Janibacter cremeus sp. nov., an actinobacterium isolated from sea sediment

A novel Gram-stain-positive actinobacterium, designated HR08-44T, was isolated from a sea-sediment sample collected from the foreshore of Rishiri Island, Japan, and its taxonomic position was investigated by a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequence comparisons revealed that strain HR08-44T was closely related to the members of the genus Janibacter , with pairwise sequence similarities of 97.3–98.8 %. Strain HR08-44T had peptidoglycan type A1γ, with meso-diaminopimelic acid as the diagnostic diamino acid. The predominant menaquinone was MK-8(H4) and the major fatty acids were iso-C16 : 0, C17 : 1ω8c, C17 : 0 and C18 : 1ω9c. These data supported the affiliation of the novel strain to the genus Janibacter . Meanwhile, the results of DNA–DNA hybridization and physiological and biochemical tests indicated that strain HR08-44T can be distinguished from recognized species of the genus Janibacter . Therefore, strain HR08-44T represents a novel species of the genus Janibacter , for which the name Janibacter cremeus sp. nov. is proposed; the type strain is HR08-44T ( = NBRC 107693T = DSM 26154T).

A Gram-stain-negative, aerobic, rod-shaped and motile bacterium, designated THG-DD7T, was isolated from tomato plant rhizosphere soil. Strain THG-DD7T grew optimally at 25–30 °C, at pH 7.0–7.5 and in the presence of 0.5 % (w/v) NaCl. According to the results of 16S rRNA gene sequence comparisons, strain THG-DD7T was most closely related to Rhodanobacter umsongensis GR24-2T (98.2 %), Rhodanobacter panaciterrae LnR5-47T (98.0 %), Rhodanobacter soli DCY45T (97.9 %), Rhodanobacter terrae GP18-1T (97.9 %) and Dyella ginsengisoli Gsoil 3046T (97.7 %). The DNA G+C content was 65.2 mol%. In DNA–DNA hybridization, the DNA relatedness levels between strain THG-DD7T and its closest phylogenetically neighbours were below 40.0 %. The predominant isoprenoid quinone was ubiquinone Q-8. The major polar lipids were diphosphatidylglycerol, phosphtidylethanolamine, phosphatidyl-N-methylethanolamine and phosphatidylglycerol. The major fatty acids were iso-C15 : 0, iso-C16 : 0, iso-C17 : 0, anteiso-C15 : 0 and iso-C17 : 1ω9c and/or C16 : 0 10-methyl (summed feature 9). These data supported the affiliation of strain THG-DD7T to the genus Rhodanobacter . The results of physiological and biochemical tests enabled strain THG-DD7T to be differentiated genotypically and phenotypically from the species of the genus Rhodanobacter with validly published names. Therefore, the novel isolate represents a novel species, for which the name Rhodanobacter koreensis sp. nov. is proposed. The type strain is THG-DD7T ( = KACC 17650T = JCM 19614T).

A novel actinomycete strain, designated YIM 75904T, was isolated from a soil sample that had been collected from a dry and hot river valley in Dongchuan county, Yunnan province, south-western China. The taxonomic position of the novel strain was investigated by a polyphasic approach. In phylogenetic analyses based on 16S rRNA gene sequences, strain YIM 75904T formed a distinct clade within the genus Amycolatopsis and appeared to be closely related to Amycolatopsis sacchari K24T (99.3 % sequence similarity). Strain YIM 75904T had a type-IV cell wall, with no detectable mycolic acids, and had MK-9(H4) as its predominant menaquonine. Its cell wall contained meso-diaminopimelic acid, galactose, glucose and arabinose, and its major cellular fatty acids were iso-C16 : 0, iso-C15 : 0, anteiso-C17 : 0 and anteiso-C15 : 0. The genomic DNA G+C content of the novel strain was 68.5 mol%. Based on the results of physiological and biochemical tests and DNA–DNA hybridizations, strain YIM 75904T represents a novel species of the genus Amycolatopsis for which the name Amycolatopsis dongchuanensis sp. nov. is proposed. The type strain is YIM 75904T ( = CCTCC AA 2011016T  = JCM 18054T).

