Endophytically colonized Citrus limon seedlings by Beauveria bassiana hampered development, reproduction and progeny fitness of Diaphorina citri

Entomopathogenic fungi are commonly applied as inundative sprays to protect plants against insect pests. Their artificial establishment as fungal endophytes to provide other benefits to the host plants aside mere protection against the primary pests has also been widely demonstrated. In the present study, two fungal strains of Beauveria bassiana and one strain of Isaria fumosorosea were assessed in a pathogenicity test against adults of Asian citrus psyllid (Diaphorina citri) and found to induce 50% reduction in the survival rate of D. citri adults within 5 days of exposure. The ability of the three fungal strains to endophytically colonize Citrus limon, the impact on plant growth and the effects of systemic colonization on 3 successive generations of D. citri feeding on colonized plants was evaluated. Citrus seedlings at 4 months post-planting were inoculated with each of the fungal strains via foliar spraying. Both strains of B. bassiana successfully colonized the seedlings. One of the B. bassiana strains (BB Fafu-13) was sustained up to 12 weeks in the colonized seedlings, whereas the other B. bassiana strain (BB Fafu-16) was only recovered up to 8 weeks post-inoculation. Isaria fumosorosea (IF Fafu-1) failed to colonize the plant. Both strains of B. bassiana induced significant improvement in plant height and flush production in endophytically colonized seedlings. In addition, endophytic B. bassiana caused 10–15% D. citri adult mortality within 7 days of exposure. Female D. citri feeding on B. bassiana challenged plants laid fewer eggs as compared to those feeding on endophyte-free seedlings, while reduction in adult emergence was recorded on B. bassiana treated plants. With this study, we present the first evidence of B. bassiana artificial establishment as fungal endophyte in citrus plants and its negative effects on D. citri.

RESUMO Avaliou-se a patogenicidade de diversas espécies de fungos entomopatogênicos a ninfas de 2o a 4o ínstares do psilídeo Diaphorina citri. Foram feitos bioensaios com Beauveria bassiana, Metarhizium anisopliae, Lecanicillium muscarum, L. longisporum, Paecilomyces fumosoroseus, P. farinosus, Syngliocladium sp. na concentração de 5 x 107 conídios/mL para cada patógeno, com exceção de Hirsutella thompsonii que foi aplicado na concentração de 2,8 x 107 conídios/mL. Utilizaram-se mudas de murta, Murraya paniculata, infestadas com as ninfas do inseto que foram pulverizadas com as suspensões conidiais dos patógenos. Os fungos B. bassiana, M. anisopliae, H. thompsonii, L. muscarum e P. fumosoroseus foram patogênicos para as ninfas do psilídeo. O isolado Esalq-PL63, de B. bassiana, causou mortalidade de aproximadamente 72% das ninfas, sete dias após a inoculação. Constatou-se que B. bassiananão conseguiu completar o desenvolvimento no corpo do hospedeiro, uma vez que a fase de conidiogênese não ocorreu. Mesmo assim, o isolado Esalq-PL63 pode ser considerado um promissor agente de controle microbiano de ninfas de D. citri por causar altos índices de mortalidade.

The inoculation methods, the fungal strains, and several other factors are known to influent the success of fungal entomopathogens colonization in plants. The physiological status of the plant could also be another determinant. In the present study, the ability of three strains of Beauveria bassiana and one strain of Metarhizium anisopliae to successfully colonize Citrus limon plants and the influence of seedling age on endophytic colonization success was examined. Three, 4, and 6 months old seedlings were inoculated with 10 mL of 1 × 108 conidial·mL−1 suspensions of each of the four fungal strains via foliar spraying. All fungal strains successfully colonized citrus seedlings and were sustained up to 2 months in colonized plants irrespective of the seedling age, with differences in the mean percentage colonization recorded at various post-inoculation periods among the fungal strains. The highest percent endophytic fungi recovery rate was recorded in the 3 months old seedlings, where fungal mycelia of inoculated fungi were successfully re-isolated from 65.6% of the untreated newly developed leaf and stem tissues. One strain of B. bassiana, BB Fafu-12, significantly improved seedling height and leaf number. The study demonstrates the influence of seedling age on B. bassiana and M. anisopliae successful colonization in the citrus plant.

O objetivo deste trabalho foi avaliar a patogenicidade de Beauveria bassiana a ninfas de Diaphorina citri (Hemiptera: Psyllidae) e verificar a compatibilidade do fungo com produtos fitossanitários e sua persistência em plantas de citros. Ninfas de D. citri foram pulverizadas com B. bassiana, nas concentrações 5x10(6), 1x10(7), 5x10(7), 1x10(8), 5x10(8) e 1x10(9) conídios mL-1, para determinação da concentração letal. Para avaliação da compatibilidade do fungo com produtos fitossanitários, extrato de nim e cinco inseticidas de quatro grupos químicos diferentes foram incorporados individualmente ao meio de cultura BDA em que o fungo foi cultivado. Avaliaram-se o crescimento vegetativo, a esporulação e a viabilidade do entomopatógeno. Plantas de citros, mantidas em casa de vegetação, foram tratadas primeiramente com os produtos fitossanitários e depois com o entomopatógeno. Avaliaram-se os tempos de exposição de 24 horas e de 7 e 14 dias. O fungo foi patogênico às ninfas de D. citri; a CL50 foi de 0,4x10(7) e a CL90 de 6,7x10(7) conidios mL-1, no décimo dia de avaliação. Em laboratório, os produtos fitosssanitários reduzem o crescimento do fungo. Em casa de vegetação, os produtos não afetam a sobrevivência do fungo nas plantas de citros.

Natural enemies associated to Diaphorina citri in the state of Nayarit were identified. The study was carried out in three orchards of Persian lime from May to October 2011. The identified depredator species were Olla v-nigrum, Chilocorus cacti, Cycloneda sanguinea, and Ceraeochrysa sp.; as well as the parasitoid Tamarixia radiata and the entomopathogenic Beauveria bassiana. Nephus sp. Mulsant, 1850 and Pentilia sp. Mulsant, 1850 are new distribution records to Mexico.

Este é o primeiro registro da espécie Diaphorina citri Kuwayama (Hemiptera: Liviidae) para o estado de Roraima, Brasil. Exemplares deste inseto foram coletados no município de Boa Vista sobre plantas de Citrus limon (L.) e Murraya paniculata (L.) Jack. (Rutaceae). Observou-se a predação de ninfas de D. citri por larvas de Ceraeochrysa fairchildi Banks (Neuroptera: Chrysopidae).
