Mariniluteicoccus flavus gen. nov., sp. nov., a new member of the family Propionibacteriaceae, isolated from a deep-sea sediment

A Gram-staining-positive, aerobic, non-motile, irregular coccus, designated strain YIM M13146T, was isolated from a sediment sample collected from the South China Sea at a depth of 2439 m, and its taxonomic position was determined by a polyphasic approach. Optimal growth of the strain was observed at 30 °C (range 5–40 °C), pH 7.0 (pH 6.0–9.0) and 0–1 % NaCl (0–6 %, w/v) on/in tryptic soy agar/broth. Strain YIM M13146T had the major cellular fatty acid anteiso-C15 : 0, the predominant respiratory menaquinone MK-9(H4), peptidoglycan type A3γ (ll-DAP–Gly) containing alanine, glycine, glutamic acid and ll-diaminopimelic acid (ll-DAP) and the polar lipids phosphatidylcholine, diphosphatidylglycerol, one unknown phospholipid and several glycolipids. The G+C content of the DNA was 67.2 mol%. Phenotypic and chemotaxonomic characteristics together with 16S rRNA gene sequence analyses showed that strain YIM M13146T was distinct from its close phylogenetic relatives in the genera Propioniferax and Granulicoccus of the family Propionibacteriaceae . Hence, a new genus and species, Mariniluteicoccus flavus gen. nov., sp. nov., is proposed. The type strain of Mariniluteicoccus flavus is YIM M13146T ( = DSM 25892T = CCTCC AB 2012055T).

Two novel Gram-staining-positive, rod-shaped, endospore-forming and moderately thermophilic bacteria, designated strains DX-3T and GIESS002, were isolated from sludge composts from Guangdong Province, China. Analysis of 16S rRNA gene sequences revealed that the isolates were closely related to each other with extremely high similarity (99.6 %), and were members of the family Bacillaceae . However, these two isolates formed a novel phylogenetic branch within this family. Their closest relatives were the members of the genera Ornithinibacillus , Oceanobacillus and Virgibacillus . Cells of both strains were facultatively anaerobic and catalase- and oxidase-positive. The cell-wall peptidoglycan type was A1γ (meso-diaminopimelic acid direct). The predominant isoprenoid quinone was MK-7. The main polar lipids were diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. The major cellular fatty acid was iso-C15 : 0. The DNA G+C content was 43.2–43.7 mol%. The results of a polyphasic taxonomic study indicated that strains DX-3T and GIESS002 represent a novel species in a new genus in the family Bacillaceae , order Bacillales , for which the name Compostibacillus humi gen. nov., sp. nov. is proposed. The type strain is DX-3T ( = KCTC 33104T = CGMCC 1.12360T).

Eight facultatively anaerobic rod-shaped bacteria were isolated from raw milk and two other dairy products. Results of phylogenetic analyses based on 16S rRNA gene sequences showed that the isolates are placed in a distinct lineage within the family Propionibacteriaceae with Propioniciclava sinopodophylli and Propioniciclava tarda as the closest relatives (94.6 and 93.5 % similarity, respectively). The cell-wall peptidoglycan contained meso-diaminopimelic acid, alanine and glutamic acid and was of the A1γ type (meso-DAP-direct). The major cellular fatty acid was anteiso-C15 : 0 and the major polar lipids were diphosphatidylglycerol, phosphatidyglycerol and three unidentified glycolipids. The quinone system contained predominantly menaquinone MK-9(H4). The G+C content of the genomic DNA of strain VG341T was 67.7 mol%. The whole-cell sugar pattern contained ribose, rhamnose, arabinose and galactose. On the basis of phenotypic and genetic data, eight strains (VG341T, WS4684, WS4769, WS 4882, WS4883, WS4901, WS4902 and WS4904) are proposed to be classified as members of a novel species in a new genus of the family Propionibacteriaceae , for which the name Brevilactibacter flavus gen. nov., sp. nov. is proposed. The type strain is VG341T (=WS4900T=DSM 100885T=LMG 29089T) and seven additional strains are WS4684, WS4769, WS4882, WS4883, WS4901, WS4902 and WS4904. Furthermore, we propose the reclassification of P. sinopodophylli as Brevilactibacter sinopodophylli comb. nov.

