A possibilidade de proibição do uso de antibióticos como promotores de crescimento para animais de produção tem feito com que consumidores e produtores procurem por alternativas. Os probióticos têm se mostrado promissores em cumprir este papel e por esta razão, setenta e nove bezerros da raça Holandesa foram utilizados para avaliar a adição de probiótico constituído por Lactobacillus acidophilus, Enterococcus faecium e Saccharomyces cerevisiae ao aleitamento. Os animais foram distribuídos em um delineamento inteiramente casualizado, com arranjo fatorial de tratamentos 3x2, correspondendo ao tipo de dieta líquida (leite integral, sucedâneo ao 3° dia ou sucedâneo ao 15° dia de idade, com adição ou não de probiótico). O período experimental foi do nascimento até 15 dias após a desmama. O fornecimento de probiótico a bezerros(as) aleitados(as) com sucedâneo a partir dos 3 dias de idade melhorou o ganho de peso (com probiótico=0,22 vs. sem probiótico=0,16 kg dia-1) e conversão alimentar (2,62 vs. 3,85) até a desmama, e conversão alimentar (1,66 vs. 2,03) pós-desmama. Os bezerros aleitados com sucedâneo consumiram mais concentrado em relação aos que receberam leite integral (sucedâneo=0,22 vs. leite=0,19 kg dia-1), mas consumiram menos matéria seca total (0,61 vs. 0,67 kg dia-1) e apresentaram menor peso à desmama (49 vs. 59 kg). Quando o uso de sucedâneo foi iniciado aos 3 dias, os bezerros apresentaram maior consumo de concentrado (sucedâneo aos 3 dias=0,25 vs. sucedâneo aos 15 dias=0,20 kg dia-1) e menor peso à desmama (47,3 vs. 51 kg) em relação aos bezerros aleitados com sucedâneo aos 15 dias.

O uso de probióticos a base de leveduras melhora o ganho de peso e o sistema imunológico de bovinos frente a grandes desafios imunológicos, no entanto permanece a dúvida se seu uso também apresentaria efeito em desafios mais tênues. Desta maneira, o presente trabalho verificou se o uso de probiótico a base de Enterococcus faecium e Saccaromyces cerevisiae via oral interfere na imunidade inata e no peso de bezerros sadios, em fase de crescimento, alimentados principalmente com silagem de milho. Para tanto, 12 bezerros da raça Jersey (120 kg ±50 kg e 4-7 meses de idade), foram alocados nos grupos controle (n=6) ou tratamento (n=6), de acordo com a oferta de probiótico (2g/dia, via oral) durante 45 dias. Nos dias 0, 15, 30 e 45, os animais foram submetidos a pesagens, exames clínicos, e colheita de sangue para realização de hemograma e mensuração do metabolismo oxidativo neutrofílico. Observou-se que, o probiótico promoveu pequeno e pontual incremento na função neutrofilica sanguínea (P=0,10, D15), incremento nos valores de hematócrito e hemoglobina (P=0,03 e 0,05, D45), além de maior ganho de peso nas fases iniciais da administração do suplemento (P=0,05), permitindo concluir que o suplemento é benéfico para animais em fase de adaptação de trocas de dieta e de manejo, mesmo quando estes desafios são tênues.

The aim of the study was to examine the possibility of replacing antibiotic growth promoters with a new probiotic preparation. One gram of the preparation contains: 4.7 × 107 of LAB (<i>Lactobacillus casei/paracasei</i> LOCK 0920, <i>L. brevis</i> LOCK 0944, <i>L. plantarum</i> LOCK 0945), 2.0 × 103 of yeast (<i>Saccharomyces cerevisiae</i> LOCK 0140) and a 50 mg of extract from Yucca Schidigera. The experiment was conducted on 99 female broilers divided into three groups consisting of 33 chickens, kept separately. A wheat- and soybean meal-based diet was divided into three parts: A (containing 1 g/kg of probiotic for starter and grower diets and 0.5g/kg for finisher diet), B (containing 14 mg/kg of flavomycin), and C (unsupplemented). The diets were fed from Day 1 to Day 41 of life. Final BW was on average 2.4 kg in all groups, FCR was 1.63 kg feed/kg BWG, neither BWG nor FCR nor mass of the liver, pancreas and gastrointestinal tract were significantly influenced by supplementing the diet with either additive. Special attention should be drawn to the fact that supplementing feeds with a probiotic preparation already after one week of breeding considerably decreased the number of <i>Clostridium</i> bacteria in broilers’ faeces. Nevertheless, it should be emphasized that in this study the excreta of chickens fed with the feed mixed with a probiotic contained the lowest changeability of the number bacteria of the <i>Enterobacteriaceae</i> family and bacteria belonging to coli groups in individual weeks of breeding. It may be concluded that the studied probiotic can be considered as a substitute for antibiotic growth promoters in broiler diets.

