To elucidate the possible role of Rho A/Rho-kinase on lysophosphatidic acid (LPA)-induced contraction in intact guinea-pig ileal smooth muscle, we examined effects of pretreatment with a specific inhibitor of Rho-kinase (Y-27632) on the LPA-induced contraction and MLC20 phosphorylation. In addition, we investigated whether LPA actually elicits an activation of Rho A by studying subcellular distribution of Rho A in unstimulated and stimulated smooth muscles by LPA. LPA induced a less intense, but sustained, contraction compared with ACh, and was accompanied by significant increases in MLC20 phosphorylation. The effects of LPA on tension and MLC20 phosphorylation were inhibited by Y-27632. The ACh-induced contraction, but not increases in MLC20 phosphorylation, was partially inhibited by Y-27632. High K+-induced contraction was unaffected by the inhibitor. LPA stimulated translocation of Rho A from the cytosol to the membrane fraction of the muscle. Translocation of Rho A was also induced by ACh and high K+. These results suggest that LPA-induced contraction of intact ileal smooth muscle is dominated through activation of Rho A and Rho-kinase and subsequent increases in MLC20 phosphorylation.Key words: lysophosphatidic acid, Rho, Rho-kinase, ileal smooth muscle.

The influence of modulation of cytoskeleton by colchicine, vinblastine, cytochalasine B and docetaxel on contractile reactions of smooth muscle cells caused by electric stimulus, depolarization, phenylephrine has been investigated by the mechanographical method, by the methods of the double sucrose gup junction. It is established, that induced by a isoosmotic hyperpotassium solution of reduction of smooth muscle of the rat’s aorta, and also caused depolarization stimulus potentials of action and reductions smooth muscle cells from guinea pig urethra, depend more on the condition of microfilaments cytoskeleton than on microtubules. The reduction of smooth muscles cells of an aorta of the rat, caused by isoosmotic striction, is suppressed under the destruction microfilaments whereas the reduction in a hyperosmotic solution depends on a condition of both microfilaments, and microtubules. Cytoskeleton’s microfilaments of aorta’s smooth muscles and microtubules of smooth muscles of cells ureter are involved in mechanisms of action phenylephrine’s action on contractile activity of smooth muscle cells of an aorta and ureter.

The role of β1- and β2-adrenergic receptor stimulation in modulating adenosine 3′,5′-cyclic monophosphate (cAMP)-regulated Cl− and Ca2+ currents was investigated with use of guinea pig ventricular myocytes. Activation of the Cl− current by the nonselective β-receptor agonist isoproterenol (Iso) was not affected by the β2-receptor antagonist ICI-118,551 (ICI), but it was blocked by the β1-receptor antagonist atenolol. The inability of β2-receptor stimulation to activate the Cl− current was confirmed by the lack of response to the selective β2-receptor agonists salbutamol and zinterol. Responses to β2-adrenergic receptor stimulation were also looked for in pertussis toxin (PTX)-treated myocytes because PTX increases the sensitivity of responses to Iso, and PTX has been reported to increase the responsiveness to β2- but not β1-receptor stimulation. PTX treatment increased the sensitivity of the Cl− current to activation by Iso in the presence of ICI, indicating that PTX increases β1-receptor responsiveness. PTX treatment also resulted in the ability of salbutamol to activate the Cl− current. However, the response to salbutamol was blocked by atenolol but not by appropriate concentrations of ICI, suggesting that salbutamol was activating β1-receptors. These results indicate that PTX treatment increases the sensitivity to β1-receptor stimulation, without affecting β2-responsiveness. To verify that the lack of response to β2-receptor stimulation was not unique to the Cl− current, the effects of β2-receptor agonists on the L-type Ca2+current were also examined. The Ca2+ current was only affected by high concentrations of zinterol or salbutamol, and such responses were blocked by atenolol, but not by ICI, suggesting that activation of β1-receptors was involved. These results indicate that β1- but not β2-adrenergic receptor stimulation plays an important role in modulating the cAMP-regulated Cl− and Ca2+ currents in guinea pig ventricular myocytes.

Objectives: This study focuses on the role of the mediator of smooth muscles i.e. nitric oxide in the effect of rosiglitazone on the relaxation of contractions in guinea pig trachea induced by histamine. Study Design: Experimental Study. Setting: Department of Pharmacology, Army Medical College, Rawalpindi. Period: 1st December 2018 till 14th December 2018. Material & Methods: The activity of the trachealis mucle of the guinea pigs was noted by the use of oxygenated solution of Krebs-Henseleit. The contractions were noted with rosiglitazone alone as well as in presence of an ester of L-nitro-arginine-methyl, which can antagonize the activity of NO. The activity was measured by the use of an isometric force displacement transducer and was then recorded on an oscillograph with four channels. Results: The ester L-nitro-arginine-methyl was able to reduce the relaxant effect of rosiglitazone on the contractions induced by histamine. Conclusion: It can be suggested that the relaxation induced by rosiglitazone is mediated in part through NO.

The aim of this study was to determine whether polydatin, a glucoside of resveratrol isolated from the root of Polygonum cuspidatum, warranted development as a potential therapeutic for ameliorating the pain originating from gallbladder spasm disorders and the underlying mechanisms. Guinea pig gallbladder smooth muscles were treated with polydatin and specific inhibitors to explore the mechanisms underpinning polydatin-induced relaxation of carbachol-precontracted guinea pig gallbladder. Our results shown that polydatin relaxed carbachol-induced contraction in a dose-dependent manner through the nitric oxide/cyclic guanosine monophosphate/protein kinase G and the cyclic adenosine monophosphate/protein kinase A signaling pathways as well as the myosin light chain kinase and potassium channels. Our findings suggested that there was value in further exploring the potential therapeutic use of polydatin in gallbladder spasm disorders.