A Gram-positive, non-spore-forming bacterium (GW-12028T) of unknown origin showing filamentous growth and producing spherical sporangia was studied for its taxonomic allocation. The 16S rRNA gene sequence analysis and subsequent similarity studies showed that strain GW-12028T belongs to the genus Streptosporangium , and is most closely related to Streptosporangium pseudovulgare DSM 43181T (99.9 %) and Streptosporangium nondiastaticum DSM 43848T (99.6 %) and more distantly related to Streptosporangium fragile IFO 14311T (98.4 %) and other species of the genus Streptosporangium (95.8 to 98 %). Chemotaxonomic analyses showed that the peptidoglycan diamino acid was meso-diaminopimelic acid. Whole-cell hydrolysates contained madurose as the diagnostic sugar and exhibited a quinone system that contained predominantly menaquinones with nine isoprenoic units in the side chain [MK-9, MK-9(H2), MK-9(H4)]. The polar lipid profile consisted of diphosphatidylglycerol, phosphatidylglycerol, phoshatidylethanolamine, hydroxyphosphatidylethanolamine, phosphatidylinositol, phosphatidylinositol-mannosides, four unidentified glycolipids, a phospholipid and an aminolipid. The polyamine pattern contained the major compounds spermine and spermidine. The major fatty acids were 10-methyl C17 : 0, iso-C16 : 0 and C15 : 0. These chemotaxonomic traits are common to other species of the genus Streptosporangium . DNA–DNA hybridizations and physiological and biochemical tests in comparison with the type strains of the most closely related species, S. nondiastaticum and S. pseudovulgare , allowed genotypic and phenotypic differentiation of strain GW-12028T. This strain represents a novel species, for which we propose the name Streptosporangium sandarakinum sp. nov., with the type strain GW-12028T ( = LMG 27062T  = DSM 45763T).

A novel actinomycete, designated strain NEAU-FHS4T, was isolated from the root of black false hellebore (Veratrum nigrum L.). Strain NEAU-FHS4T formed single spores with smooth surfaces on substrate mycelium. The novel strain contained meso-diaminopimelic as amino acid of the peptidoglycan and xylose and glucose as whole-cell sugars. The predominant menaquinones were MK-10(H6) and MK-10(H8). Mycolic acids were not detected. The diagnostic phospholipids were phosphatidylethanolamine, diphosphatidylglycerol and phosphatidylinositol. The predominant cellular fatty acids were iso-C16 : 0, C16 : 0, C18 : 0 and anteiso-C17 : 0. Phenotypic and chemotaxonomic analysis showed that the novel isolate had characteristics typical of members of the genus Plantactinospora . 16S rRNA gene sequence analysis also indicated that strain NEAU-FHS4T belonged to the genus Plantactinospora , with highest sequence similarities to Plantactinospora mayteni YIM 61359T (98.88 %) and Plantactinospora endophytica YIM 68255T (98.85 %). The results of DNA–DNA hybridization and physiological and biochemical tests allowed genotypic and phenotypic differentiation of the novel strain from the most closely related strains. Based on morphological, chemotaxonomic and phylogenetic data, strain NEAU-FHS4T is considered to represent a novel species of the genus Plantactinospora , for which the name Plantactinospora veratri sp. nov. is proposed. The type strain is NEAU-FHS4T ( = CGMCC 4.7143T = DSM 46718T).

A Gram-staining-positive, aerobic and non-motile strain, 0704P10-1T, was isolated from hydrothermal sediment of the Indian Ocean. Phylogenetic, phenotypic and chemotaxonomic data for the organism supported that it belonged to the genus Janibacter . Strain 0704P10-1T showed 97.2–98.7 % 16S rRNA gene sequence similarities to the type strains of recognized members of the genus Janibacter . It contained meso-diaminopimelic acid as the diagnostic diamino acid in the cell wall. MK-8(H4) was the only menaquinone detected. The major fatty acids were iso-C16 : 0, C17 : 1ω8c and 10-methyl C17 : 0. Meanwhile, the results of DNA–DNA hybridization studies and other physiological and biochemical tests allowed the genotypic and phenotypic differentiation of strain 0704P10-1T from closely related species. Thus, strain 0704P10-1T represents a novel species of the genus Janibacter , for which the name Janibacter indicus sp. nov. is proposed. The type strain is 0704P10-1T ( = LMG 27493T = CGMCC 1.12511T).

A Gram-stain-positive, novel actinobacterium, designated strain JL-6T, was isolated from the litter of a bamboo (Sasa borealis) forest in Damyang, Korea. Strain JL-6T had white‐grey, smooth, cylindrical spores that were borne in straight, long spore-chains. The novel strain grew aerobically at 15–28 °C (optimum, 28 °C), pH 4.0–8.0 (optimum, pH 5.5) and with 0–1.5 % (w/v) NaCl. The cell-wall peptidoglycan contained ll-diaminopimelic acid, glutamic acid, alanine and glycine. The predominant menaquinones were MK-9(H6) and MK-9(H8). Whole-cell hydrolysates mainly contained glucose and ribose. Phosphatidylinositol and phosphatidylcholine were the diagnostic phospholipids. The G+C content of the genomic DNA was 72.8 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain JL-6T belonged to the genus Streptomyces with sequence similarities ranging from 97.3 % to 98.3 %. However, DNA–DNA hybridization between JL-6T and the closest related strain, Streptomyces turgidiscabies , ATCC 700248T and other closely related species in the genus Streptomyces showed <50 % relatedness. Based on these observations, strain JL-6T is proposed to represent a novel species of the genus Streptomyces , for which the name Streptomyces graminilatus sp. nov. is proposed. The type strain is JL-6T ( = KACC 16470T = NBRC 108882T).

