The objective of this study was to evaluate the relationship between the polymorphism within GHRH gene (GHRH/HaeIII) and milk production traits for Polish Holstein-Friesian using PCR-RFLP technique for genotyping. The molecular background of this mutation was defined as the A→C transversion at the intron 2. A total of 220 cows were examined. The following frequencies were established: 0.0227 for genotype AA, 0.3227 for AB, and 0.6546 for BB; 0.1841 for the allele GHRHA and 0.8159 for GHRHB. A highly significant association was found between the GHRH/HaeIII polymorphism and milk performance. The BB-genotype cows were characterised by a higher fat yield and percentage (P ≤ 0.01).

Bovine mastitis is a widespread disease of the mammary gland, highly contributing to the increase in veterinary costs in dairy industry. In the present study, the genetic polymorphism within bovine L-selectin gene was analysed and its impact on clinical mastitis occurrence, somatic cell score (SCS), and milk production traits in Polish Holstein-Friesian cows was examined. Polymorphism within L-selectin gene, molecule responsible for neutrophil attachment to endothelium, might have a potential role in immune response to bacterial infections and udder health. Two hundred and six Polish Holstein-Friesian cows were genotyped by polymerase chain reaction-restriction fragment length polymorphism method. Two single nucleotide polymorphisms mutations within the coding sequence of L-selectin gene were identified (c.165G>A and c.567C>T). The effect of c.165G>A and c.567C>T mutations on SCS was highly significant (P = 0.0019 and P = 0.0003, respectively). Strong associations (P ≤ 0.0001) were also observed between L-selectin polymorphism and milk production traits (milk yield, milk fat percentage, and milk protein percentage). However, the polymorphism in the analysed gene had no influence on the resistance or susceptibility of cows to clinical mastitis (only the tendency toward significance, P = 0.06 for c.567C>T mutation was found). Potential exploitation of the information on the identified associations in genetic selection needs to confirm the obtained results in further investigations.

PCR-RFLP analysis of PCR products were carried out using Aci I / SSi I for 100 Sahiwal and 115 Karan Fries cattle. In Sahiwal cattle and Karan Fries cattle, 438bp has three genotypes AA (438), AB (438+299+139 bp) and BB (299+139 bp). In Sahiwal cattle these genotypes are highly significant for FL305DPY but in Karan Fries cattle these genotypes are highly significant for FL305DMY, FLTMY, FL305DFY and FL305DPY. In Sahiwal cattle, mean ± SE of AA genotype for FL305DMY, FLTMY, FL305DFY, FL305DSNFY, FL305DPY were 1809.90 ± 15.7 kg, 2029.4 ± 15.6 kg, 99.90 ± 0.66 kg, 154.87 ± 0.17 kg and 44.81 ± 0.06 kg, respectively and for AB genotype were 1800.76 ± 9.48 kg, 1993.99 ± 9.42 kg, 100.54 ± 0.39 kg, 154.79 ± 0.10 kg, 43.99 ± 0.04 kg, respectively and for BB genotype were 1830.0 ± 14.10 kg, 2032.80 ± 14.0 kg, 100.24 ± 0.59 kg, 155.11 ± 0.15 kg, 42.98 ± 0.05 kg, respectively. Heterozygous AB genotype was found to be superior for, FL305DFY trait. AA genotype was significantly superior for FL305DPY traits whereas BB genotype was found to be superior for FL305DMY, FLTMY, FL305DSNFY. In Karan Fries cattle, the mean ± SE of AA genotype for FL305DMY, FLTMY, FL305DFY, FL305DSNFY, FL305DPY were 3442.17 ± 8.39 kg, 4461.93 ± 8.39 kg, 124.96 ± 7.20 kg, 277.35 ± 0.08 kg and 112.51 ± 0.08 kg, respectively and for AB genotype were 3572.69 ± 5.93 kg, 4592.45 ± 5.93 kg, 140.17 ± 5.09 kg, 278.60 ± 0.06 kg, 113.91 ± 0.05 kg, respectively and for BB genotypes were 3502.41 ± 9.19 kg, 4522.17 ± 9.19 kg, 136.91 ± 7.89 kg, 277.93 ± 0.09 kg, 113.19 ± 0.08 kg, respectively.

The aim of this study was to identify SNPs mutations in the CATHL2 gene and determine their potential association with dairy performance traits in Polish Black-and-White Holstein-Friesian (phf) cows. Genotypes of individuals were identified by PCR-RFLP. The frequencies of CATHL2/DdeI alleles were C ‒ 0.693 and T ‒ 0.307, and for CATHL2/HhaI polymorphisms, G ‒ 0.763 and C ‒ 0.237. The statistical analysis showed that cows with the CC (CATHL2/DdeI) and CG (CATHL2/HhaI) genotype produced higher milk yield than the other cattle genotypes. In the case of CATHL2/DdeI and CATHL2/HhaI polymorphisms, the highest somatic cell count was found in heterozygous CT and CG cows.

