This chapter introduces the concept of formulation development assisted by computer applications. Development and optimization of various types of pharmaceutical emulsions microemulsions, self- microemulsifying systems, and double emulsions are presented. Illustrative examples are presented to demonstrate the ability of computer-aided tools to facilitate formulation development. Various techniques, such as design of experiments and arti?cial neural networks, are implemented for optimization of the formulation and/or processing parameters. Furthermore, some of the critical quality attributes and processing parameters are optimized simultaneously. The examples presented should serve as the foundation for the future quality- by-design development of pharmaceutical emulsion and (self) microemulsion formulations.

A strategy based on sequential design of experiments (screening, optimization and confirmation) was used to develop a tablet formulation of ibuprofen (400 mg) that is manufactured by direct compression. This formulation has a high content of ibuprofen (76%), in spite of the poor flowability of the drug substance. Sequential design of experiments proved to be an effective and efficient strategy in formulation development.

Zespół metod wykorzystywanych do planowania eksperymentów określa się jako Design of Experiments -DOE. Celem stosowania DOE jest uzyskanie jak największej ilości wartościowych i wiarygodnych informacji o badanym wyrobie lub procesie na podstawie jak najmniejszej liczby doświadczeń. W artykule zwrócono uwagę na stosunkowo mało znane metody opracowane przez Shainina oraz Taguchiego. Przedstawiono dobór parametrów metodą Shainina i Taguchiego na przykładzie procesu toczenia.
