, 2008 and 2015, and to make a projection for 2030. Therefore, NDVI from the sensor MODIS was analysed and the transition matrix was calculated by the DINAMICA EGO. The methods adopted were open sources. The results were worrisome, indicating alterations of the vegetation cover of the Pantanal, with an increase of short vegetation (grasslands or pastures) in the evaluated period. The projection pointed out that in 2030 the Brazilian Pantanal wetland area will be covered by 78% of short vegetation and only 14% of dense (arboreal-shrubby) vegetation. The approach can be a useful tool for conservation of the Brazilian Pantanal wetland.

Objective: Present study evaluates the antifungal potential of Allium ampeloprasum extracts against two phytopathogens of economic importance, namely Alternaria triticina and Magnaporthe oryzae which are found to be one of the greatest threats to wheat and rice crop worldwide.Methods: Four different organic solvents, i.e., water, methanol, acetone, and hexane were used for extract preparation based on the increasing order of polarity. Further, different concentrations of these extracts were used to perform a quantitative assay for measuring the antifungal activity of extracts. Two-way analysis of variance was used to statistically analyze the results through GraphPad Prism v. 7.00.Results: The best results were shown by water extracts with a maximum inhibition of 99.39±0.2% against A. triticina and 99.39±2.2% against M. oryzae. Water extracts gave minimal inhibitory concentration 90 values at 9.94 μg/mL against M. oryzae and at 16.73 μg/mL against A. triticina.Conclusion: Water extracts of A. ampeloprasum have relatively higher potential for being used as an antifungal agent as compared to other organic extracts used in this study. Further study on the chemical composition of A. ampeloprasum water extracts can reveal potent anti-phytopathogenic fungal compounds which can be used to develop biofungicides.

The treatment of benzylidenemalononitriles 3a-c with phenylhydrazines 4a-n in refluxing ethanol did not provide pyrazole derivatives but furnished hydrazones 1a-o. The structure of hydrazones was secured by X-Ray analysis. Newly synthesized hydrazones 1a-o were tested against 8 Candida spp. strains in a dose response assay to determine the minimum inhibitory concentration (MIC99). Five compounds 1c, 1d, 1i, 1k and 1l were identified as promising antifungal agents against Candida spp. (C. albicans SC5314, C. glabrata, C. tropicalis, C. parapsilosis and C. glabrata (R azoles)) with MIC99 values ranging from 16 to 32 µg/mL. To further evaluate the antifungal potential of the active compounds, they have been assayed against a mammalian cell line HEK293 to determine general cell toxicity and on NCI-60 cancer cell lines panel, demonstrating selectivity antifungal activity over cytotoxicity.

Osteogenesis activity of fractions extracted from Clinacanthus nutans was evaluated on an in vitro model using osteoblast MC3T3-E1 cells. The results showed that all fractions, including ethanol (EtOH), n-hexane, ethyl acetate (EtOAc) and butanol (BuOH) were not significantly toxic to the osteoblast cells at the test concentrations of 5, 10, 25, and 50 µg/mL. The EtOH and EtOAc fractions exhibited the highest osteogenesis activity in terms of enhancement of alkaline phosphatase (ALP) and mineralization activity of MC3T3-E1 cells, especially the EtOAc fraction which increased ALP activity up to > 30% and mineralization activity up to > 100%. Thus, the EtOAc fraction shows osteogenesis activity through stimulating activites of the two markers for bone generation including ALP and mineralization in osteoblast cells. The fraction is now under extensive investigation to isolate and fully understand the modes of action of the active compounds.