The correlation between polymorphisms in the IGF-1 gene and production traits in beef cattle is well known. The effect of insulin-like growth factor on the value of milk traits is not yet adequately understood. The aim of the study was to attempt to describe the effect of IGF-1/SnaBI substitution on selected milk performance parameters of the Black-and-White variety of Holstein-Friesian cows. Three genotypes were identified: CC, CT and TT. The results showed a correlation between IGF-1/SnaBI genotypes and milk yield (highest for CC homozygotes and lowest for CT heterozygotes). No relationship could be established between the genotype and the quality characteristics of milk.

Abstract The objective of this study was to examine associations between milking temperament, parity and milk production traits in primiparous and multiparous dairy cows. Twenty-one primiparous and nineteen multiparous Holstein Friesian cows were involved in the investigation on a Hungarian dairy farm. Cows’ stepping behaviour, milk yield and average milking speed were recorded once a month over an entire lactation, during morning milking. Milking temperament was scored by direct human observation on a 5-point-scale (1=very nervous, 5=very quiet) during udder preparation and milking. Multiparous cows showed a little more excitable behaviour at milking than during udder preparation (Mann-Whitney U=14165.00, P=0.032), and they were calmer at premilking preparation than primiparous cows (Mann-Whitney U=14046.00, P=0.001). Milking temperament of multiparous cows during udder preparation was associated with milking speed: nervous cows let down their milk slower (F=9.102, df= 1, P=0.003). Further experiments are needed to repeat the milking temperament test along with sensors measuring heart rate variability of cows in order to better understand the milking behaviour of cows.

The aim of this study was to evaluate the change of temperament of Holstein cows scored before and during milking during the entire lactation and to calculate correlation between temperament measured in the two moments as well as milk production traits and temperament. Observations were carried out on 17 primiparous Holstein Friesian cows starting their lactation within 85 days. Measurements of temperament and milk production traits were taken once in each month of the lactation, during the morning milking, in a milking parlour. Temperament was assessed in a 5-point scale during udder preparation and milking procedure (1: very nervous, 5: very quiet). Data of milk yield and milk flow were collected individually on each test day morning. Results showed changes during the lactation in temperament of cows scored directly before milking (P < 0.01), however, temperament during milking did not alter with months (P > 0.10). By correlation coefficients, behaviour assessed in the two moments cannot be regarded as the same. In addition loose, significant relations were found only between milk traits and temperament before milking.

Teat conformation characteristics, udder height, total milking yield, 2-min yield, 2-min yield adjusted for the linear and quadratic effects of total milking yield, and 305-day milk and fat yields were recorded on 377 Holstein-Friesian cows in 18 herds. Simple relationships of teat conformation measures and udder height to milk flow rate and production traits which were significant (P < 0.05) were as follows. Mean 2-min, total milking, and 305-day milk yields were lower for plumb front teats than for front teats that were not plumb. Cows with plumb rear teats also had lower total milking yields. Cows with cylindrical teats, both front and rear, had lower total milking yields than cows with funnel teats. Teat length was positively correlated with total milking, 305-day milk and 305-day fat yields. Teat diameter measures taken at proximal, medial and distal locations of the teat were positively correlated with all production traits, and rear teat diameter measures were positively correlated with 2-min yield. Udder height was positively correlated with adjusted 2-min yield and negatively correlated with total milking, 305-day milk and 305-day fat yields. Least squares analyses, which corrected for the effects of herd, age of cow at calving, and stage of lactation, were conducted, and significant (P < 0.05) effects were as follows. Cows with cylindrical teats produced 0.7–0.8 kg less total milk at milking than cows with funnel teats. Rear teat diameter was more highly related to milk production than front teat diameter. Proximal and distal diameter of the rear teat was positively associated with all three production traits, and medial rear teat diameter was positively associated with total milking and 305-day milk yields. Also, proximal diameter of the rear teat had a positive relationship with 2-min yield. Total milking yield was positively influenced by front teat proximal and medial diameters, but medial diameter had a negative effect on adjusted 2-min yield. Udder height influenced all traits. High udders had relatively higher milk flow rates but lower milk and fat production. Each centimetre increase in udder height was associated with a 46-kg decrease in 305-day milk. Heritabilities of 2-min yield, adjusted 2-min yield, 305-day milk yield and 305-day fat yield were 0.17, 0.27, 0.18, and 0.50, respectively.