A novel Gram-staining-positive, rod-shaped, motile, strictly aerobic, endospore-forming bacterium, designated WCC 4585T, was isolated from a pharmaceutical production line. The organism grew optimally at 30 °C, at pH 8 and in the presence of 0.5 % (w/v) NaCl. Oval endospores were formed subterminally and terminally in swollen sporangia. The cell-wall diamino acid was meso-diaminopimelic acid (type A1γ) and the genomic DNA G+C content was 38.7 mol%. The major menaquinone was MK-7. The cellular fatty acid profile contained major amounts of iso-C15 : 0, anteiso-C15 : 0 and anteiso-C17 : 0, and the cellular phospholipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and aminophospholipid. The isolate was most closely related to Bacillus oceanisediminis H2T, Bacillus infantis SMC 4352-1T, Bacillus firmus NCIMB 9366T, Bacillus circulans ATCC 4513T and Bacillus horneckiae DSM 23495T with which it shared less than 98.0 % 16S rRNA gene sequence similarity. DNA–DNA relatedness values between strain WCC 4585T and five type strains of related species were ≤27 % and sequence similarity values based on groEL sequences were ≤88.7 %. On the basis of the characteristics presented, strain WCC 4585T is proposed to represent a novel species, Bacillus gottheilii sp. nov. The type strain is WCC 4585T( = DSM 23668T = CCUG 59876T = LMG 25856T).

A novel halophilic, filamentous, actinomycete strain, designated CXB832T, was isolated from a salt pond in Qingdao, China. Optimal growth occurred at 37 °C, pH 7.0–8.0 and 9–12 % (w/v) NaCl. Strain CXB832T formed pale yellow to deep yellow branched substrate mycelium without fragmentation. Abundant white aerial mycelium differentiated into long chains of spores and the spores were rod-shaped with smooth surfaces. Strain CXB832T contained meso-diaminopimelic acid as the diagnostic diamino acid of the cell-wall peptidoglycan, and glucose and xylose as the major whole-cell sugars. The phospholipids were diphosphatidylglycerol, phosphatidylglycerol, phospholipids, glycolipid and unidentified lipids. MK-10(H8), MK-9(H8), MK-10(H2) and MK-10(H6) were the predominant menaquinones. The major fatty acids were i-C16 : 0 (30.71 %), ai-C17 : 0 (13.31 %) and C16 : 0 (11.28 %). The G+C content of the DNA was 60.1 mol%. Comparative analysis of 16S rRNA gene sequences showed that the novel strain was most closely related to genera within the family Nocardiopsaceae , but formed a separate lineage. The highest sequence similarities were to Nocardiopsis arabia DSM 45083T (95.4 %) and Haloactinospora alba DSM 45015T (94.9 %). On the basis of phenotypic, chemotaxonomic and phylogenetic distinctiveness, strain CXB832T represents a new genus and novel species in the family Nocardiopsaceae , for which the name Salinactinospora qingdaonensis gen. nov., sp. nov. is proposed. The type strain of the type species is CXB832T ( = DSM 45442T = LMG 25567T).

A novel actinobacterium, designated IPBSL-7T, was isolated from a drilling core 297 m deep obtained from the Iberian Pyrite Belt. The strain was isolated anaerobically using nitrate as the electron acceptor. 16S rRNA gene sequence analysis revealed that it was related to Tessaracoccus flavescens SST-39T (95.7 % similarity), Tessaracoccus bendigoensis Ben 106T (95.7 %), Tessaracoccus lubricantis KSS-17SeT (95.6 %) and Tessaracoccus oleiagri SL014B-20A1T (95.0 %), while its similarity to any other member of the family Propionibacteriaceae was less than 94 %. Cells were non-motile, non-spore-forming, Gram-positive, oval to rod-shaped, and often appeared in pairs or small groups. The strain was facultatively anaerobic, oxidase-negative, catalase-positive and capable of reducing nitrate. Colonies were circular, convex, smooth and colourless. The organism could grow at between 15 and 40 °C, with an optimal growth at 37 °C. The pH range for growth was from pH 6 to 9, with pH 8 being the optimal value. Strain IPBSL-7T had peptidoglycan type A3-γ′, with ll-diaminopimelic acid as the diagnostic diamino-acid and glycine at position 1 of the peptide subunit. The dominant menaquinone was MK-9(H4) (93.8 %). The major cellular fatty acid was anteiso-C15 : 0 (55.0 %). The DNA G+C content was 70.3 mol%. On the basis of phenotypic and phylogenetic results, strain IPBSL-7T can be differentiated from previously described species of the genus Tessaracoccus and, therefore, represents a novel species, for which the name Tessaracoccus lapidicaptus sp. nov. is proposed. The type strain is IPBSL-7T ( = CECT 8385T = DSM 27266T).

