A Selective Medium for the Recovery and Enumeration of Fomitopsis meliae, Causing Lemon Canker and Wood Rot

A selective medium (FMS medium) was developed for the isolation and quantification of Fomitopsis meliae, the causal agent of lemon canker and brown wood rot, from plants, soil, and air. The optimal concentration and combination of fungicides and antibiotics was evaluated to determine the most selective condition for growing F. meliae. The resultant composition of the medium (FMS) per litre (pH 3.5) was: 16 mg thiophanate-methyl, 8 mg dichloran, 5 mg 2-phenylphenol, 100 mg fluopyram, 0.5 mg fludioxonil, 100 mg chloramphenicol, 100 mg streptomycin, 15 g malt extract, 2.5 g mycological peptone, and 15 g agar. The fungus was successfully isolated and enumerated from air, soil and plant tissues using FMS medium. Furthermore, FMS medium almost completely inhibited the growth of other plant pathogenic fungi, soil and air saprophytes. This selectivity was high enough to estimate spore inoculum of F. meliae in an air sample or as a spore trapping device in commercial lemon orchards. FMS medium will be useful for studying epidemiology and management of F. meliae.

Fungal phytotoxins are natural secondary metabolites produced by plant pathogenic fungi during host–pathogen interactions. They have received considerable particular attention for elucidating disease etiology, and consequently to design strategies for disease control. Due to wide differences in their chemical structures, these toxic metabolites have different ecological and environmental roles and mechanisms of action. This review aims at summarizing the studies on the possible use of these metabolites as tools in biological and integrated weed management, e.g. as: novel and environmentally friendly herbicides; lead for novel compounds; sources of novel mechanisms of action. Moreover, the limiting factors for utilizing those metabolites in practice will also be briefly discussed.

This study was carried out to identify the unknown different symptoms and their causes as plant pathogenic fungi from Al-Jabal Al-Akhdar District. Plant materials with fungal signs and symptoms were collected and examined. The main fungi consistently isolated from symptomatic leaves and twigs were Pestalotiopsis spp. Morphology, colony characteristics, and pathogenicity of the isolates were examined. My report the occurrence of Pestalotiopsis spp. on leaves of mastic (Pistacia lentiscus) for the first time in Libya.

To develop efficient control measures against fungal plant pathogens, the dynamics of host plant colonization during disease development and the interactions among fungi within host plant tissues need to be clarified. These studies require accurate quantitative estimation of specific fungal biomass in plant tissues. This has been approached by direct-microscopic methods, cultural methods, chemical determinations of fungal components, serological methods, and molecular methods. Among these methods, serological and molecular methods provide rapid, specific, and sensitive quantitative measures of fungal biomass in host plant tissues. Therefore, studies on fungal dynamics of host plant colonization using these two methods are presented. Some examples of species interactions among pathogenic fungi within host plants, such as synergism and competition, are reviewed and the usefulness of serological and molecular methods for studies on these interactions is presented. These quantitative methods will provide helpful information for understanding the ecology of plant pathogenic fungi, such as the dynamics of host plant colonization and species interactions.
