The effects of a simple saline solution prepared using two different pH (4.4 and 8.5) on sperm motility in burbot were investigated. Results were recorded during a 96-hour chilled storage (4°C) in 24-hour intervals. Measurements were focused on the detailed characteristics of motility using 12 parameters obtained from the Computer-assisted Sperm Analysis (CASA). Significantly higher progressive motility (pMOT), distance average path (DAP), distance curved line, distance straight line (DSL), average path velocity (VAP), curvilinear velocity, straight line velocity, and beat cross frequency (BCF) were observed with the activating solution buffered at pH 8.5 in comparison with pH 4.4. Already after 24 h a significant reduction was measured in pMOT (0 h: 49 ± 24%, 24 h: 12 ± 7%). Similar decreasing tendency was recorded only after 72 h in DAP (0 h: 26 ± 4 µm/s, 72 h: 19 ± 9 µm/s), DSL (0 h: 21 ± 5 µm/s, 72 h: 17 ± 8 µm/s), VAP (0 h: 59 ± 9 µm/s, 72 h: 43 ± 21 µm/s), and BCF (0 h: 28 ± 2 Hz, 72 h: 18 ± 10 Hz). The response of different investigated CASA parameters to different treatments varied in our experiments. According to our studies, numerous burbot sperm motility parameters are sensitive to chilled storage and to low pH of the activating solution. Our results could support the effective sperm quality assessment and successful artificial propagation process in burbot.

A histological analysis was conducted on the gills of 15 Piaractus mesopotamicus and 19 Prochilodus lineatus specimens collected between April and November 2004 from the Aquidauana River, State of Mato Grosso do Sul, Central-West Brazil, to describe the anatomopathological characteristics of the gills of these freshwater fish. Gill samples were fixated in 10% buffered formalin and processed following histological routine procedures. The histological examination of the gills of P. mesopotamicus revealed intralamellar monogenean and mixosporean cysts of Henneguya piaractus at several developmental stages over the entire (basal, median and distal) lamella. Intraepithelial cysts caused lamella dilation and deformity of adjacent lamellae. In P. lineatus gills, monogenean cysts were detected. In both host species, hyperplasia of the gill epithelium and structural disorganization of secondary lamellae was seen diffusely in the gills, leading to fused lamellae in the gills. In few cases, there was found mononuclear inflammatory cells and hemorrhagic focal points distally in the lamellae.

Sperm of the following Neotropical Characiformes fish species were tested for cold storage: Brycon lundii, Piaractus mesopotamicus, Leporinus elongatus, Leporinus friderici, Prochilodus lineatus and Prochilodus marggravii. Each sperm sample was split into two aliquots. The first was placed into a plastic bag with air or oxygen and the second, in a plastic tube with air. The samples were maintained at temperatures between 1.7-4.9 ºC. The rate of sperm motility was estimated using a 50 mM NaCl solution as the activating solution. The shortest sperm storage duration (7 h) was recorded for L. friderici, when the sperm motility rate reached ~ 30%, whereas the longest duration (20 h) was obtained with the sperm of P. lineatus. A fertilisation test using Prochilodus marggravii sperm refrigerated for 8 h yielded 88-90% of viable embryos. The refrigerated storage method could be of practical applications, especially in fish reproductive management at hatchery stations.

Este estudo descreveu a histopatologia de rim, baço e fígado de Piaractus mesopotamicus, Prochilodus lineatus e Pseudoplatystoma fasciatum, parasitados por mixosporídios, capturados no Rio Aquidauana, MS. Após necropsia, amostras do fígado, rim cefálico e baço foram colhidas, fixadas em formalina a 10 % tamponada e processadas de acordo com a rotina histológica. Os cortes foram feitos à espessura de 5 μm e corados com hematoxilina-eosina. Foram encontrados Myxobolus porofilus em P. lineatus, M. colossomatis em P. mesopotamicus e Myxobolus spp. nas três espécies de hospedeiros. Cistos de mixosporídios no exame histopatológico foram vistos no fígado e baço de P. mesopotamicus. Mais de 50% das amostras de fígado de P mesopotamicus e P lineatus apresentou hepatodistrofia difusa. Mais de 80 % das amostras de fígado de P. fasciatum apresentou formações hialinas concêntricas e esteatose em 50% das amostras. Em 95,23 % das amostras de rins de P. mesopotamicus, foram observadas alterações teciduais, e em mais de 60 % dos casos nefrodistrofia difusa moderada e congestão de sinusóides glomerulares. Alterações teciduais nas amostras de rins de P. lineatus foram observadas em menos de 20 % da amostra. No baço dos peixes ora examinados não foram encontradas lesões dignas de relato.

