Noninvasive DNA sampling and camera trapping suggest dramatic regional conservation status of an understudied carnivore: the Rüppell’s fox in Tunisia

Purpose: To recruit healthy full- and preterm infants into genetic research and determine the effectiveness of a noninvasive DNA sampling technique for comparing epigenetic modifications. Background: Noxious stimuli during a vulnerable period of infant neuronal plasticity may trigger long-term epigenetic changes affecting neurodevelopment, pain modulation, and reactivity. Recognizing epigenetic pain findings is problematic because parents are reluctant to enroll newborns into genetic research. Methods: Design: Within-subject change over time candidate-gene DNA methylation association study. Setting/ sample: Urban teaching hospital’s neonatal intensive care unit and newborn nursery. Convenience sample of healthy full- (>37 weeks, n = 6) and preterm (<37 weeks, n = 6) infants. Procedure: Parents participated in a genetic presentation prior to informed consent. Infant buccal saliva was collected after admission to the unit and prior to discharge. Analysis: The methylation pattern at the 5′ end of µ-opioid receptor gene ( OPRM1) was examined. DNA was treated with bisulfite to convert all cytosines to uracil residues, leaving methylated cytosines unchanged. Sequencing of untreated and bisulfite-converted DNA was carried out. The sequences of unconverted and bisulfite-converted DNA were aligned with ClustalW, fidelity of the polymerase chain reaction and the sequencing reaction evaluated, and the methylation pattern identified. Results: Recruitment and assessment of a noninvasive DNA sampling technique for comparing epigenetic modifications were successful; however, infant stress did not produce a change in OPRM1 methylation expression. Relevance: This study established the feasibility of recruiting healthy full-term infants into genetic research and the effectiveness of noninvasive DNA sampling for comparing epigenetic modification in infants.

A new locality for Ophiodes intermedius is reported for Uruguay. The new record at Punta Gorda, Departamento Colonia, is the first departmental record, and extends the known distribution in Uruguay approximately 150 km to the south. This taxon is restricted to the fast developing western littoral of Uruguay, and a frequent assessment of its regional conservation status is recommended.

The northern hopping-mouse (Notomys aquilo) is a cryptic and enigmatic rodent endemic to Australia’s monsoonal tropics. Focusing on the insular population on Groote Eylandt, Northern Territory, we present the first study to successfully use live traps, camera traps and radio-tracking to document the ecology of N. aquilo. Searches for signs of the species, camera trapping, pitfall trapping and spotlighting were conducted across the island during 2012–15. These methods detected the species in three of the 32 locations surveyed. Pitfall traps captured 39 individuals over 7917 trap-nights. Females were significantly longer and heavier, and had better body condition, than males. Breeding occurred throughout the year; however, the greatest influx of juveniles into the population occurred early in the dry season in June and July. Nine individuals radio-tracked in woodland habitat utilised discrete home ranges of 0.39–23.95 ha. All individuals used open microhabitat proportionally more than was available, and there was a strong preference for eucalypt woodland on sandy substrate rather than for adjacent sandstone woodland or acacia shrubland. Camera trapping was more effective than live trapping at estimating abundance and, with the lower effort required to employ this technique, it is recommended for future sampling of the species. Groote Eylandt possibly contains the last populations of N. aquilo, but even there its abundance and distribution have decreased dramatically in surveys over the last several decades. Therefore, we recommend that the species’ conservation status under the Environment Protection and Biodiversity Conservation Act 1999 be changed from ‘vulnerable’ to ‘endangered’.

AbstractCamera-trapping and capture-recapture models are the most widely used tools for estimating densities of wild felids that have unique coat patterns, such as Eurasian lynx. However, studies dealing with this species are predominantly on a short-term basis and our knowledge of temporal trends and population persistence is still scarce. By using systematic camera-trapping and spatial capture-recapture models, we estimated lynx densities and evaluated density fluctuations, apparent survival, transition rate and individual's turnover during five consecutive seasons at three different sites situated in the Czech–Slovak–Polish borderland at the periphery of the Western Carpathians. Our density estimates vary between 0.26 and 1.85 lynx/100 km2 suitable habitat and represent the lowest and the highest lynx densities reported from the Carpathians. We recorded 1.5–4.1-fold changes in asynchronous fluctuated densities among all study sites and seasons. Furthermore, we detected high individual’s turnover (on average 46.3 ± 8.06% in all independent lynx and 37.6 ± 4.22% in adults) as well as low persistence of adults (only 3 out of 29 individuals detected in all seasons). The overall apparent survival rate was 0.63 ± 0.055 and overall transition rate between sites was 0.03 ± 0.019. Transition rate of males was significantly higher than in females, suggesting male-biased dispersal and female philopatry. Fluctuating densities and high turnover rates, in combination with documented lynx mortality, indicate that the population in our region faces several human-induced mortalities, such as poaching or lynx-vehicle collisions. These factors might restrict population growth and limit the dispersion of lynx to other subsequent areas, thus undermining the favourable conservation status of the Carpathian population. Moreover, our study demonstrates that long-term camera-trapping surveys are needed for evaluation of population trends and for reliable estimates of demographic parameters of wild territorial felids, and can be further used for establishing successful management and conservation measures.