A novel Gram-staining-positive actinobacterium, designated H85-3T, was isolated from a sea sediment sample and its taxonomic position was investigated by a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequence comparisons revealed that strain H85-3T was closely related to the members of the genus Zhihengliuella with pairwise sequence similarities of 97.4–98.6 %. The peptidoglycan of strain H85-3T was found to be of the A4α type with lysine as the diagnostic diamino acid. The menaquinones were MK-9, MK-10 and MK-8 (56 : 30 : 14) and the major cellular fatty acids were anteiso-C15 : 0, iso-C15 : 0 and anteiso-C17 : 0. These data supported the affiliation of strain H85-3T to the genus Zhihengliuella . Meanwhile, the results of DNA–DNA hybridization, along with the differences in some phenotypic characteristics, indicated that strain H85-3T should be distinguished from the recognized species of the genus Zhihengliuella . Therefore, strain H85-3T represents a novel species of the genus Zhihengliuella , for which the name Zhihengliuella flava sp. nov. is proposed; the type strain is H85-3T ( = NBRC 109021T = DSM 26152T). An emended description of the genus Zhihengliuella is also proposed.

A Gram-positive, aerobic, coccus-shaped, non-endospore-forming bacterium (Gsoil 633T) was isolated from soil from a ginseng field in Pocheon province in South Korea. The novel isolate was characterized in order to determine its taxonomic position. On the basis of 16S rRNA gene sequence similarities, strain Gsoil 633T was shown to belong to the family Propionibacteriaceae. The closest phylogenetic relative was Microlunatus phosphovorus DSM 19555T, with 96.1 % sequence similarity; the sequence similarity to other members of the family was less than 95.4 %. The isolate was characterized chemotaxonomically as having ll-2,6-diaminopimelic acid in the cell-wall peptidoglycan, MK-9(H4) as the predominant menaquinone and anteiso-C15 : 0, iso-C15 : 0 and iso-C16 : 0 as the major fatty acids. The G+C content of the genomic DNA was 69.8 mol%. The morphological and chemotaxonomic properties of the isolate were consistent with those of M. phosphovorus, but the results of physiological and biochemical tests allowed the phenotypic differentiation of strain Gsoil 633T from this species. Therefore, strain Gsoil 633T represents a novel species, for which the name Microlunatus ginsengisoli sp. nov. is proposed. The type strain is Gsoil 633T (=KCTC 13940T=DSM 17942T).

A rod-shaped, endospore-forming, Gram-reaction-positive bacterium, designated strain TC22-2bT, was isolated from compost in Tochigi, Japan. Phylogenetic analysis based on 16S rRNA gene sequences showed that the strain belonged to a cluster comprising species of the genus Paenibacillus and was most closely related to the type strain of Paenibacillus elgii (93.4 % similarity). The major cellular fatty acids were C16:0 (25.5 %), iso-C16:0 (23.6 %) and anteiso-C15:0 (21.5 %). The predominant menaquinone was MK-7. The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. The diamino acid found in the cell wall peptidoglycan was meso-diaminopimelic acid, and the DNA G+C content was 59.1 mol%. The results of physiological and biochemical tests enabled the phenotypic differentiation of strain TC22-2bT from the most closely related species with validly published names. Phylogenetic and phenotypic evidence reveals that strain TC22-2bT represents a novel species of the genus Paenibacillus , for which the name Paenibacillus thermoaerophilus sp. nov. is proposed. The type strain of the novel species is TC22-2bT ( = DSM 26310T  = JCM 18657T).

A novel actinobacterial strain, MBRL 353T, was isolated from a sample collected from a limestone quarry at Hundung, Manipur, India. Comparison of 16S rRNA gene sequences of strain MBRL 353T and other members of the genus Rhodococcus showed sequence similarities ranging from 95.5 to 98.2 %, with strain MBRL 353T showing closest sequence similarity to Rhodococcus triatomae IMMIB RIV-085T (98.2 %) and Rhodococcus equi DSM 20307T (97.2 %). DNA–DNA hybridization results, however, revealed that DNA–DNA relatedness values between strain MBRL 353T and R. triatomae DSM 44892T (43.4 %) and R. equi DSM 20307T (33.4 %) were well below the 70 % limit for species identification. Strain MBRL 353T contained meso-diaminopimelic acid as the diagnostic diamino acid and galactose and arabinose in the cell wall. Mycolic acids were present. The major fatty acids were C16 : 0 (45.7 %), C18 : 1ω9c (18.2 %) and 10-methyl C18 : 0 (11.3 %). The only menaquinone detected was MK-8(H2), while the major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositol mannoside and one unknown phospholipid. The G+C content of the genomic DNA was 69.2 mol%. The phenotypic and genotypic data showed that strain MBRL 353T merits recognition as a representative of a novel species of the genus Rhodococcus for which the name Rhodococcus canchipurensis sp. nov. is proposed; the type strain is MBRL 353T ( = KCTC 19851T = JCM 17578T).