A Gram-stain-positive, facultatively anaerobic and rod-shaped bacterium, designated strain xj7T, was isolated from roots of Typha angustifolia L. growing in Beijing Cuihu Wetland, China. The isolate was identified as a member of the genus Paenibacillus based on phenotypic characteristics and phylogenetic inference. The novel strain was spore-forming, motile, catalase-positive and oxidase-negative. Optimal growth of strain xj7T occurred at 28–30 °C and pH 7.0–7.5. Diphosphatidylglycerol was the most abundant polar lipid and occurred along with phosphatidylglycerol, phosphatidylethanolamine, one unknown phospholipid and three unknown aminophospholipids. The diamino acid found in the cell-wall peptidoglycan was meso-diaminopimelic acid. The predominant isoprenoid quinone was MK-7. The major fatty acid components were anteiso-C15 : 0 (56.1 %), iso-C16 : 0 (9.1 %), C16 : 0 (8.0 %), iso-C14 : 0 (6.3 %) and iso-C15 : 0 (5.1 %). The G+C content of genomic DNA was 47.9 mol%. Phylogenetic analysis based on the 16S rRNA gene sequence showed that strain xj7T fell within the evolutionary radiation encompassed by the genus Paenibacillus , its closest neighbours were Paenibacillus borealis KK19T (97.5 %) and Paenibacillus durus DSM 1735T (97.1 %). However, the DNA–DNA relatedness values between strain xj7T and P. borealis KK19T and between strain xj7T and P. durus DSM 1735T, were both 35 %. Based on phenotypic, chemotaxonomic and phylogenetic properties, strain xj7T is considered to represent a novel species of the genus Paenibacillus , for which the name Paenibacillus typhae sp. nov. is proposed. The type strain is xj7T ( = CGMCC 1.11012T = DSM 25190T).

A novel marine actinomycete, designated strain KJ-029T, was isolated from a marine sediment sample (water depth of 226 m) in Kagoshima, Japan. 16S rRNA gene sequence analysis revealed that the new isolate was most closely related to Micromonospora craniellae LHW 63014T (99.3 % similarity). Phylogenetic analyses of the genus Micromonospora based on 16S rRNA gene sequences showed that strain KJ-029T was clustered with Micromonospora craniellae LHW 63014T and Micromonospora endophytica 202201T. However, digital DNA–DNA hybridization analyses presented low levels of relatedness in the range of 24.8–32.9 % between strain KJ-029T and the above closely related strains. The novel strain contained meso-diaminopimelic acid and 3-OH-diaminopimelic acid, d-glutamic acid, glycine and d-alanine in the cell-wall peptidoglycan. The acyl type of the peptidoglycan was glycolyl and mycolic acids were absent. The major menaquinone was MK-9(H4). The whole-cell sugars consisted of glucose, mannose, xylose and ribose. Phosphatidylethanolamine was detected as the major phospholipid and corresponded to phospholipid type II. The predominant cellular fatty acid was iso-C16 : 0. The DNA G+C content of the genomic DNA was 71.5 mol%. Based on the present polyphasic study, strain KJ-029T represents a novel species of the genus Micromonospora , for which the name Micromonospora pelagivivens sp. nov. is proposed. The type strain is KJ-029T (=NBRC 113519T=TBRC 9233T).

