Susceptibility of nymphs and adults of Diaphorina citri to the entomophathogenic fungus Hirsutella citriformis

Background Among entomopathogenic fungi, H. citriformis has been recognized as potential biocontrol agent against the Asian citrus psyllid Diaphorina citri (Hemiptera: Liviidae). Nevertheless, this fungus is poorly characterized. Previous molecular studies have shown high sequence similarities among strains, but significant differences in Diaphorina citri virulence. Objective The aim of the present study was to determine mycelial compatibility and anastomosis, and nucleus numbers in mycelium and conidia of eight H. citriformis strains isolated from mycosed D. citri adults collected from several Mexican states. Methods Mycelial compatibility and anastomosis evaluation was performed after pairing strains, leading to 36 confrontations, and cultured in chlorate minimum medium to obtain mutants for vegetative compatibility group. Results Hypha or conidia nuclei were visualized with safranin-O and 3% KOH, and 0.05% trypan blue–lactophenol solution. H. citriformis strains showed compatibly and anastomosis events after confrontation. In addition, they showed one nucleus per conidium and mycelium section. It was not possible to obtain H. citriformis nit mutants from the chlorate concentrations tested. Conclusions To date, this is the first report demonstrating mycelial compatibility, anastomosis occurrence, and hyphae and conidia nuclei number among H. citriformis strains.

Pemanfaatan cendawan entomopatogen Hirsutella citriformis dalam mengendalikan hama utama tanaman jeruk Diaphorina citri mempunyai arti penting dalam mendukung konsep pengendalian hama terpadu. Entomopatogen dapat mengendalikan imago Diaphorina citri pada konsentrasi 108 konidia/ml, terutama pada musim penghujan. Aplikasi pestisida (insektisida dan fungisida) di lapang  dilakukan oleh petani diduga berpengaruh terhadap H.citriformis belum pernah diteliti. Tujuan penelitian ini adalah mengetahui pengaruh insektisida dan fungisida terhadap perkembangan H.citriformis secara in vitro dan in vivo. Penelitian dilaksanakan di Laboratorium Terpadu dan Rumah Kasa Balai Penelitian Tanaman Jeruk dan Buah Subtropika, Tlekung, mulai Juni 2008 sampai dengan Juni 2009. Penelitian in vitro, menggunakan rancangan acak lengkap dengan sembilan perlakuan, yaitu insektisida berbahan aktif abamektin, sipermetrin, profenofos, dimetoat serta fungisida berbahan aktif  propinep, mankozeb, benomil, bupirimat serta kontrol, dengan tiga ulangan. Penelitian in vivo,  dilakukan pada benih jeruk menggunakan rancangan acak kelompok dengan tiga perlakuan: satu macam insektisida  dan fungisida  terbaik dari hasil penelitian in vitro yaitu yang tidak menekan pertumbuhan H citriformis dan kontrol, masing-masing dengan lima ulangan.  Parameter pengamatan in vitro meliputi diameter koloni H.citriformis dan persentase tingkat hambatan, sedang pengamatan in vivo di rumah kasa  meliputi periode inkubasi pada D.citri dan jumlah imago terinfeksi yang mati. Hasil penelitian laboratorium menunjukkan bahwa  H. citriformis dapat tumbuh cukup optimal pada media buatan yang dicampur dengan insektisida abamektin, sipermetrin, dan profenofos serta fungisida berbahan aktif propinep dan mankozeb. Hasil penelitian di rumah kasa menunjukkan bahwa insektisida profenofos lebih kompatibel disemprot bersama-sama dengan H.citriformis dibanding fungisida bupirimat dalam mengendalikan serangga imago D.citri

Objective: To calculate the monthly relative abundance of Thysanoptera species, according to the Margalef, Simpson and Shannon-Wiener diversity indices. Design / Methodology / Approach: The work was carried out in three geographic units with conventional management, during January-December, 2019 in the Reserva de la Biosfera Zicuirán-Infiernillo (Biosphere Reserve), Michoacán, Mexico. In each unit, 10 trees were selected through simple random sampling. Thrips counts were performed on ten shoots per tree every 15 d, for a total of 7200 shoots in the three geographic units. Thysanoptera individuals were placed in entomological jars. The variables were: number of thrips collected per shoot in sampled tree and geographic unit (orchard). To estimate the specific richness and structure of species, the program "calculation of diversity indices DIVERS" was used. Results: In the three geographical units studied, the recorded presence of Thysanoptera accounted for 12 to 17 species. For Nueva Italia 12 recorded species, two were permanent (16.66%), five abundant (41.66%), one scarce (8.3%) and four rare (33.33%). In Zicuirán, three species were permanent (17.64%), six abundant (35.29%), two scarce (11.76%) and six rare (35.29%). In Los Hoyos, four species were permanent (26.66%), four abundant (26.66%) and seven rare (46.66%). The abundance of species was represented by the genus Frankliniella and the species Scolothrips sexmaculatus and Scirtothrips citri. The highest species richness and abundance was found from January to May. In October and November, the value of the calculated indices was zero, which shows less richness and abundance of individuals. The best species uniformity was recorded during January and December, which meant a more stable and homogeneous relation. Study limitations/Implications. Pest resurgence, presence of Candidatus Liberibacter spp. and its vector Diaphorina citri. Findings / Conclusions: in Nueva Italia, 12 species were taxonomically determined; in Los Hoyos 15, and in Zicuirán 17 species, which are reported for the first time in the state of Michoacán, Mexico. At the geographic unit "Los Hoyos" diversity was higher, uniform and stable.
