Phylogeography and population genetics of the maize stalk borer Busseola fusca (Lepidoptera, Noctuidae) in sub-Saharan Africa

The stem borer Busseola fusca (Fuller) (Lepidoptera: Noctuidae) is an important pest of maize and sorghum in sub-Saharan Africa. This insect has oligophagous feeding habits, feeding mostly on maize and sorghum with a narrow range of wild Poaceous plant species. We hypothesised that first instar B. fusca larvae, the critical stage for successful establishment on a host plant, can establish and then grow on a particular plant as a result of induction of a complement of digestive enzymes that mediates host acceptance at first instars. A fast semi-quantitative analysis of potentially digestive enzymatic activities present in the first larvae previously fed for 4 days on leaves of host and non-host plants was performed using the API-ZYM kit system able to detect a multiplex of enzyme activities. Regardless of the plant species, the larvae exhibited higher activities of the carbohydrate metabolising enzymes than of aminopeptidases and proteases. In addition, highest activities of carbohydrates degrading enzymes were exhibited by larvae that consumed leaves of the most preferred plant species of B. fusca. Conversely, esterases were only detected in neonate larvae that consumed leaves of the less preferred and non-host plants. No alkaline phosphatase and lipase activities were detected. The significance of these results was discussed in terms of food requirements of first instar larvae when settling on a plant.

Abstract Busseola fusca (Fuller) (Lepidoptera: Noctuidae), the maize stalk borer, is a widespread crop pest in sub-Saharan Africa that has been the focus of biological research and intensive management strategies. Here, we present a comprehensive annotated transcriptome of B. fusca (originally collected in the Western Province of Kenya) based on ten pooled libraries including a wide array of developmental stages, tissue types, and exposures to parasitoid wasps. Parasitoid wasps have been used as a form of biocontrol to try and reduce crop losses with variable success, in part due to differential infectivities and immune responses among wasps and hosts. We identified a number of loci of interest for pest management, including genes potentially involved in chemoreception, immunity, and response to insecticides. The comprehensive sampling design used expands our current understanding of the transcriptome of this species and deepens the list of potential target genes for future crop loss mitigation, in addition to highlighting candidate loci for differential expression and functional genetic analyses in this important pest species.

SUMMARYIntestinal schistosomiasis continues to be a major public health problem in sub-Saharan Africa, and is endemic in communities around Lake Victoria. Interest is growing in the molecular evolution and population genetic structure of Schistosoma mansoni and we describe a detailed analysis of the molecular epidemiology and phylogeography of S. mansoni from Lake Victoria. In total, 388 cytochrome oxidase 1 (COI) sequences were obtained from 25 sites along the Ugandan, Tanzanian and Kenyan shorelines of Lake Victoria, and 122 unique barcodes were identified; 9 corresponded to previously discovered barcodes from Lakes Victoria and Albert. A subset of the data, composed of COI sequences from miracidia from 10 individual children, was used for population genetics analyses; these results were corroborated by microsatellite analysis of 4 isolates of lab-passaged adult worms. Overall, 12 barcodes were found to be shared across all 3 countries, whereas the majority occurred singly and were locally restricted. The population genetics analyses were in agreement in revealing high diversity at the level of the human host and negligible population structuring by location. The lack of correlation between genetic distance and geographical distance in these data may be attributed to the confounding influence of high intra-individual diversity as well as human migration between communities.

The fall armyworm (Spodoptera frugiperda J.E Smith) (Lepidoptera: Noctuidae) invaded Africa in 2016, and has since spread to all countries in sub-Saharan Africa, causing devastating effects on mainly maize and sorghum. The rapid spread of this pest is aided by its high reproductive rate, high migration ability, wide host range and adaptability to different environments, among others. Since its introduction, many governments purchased and distributed pesticides for emergency control, with minimal regard to their efficacy. In this chapter, we review efforts towards managing this pest, highlight key challenges, and provide our thoughts on considerations for sustainable management of the pest.

Abstract It has been over five years since the first report of an outbreak of the fall armyworm, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) in Africa. The highly invasive pest, native to the Americas, has since spread across the African continent attacking many crops and causing significant yield loss to Africa’s staple crop, maize. From the onset of the outbreak, there have been massive and varied responses from farmers, governments and nongovernmental organizations. This mini-review provides various perspectives on S. frugiperda control in sub-Saharan Africa, building on previously published evidence, and experiences of the authors. It also highlights new technologies and lessons learned so far from the S. frugiperda outbreaks in sub-Saharan Africa, based on which suggestions on possible integrated management approaches are proffered.