A novel actinobacterial strain was isolated from a seawater sample collected on Mara Island, Jeju, Republic of Korea. Cells of this organism were aerobic, Gram-positive, non-spore-forming, non-motile cocci that occurred singly or in pairs. Colonies were circular, smooth, convex and white–cream in colour. Phylogenetic analyses based on 16S rRNA gene sequences showed that the organism belonged to the family Dermacoccaceae and formed a monophyletic clade between the type strains of Demetria terragena (96.8 % similarity) and Branchiibius hedensis (95.2 % similarity). The cell-wall peptidoglycan contained l-lysine, alanine, aspartic acid, glutamic acid, glycine and serine, indicating that the isolate possessed peptidoglycan type A4α. The whole-cell sugars were galactose, glucose, mannose, xylose, arabinose, ribose and rhamnose. The major menaquinone was MK-8(H4). The polar lipids contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, an unknown phospholipid and five unknown lipids. The cellular fatty acid profile was represented by large amounts of iso-methyl branched and monounsaturated iso- and anteiso-methyl branched acids, along with the presence of a diagnostic 10-methyl acid. The G+C content of the DNA was 71 mol%. On the basis of data from polyphasic analyses presented here, strain MSW-24T is considered to represent a novel species of a new genus in the family Dermacoccaceae , for which the name Tamlicoccus marinus gen. nov., sp. nov. is proposed. The type strain of Tamlicoccus marinus is MSW-24T ( = KCTC 19485T = DSM 21415T).

Two Gram-staining-positive, rod-shaped and endospore-forming bacteria that represent a single species, designated strains GD05T and GD051, were isolated from a tropical forest soil and a hot spring sediment, respectively. Cells of both strains were facultatively anaerobic, catalase- and oxidase-positive, and could grow optimally at 50 °C, pH 8.0 and with 1 % (w/v) NaCl. Analysis of the 16S rRNA gene sequence revealed that these two isolates belonged to the family Bacillaceae , but did not show sequence similarities of more than 95 % to members of other related genera. The G+C content of the genomic DNA was 43.7–44.1 mol%. The major cellular fatty acids were anteiso-C15 : 0, iso-C15 : 0, iso-C16 : 0 and anteiso-C17 : 0. The main polar lipids were diphosphatidylglycerol and phosphatidylglycerol, and the major menaquinone was MK-7. The peptidoglycan type was A1γ (meso-diaminopimelic acid direct). On the basis of this polyphasic taxonomic analysis, the novel strains represent a novel species of a new genus in the family Bacillaceae , order Bacillales , for which the name Sinibacillus soli gen. nov., sp. nov. is proposed. The type strain is GD05T ( = CCTCC AB 2013105T = KCTC 33117T).

A novel thermotolerant bacterium, designated SgZ-8T, was isolated from a compost sample. Cells were non-motile, endospore-forming, Gram-staining positive, oxidase-negative and catalase-positive. The isolate was able to grow at 20–65 °C (optimum 50 °C) and pH 6.0–9.0 (optimum 6.5–7.0), and tolerate up to 9.0 % NaCl (w/v) under aerobic conditions. Anaerobic growth occurred with anthraquinone-2,6-disulphonate (AQDS), fumarate and NO3 - as electron acceptors. Phylogenetic analysis based on the16S rRNA and gyrB genes grouped strain SgZ-8T into the genus Bacillus , with the highest similarity to Bacillus badius JCM 12228T (96.2 % for 16S rRNA gene sequence and 83.5 % for gyrB gene sequence) among all recognized species in the genus Bacillus . The G+C content of the genomic DNA was 49.3 mol%. The major isoprenoid quinone was menaquinone 7 (MK-7) and the polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and an unidentified phospholipid. The major cellular fatty acid was iso-C16 : 0. On the basis of its phenotypic and phylogenetic properties, chemotaxonomic analysis and the results of physiological and biochemical tests, strain SgZ-8T ( = CCTCC AB 2012108T = KACC 16706T) was designated the type strain of a novel species of the genus Bacillus , for which the name Bacillus thermotolerans sp. nov. is proposed.
