CATHL2 gene polymorphism in relation to production traits in Holstein-Friesian cows

The aim of this study was to identify SNPs mutations in the CATHL2 gene and determine their potential association with dairy performance traits in Polish Black-and-White Holstein-Friesian (phf) cows. Genotypes of individuals were identified by PCR-RFLP. The frequencies of CATHL2/DdeI alleles were C ‒ 0.693 and T ‒ 0.307, and for CATHL2/HhaI polymorphisms, G ‒ 0.763 and C ‒ 0.237. The statistical analysis showed that cows with the CC (CATHL2/DdeI) and CG (CATHL2/HhaI) genotype produced higher milk yield than the other cattle genotypes. In the case of CATHL2/DdeI and CATHL2/HhaI polymorphisms, the highest somatic cell count was found in heterozygous CT and CG cows.

The study included 124 Polish Black-and-White dairy cows of various share of the Holstein-Friesian (HF) breed. Lactoferrin (LTF) gene polymorphism was obtained with PCR-RFLP method using EcoRI enzyme. Two alleles of LTF, A and B, were found in the studied population. Their frequencies were 67.74% and 32.56%, respectively. The alleles controlled the occurrence of three genotypes: AA, BB and AB, of frequencies equal to 37.90%, 2.42% and 59.68%, respectively. It was established that statistically significant associations exist between the somatic cell count (SCC) and LTF genotype, lactation month and parity as well as the HF gene share. No significant association was found between somatic cell count and season. The highest somatic cell count (transformed to a logarithmic scale) was found in milk of the AB genotype, whereas the lowest one was found in cows of the AA genotype.

The correlation between polymorphisms in the IGF-1 gene and production traits in beef cattle is well known. The effect of insulin-like growth factor on the value of milk traits is not yet adequately understood. The aim of the study was to attempt to describe the effect of IGF-1/SnaBI substitution on selected milk performance parameters of the Black-and-White variety of Holstein-Friesian cows. Three genotypes were identified: CC, CT and TT. The results showed a correlation between IGF-1/SnaBI genotypes and milk yield (highest for CC homozygotes and lowest for CT heterozygotes). No relationship could be established between the genotype and the quality characteristics of milk.

Abstract. Associations between polymorphism of the bovine growth hormone (GH) gene (Leu/Val) and milk production traits of Black-and-White cattle with different proportional share of Holstein Friesian genes were analysed. A total of 1086 cows were included in the study. PCR-RFLP method was used for genotyping. The frequencies of genotypes and alleles were as follows: 0.653 for LL genotype, 0.324 for LV and 0.023 for VV, and 0.815 for GHL and 0.185 for GHV. Associations between Leu/Val polymorphism and milk production traits of cows were found only in first lactation. Cows with LL genotype had higher milk, fat and protein yield compared to LV individuals (P ≤ 0.01).

Abstract. The objective of this paper was to evaluate the relationship between the polymorphism of the GHRH and milk production traits of Polish Black-and-White. A total of 881 cows were included in the study. A PCR-RFLP method was used to genotyping. The frequencies of the genotypes and alleles were as follows: 0.0545 for AA, 0.3133 for AB and 0.6322 for BB, and 0.2111 for GHRHA and 0.7889 – GHRHB. There were no significant associations between GHRH/HaeIII polymorphism and milk production traits of the analysed cows.

The objective of this study was to evaluate the relationship between the polymorphism within GHRH gene (GHRH/HaeIII) and milk production traits for Polish Holstein-Friesian using PCR-RFLP technique for genotyping. The molecular background of this mutation was defined as the A→C transversion at the intron 2. A total of 220 cows were examined. The following frequencies were established: 0.0227 for genotype AA, 0.3227 for AB, and 0.6546 for BB; 0.1841 for the allele GHRHA and 0.8159 for GHRHB. A highly significant association was found between the GHRH/HaeIII polymorphism and milk performance. The BB-genotype cows were characterised by a higher fat yield and percentage (P ≤ 0.01).

Abstract. The aim of present study was to investigate the relationships between temperament score and milk production, as well as somatic cell count in a herd of Jersey and Holstein Friesian breeds. The temperament of 283 Jersey and 69 Holstein Friesian cows were assessed (scored) by the temperament score test (behaviour of animals was assessed in a 5-score system (1: calm, 5: nervous) while spending 30 s on the scale during weighing). The daily milk yield, fat, protein content and somatic cell count were also investigated in this study. Our investigation did not reveal any correlation between daily milk yield and temperament score. But milk somatic cell count was showed positive moderate relation with the temperament scores of Jersey (rrank=0.67; P=0.0001) and Holstein Friesian (rrank=0.66; P=0.0001) cows. Calmer cows had lower somatic cell count (Jersey: 135.40×103/cm3; Holstein Friesian: 176.07×103/cm3) compared to the more temperamental cows (Jersey: 540.44×103/cm3; P=0.0001; Holstein Friesian: 744.91×103/cm3; P=0.0001, resp.).

Abstract. Associations between polymorphism localised in the six exon of PIT1 gene (PIT1-HinfI) and milk production traits of Black-and-White cattle were analysed. A total of 900 cows were included in the study. PCR-RFLP method was used. The frequencies of the genotypes and alleles were as follows: 0.054 for AA, 0.377 for AB and 0.569 for BB, and 0.243 for PIT1A and 0.757 – PIT1B. There were no associations between PIT1-HinfI polymorphism and milk production traits of the cows.

<p class="abstrak2">Stearoyl-Coenzyme A desaturase 1 (SCD1) belongs to the fatty acid family of desaturases. In lactating ruminants, the SCD1 protein is highly expressed in the mammary gland and is relevant for the fatty acid composition of milk and dairy products. Polymorphism of SCD1 gene in Holstein-Friesian (HF) cows could be used as a basis of molecular selection of cattle in order to increase their productivity. The aim of this study was to investigate the polymorphism of SCD1 gene of Holstein-Friesian cows in Indonesia. A total of 162 blood samples of HF cows were collected from four different locations i.e. Bogor, Sukabumi, Tasikmalaya and Enrekang districts. Genotyping of SCD1 gene used PCR-RFLP method with NcoI restriction enzyme. The result showed that three genotypes (AA, AV and VV) and two alleles (A and V) have successfully found and polymorphic. A allele was dominant in all populations (0.63) and in Hardy Weinberg Equilibrium. The highest A allele was found in Sukabumi (0.78) and the lowest was in Bogor (0.55). Heterozigosity observed and expected reached 0.471 and 0.470, respectively. In conclusion, genetic polymorphism was found in all population with dominant of A allele. This finding can be used as a early genetic information of Holstein-Friesian cattle in Indonesia and to build breeding strategy for improving of productivity especially improving of healthy fat milk. </p><p class="abstrak2"><span><br /></span></p>