RESUMO: Objetivou-se avaliar a capacidade de adsorção in vitro de aflatoxina B1 (AFB1) por produtos comerciais utilizados na alimentação animal. Muitas pesquisas estão sendo realizadas para a descontaminação de AFB1 em alimentos. Os produtos comerciais utilizados frequentemente na alimentação de peixes, disponíveis na forma de probióticos, são formados por cepas de bactérias e leveduras utilizadas na maioria dos ensaios de adsorção de micotoxinas. Foram utilizados três produtos comerciais: A, composto por Bacillus subtilis, Bifidobacterium bifidum, Enterococcus faecium e Lactobacillus acidophilus; B, por leveduras secas de Saccharomyces cerevisiae provenientes de cervejaria; e C, por Bacillus subtilis, Bacillus licheniformis e Bacillus pumilus. Cinco suspensões da dose máxima recomendada pelo fabricante de cada produto (0; 25; 50; 75 e 100%) foram testadas contra AFB1 (1000 ng.mL-1) em microtubos para determinação da capacidade de adsorção. Para simular o pH do estômago e do intestino de tilápias do Nilo (Oreochromis niloticus) foram formuladas soluções tampão fosfato salino (PBS), com pH 1,5 e 7,5; respectivamente. Os microtubos foram introduzidos em uma centrífuga com agitação mecânica, a 37ºC por 1 h e depois centrifugados por 10 min a 14.000 rpm; os sobrenadantes foram quantificados por cromatografia líquida de alta eficiência. Os produtos comerciais, nas concentrações máximas, foram capazes de adsorver AFB1 em quantidades de 45,01 a 129,59; 123,90 a 215,59 e 209,98 a 370,73 ng.mL-1, respectivamente. Concluiu-se que todos os produtos comerciais analisados adsorvem AFB1 em condições simuladas de pH gastrointestinal e são candidatos potenciais para adsorção de AFB1 para futuros ensaios in vivo.

ABSTRACT A segregationally stable expression and secretion vector for Saccharomyces cerevisiae, named pYABD01, was constructed by cloning the yeast gene region encoding the mating pheromone α-factor 1 secretion signal (MFα1 s ) into the S. cerevisiae high-copy-number expression vector pYES2. The structural genes of the two leaderless peptides of enterocin L50 (EntL50A and EntL50B) from Enterococcus faecium L50 were cloned, separately (entL50A or entL50B) and together (entL50AB), into pYABD01 under the control of the galactose-inducible promoter PGAL1. The generation of recombinant S. cerevisiae strains heterologously expressing and secreting biologically active EntL50A and EntL50B demonstrates the suitability of the MFα1 s -containing vector pYABD01 to direct processing and secretion of these antimicrobial peptides through the S. cerevisiae Sec system.

ABSTRACTThis study was conducted to validate a simulated commercial baking process for hamburger buns to destroy Salmonella serovars and to determine the appropriateness of using nonpathogenic surrogates (Enterococcus faecium ATCC 8459 or Saccharomyces cerevisiae) for in-plant process validation studies. Wheat flour was inoculated (~6 log CFU/g) with three Salmonella serovars (Typhimurium, Newport, or Senftenberg 775W) or with E. faecium. Dough was formed, proofed, and baked to mimic commercial manufacturing conditions. Buns were baked for up to 13 min in a conventional oven (218.3°C), with internal crumb temperature increasing to ~100°C during the first 8 min of baking and remaining at this temperature until removal from the oven. Salmonella and E. faecium populations were undetectable by enrichment (&gt;6-log CFU/g reductions) after 9.0 and 11.5 min of baking, respectively, and ≥5-log-cycle reductions were achieved by 6.0 and 7.75 min, respectively. D-values of Salmonella (three-serovar cocktail) and E. faecium 8459 in dough were 28.64 and 133.33, 7.61 and 55.67, and 3.14 and 14.72 min at 55, 58, and 61°C, respectively, whereas D-values of S. cerevisiae were 18.73, 5.67, and 1.03 min at 52, 55, and 58°C, respectivly. The z-values of Salmonella, E. faecium, and S. cerevisiae were 6.58, 6.25, and 4.74°C, respectively. A high level of thermal lethality was observed for baking of typical hamburger bun dough, resulting in rapid elimination of high levels of the three-strain Salmonella cocktail; however, the lethality and microbial destruction kinetics should not be extrapolated to other bakery products without further research. E. faecium demonstrated greater thermal resistance compared with Salmonella during bun baking and could serve as a conservative surrogate to validate thermal process lethality in commercial bun baking operations. Low thermal tolerance of S. cerevisiae relative to Salmonella serovars limits its usefulness as a surrogate for process validations.
