Cellular adaptation to stress and metabolic cues requires a coordinated response of different intracellular compartments, separated by semipermeable membranes. One way to facilitate interorganellar communication is via membrane contact sites, physical bridges between opposing organellar membranes formed by an array of tethering machineries. These contact sites are highly dynamic and establish an interconnected organellar network able to quickly respond to external and internal stress by changing size, abundance and molecular architecture. Here, we discuss recent work on nucleus-vacuole junctions, connecting yeast vacuoles with the nucleus. Appearing as small, single foci in mitotic cells, these contacts expand into one enlarged patch upon nutrient exhaustion and entry into quiescence or can be shaped into multiple large foci essential to sustain viability upon proteostatic stress at the nuclear envelope. We highlight the remarkable plasticity and rapid remodelling of these contact sites upon metabolic or proteostatic stress and their emerging importance for cellular fitness.

Flooding and drought are major causes of reductions in crop productivity. Root distribution indicates crop adaptation to water stress. Therefore, we aimed to identify crop roots response based on root distribution under various soil conditions. The root distribution of four crops—maize, millet, sorghum, and rice—was evaluated under continuous soil waterlogging (CSW), moderate soil moisture (MSM), and gradual soil drying (GSD) conditions. Roots extended largely to the shallow soil layer in CSW and grew longer to the deeper soil layer in GSD in maize and sorghum. GSD tended to promote the root and shoot biomass across soil moisture status regardless of the crop species. The change of specific root density in rice and millet was small compared with maize and sorghum between different soil moisture statuses. Crop response in shoot and root biomass to various soil moisture status was highest in maize and lowest in rice among the tested crops as per the regression coefficient. Thus, we describe different root distributions associated with crop plasticity, which signify root spread changes, depending on soil water conditions in different crop genotypes as well as root distributions that vary depending on crop adaptation from anaerobic to aerobic conditions.

Nowadays we define Giants as persons suffering from scientifically defined physiological disorders. Since Western culture has a short memory for obsolete scientific discourse, however, the simplicity of our contemporary understanding of gigantism makes it difficult for us to understand previous attitudes toward Giants, especially as expressed in literature. As Donald Frame has remarked, ‘When most Western readers think of giants in literature, they think of Rabelais and Swift; when they think of Rabelais and Swift, they think of giants.’ However, the actual importance of Rabelais and Swift would be seriously misrepresented were we to imagine them as exponents of the traditional Western attitude toward Giants and gigantism. What is more, Rabelais, who is the source of most early modern speculation about Giants, is a particularly problematic case. His combination of agile parodic wit and extreme philosophical and theological literacy is only beginning to be satisfactorily understood. Thus his treatment of gigantological themes has until now been almost completely misrepresented because of an insufficient understanding of the cultural significance of gigantism before his time. In fact, he is at least two removes from a coherent tradition of gigantological discourse running from the Old Testament through Judaic and patristic commentary and historiography, straight into the era of humanism. While the scope of this article will not permit an intensive analysis of Rabelais' own gigantology, an analysis of the two traditions upon which he depended will implicitly demonstrate the inadequacy of the conventional wisdom which sees Pantagruel and Gargantua as a direct outgrowth of medieval French folklore, the Grandes chronicques, and the literary romances of Pulci and Folengo.