This paper presents results of a fishway evaluation performed at the Engenheiro Sérgio Motta Hydroelectric Power Plant (known as Porto Primavera) - CESP, Paraná River, Brazil. The evaluation was designed to quantify entry and passage proportions of 4 long-distance migratory fish species: Brycon orbignyanus (piracanjuba), Piaractus mesopotamicus (pacu), Prochilodus lineatus (curimba), and Rhinelepis aspera (cascudo-preto). Proportions finding and entering the fishway differed between species, ranged from 7.4 % (Prochilodus lineatus) to 55.4% (Piaractus mesopotamicus). Also, proportion passing was different between species, ranged from 31% (R. aspera) to 100% (Prochilodus lineatus). Fish that were marked and released within the fishway had greater failure rates than those that entered volitionally. Total time to pass ranged from 1.48 hours (Prochilodus lineatus) to 178.9 hours (R. aspera). Failure rates were greatest in the lower end of the fishway. Although some individuals of all species passed successfully, significant challenges remain to restoring connectivity of the upper Paraná River.

Se analizó el comportamiento productivo de dos especies de peces autóctonos bajo un sistema intensivo de producción acuapónica. El sistema contó con cuatro módulos, dos para Pacú: M2 y M3 y dos para Sábalo: M1 y M4. Cada módulo estaba constituido por un tanque de 1000 l para los peces, un tanque sobre elevado de 200 l, que recibía el agua del tanque de peces y un biofiltro de 15 l que drenaba en los sistemas hidropónicos, los cuales tenían una superficie de 4 m2 por módulo y un flujo de agua en el sistema de 2 l/minuto. Los parámetros de calidad de agua (oxígeno, pH, temperatura y conductividad eléctrica) se mantuvieron dentro de valores aceptables para la cría de los peces y para el crecimiento de la lechuga. En la serie nitrogenada (nitrito, nitrato, amonio, amoníaco) se observaron variaciones en los diferentes módulos, sin llegar a valores críticos. Respecto a los índices productivos de los peces, se obtuvo un mejor desempeño en Pacú, obteniéndose después de 37 días de cultivo una biomasa de 6.900 y 6.930 g para los módulos 2 y 3 de Pacú y 4.780 y 5.090g para los módulos 2 y 4 de Sábalo, respectivamente. Para la lechuga, la biomasa total fue de 7.482, 4.872, 8.584 y 10.672 g para los módulos 1, 2, 3 y 4, respectivamente. Los resultados indican que el Pacú es una especie de pez alternativa para la producción de acuaponía.

ABSTRACT: Brycon gouldingi is a species of neotropical fish of socioeconomic and environmental importance in the Tocantins-Araguaia Basin. Genetic studies on this species are still limited, making it difficult to evaluate the population structure and genetic diversity in natural and captive stocks. Here, we aimed to evaluate the transferability of heterologous microsatellite primers in B. gouldingi. A total of 30 primers for eight species were evaluated: Brycon hilarii, Brycon opalinus, Brycon cephalus, Brycon orbignyanus, Prochilodus lineatus, Prochilodus argenteus, Piaractus mesopotamicus, and Colossoma macropomum. The primers that showed the best amplification patterns were applied to 20 specimens of B. gouldingi, and their genetic parameters were assessed. Among the 30 primers, seven showed satisfactory transferability, six of which belonged to the genus Brycon: Bh13 (B. hilarii), BoM5, BoM13 (B. opalinus), Borg9, Borg13, and Borg59 (B. orbignyanus), and one belonged to P. argenteus (Par80). The primers for the other species tested showed non-specificity or monomorphism; and were therefore, excluded from the analyses. The number of alleles ranged between two (Borg13 and Borg59) and three (Bh13, BoM5, BoM13, Borg9 and Par80), with sizes varying between 103 bp (BoM5) and 430 bp (Borg9). Four primers showed evidence of null alleles (BoM13, Borg9, Borg13, and Par80), which could probably be attributed to the respective Hardy-Weinberg deviation. Thus, seven primers were validated for cross-amplification in B. gouldingi, which may be used in future studies involving this species.
