Raman titration experiment with magnesium salt added gradually to aqueous solution of duplexes formed by RNA homopolynucleotides polyU and polyA was performed to reveal its effect on homopolynucleotide complexes. Statistical analysis of obtained spectral set has confirmed the effect already found by less structurally sensitive methods [Nucleic Acids Res.31(17) (2003), 5101–5107] that at sufficiently high concentrations magnesium causes transformation of polyU·polyA duplexes to polyU·polyA·polyU triplexes and single polyA strands. It was found that at relatively high polynucleotide concentrations used in Raman experiment, the threshold magnesium concentration for this effect is above the concentration of duplex basepairs in solution. Due to the strong spectral changes attributed to the varied percentages of duplexes, triplexes and single strands, it was not possible to register weaker direct Raman signs of the magnesium binding to polynucleotide strand.

ABSTRACT Intravenous infusion of isotonic magnesium chloride into young cats with a resultant mean plasma magnesium concentration of 7.7 meq./100 g protein was followed by a significant lowering of the plasma calcium concentration in 90 minutes. The rate of decrease of plasma calcium is consistent with the hypothesis that calcitonin is released by magnesium in high concentrations. There was no decrease in the plasma calcium concentration in cats of the same weight thyroparathyroidectomized 60 min before an identical magnesium chloride infusion or an infusion of isotonic sodium chloride at the same flow rate. The hypercalciuric effect of magnesium could not account for the hypocalcaemic effect of magnesium. Plasma magnesium concentration during magnesium infusion into cats with an intact thyroid-parathyroid gland complex was slightly, but not significantly higher than in acutely thyroparathyroidectomized cats.

Homeopathy is one of the applications in structured water influencing human health. The objective is to search for the physical basis of homeopathy. This methodology includes a study of absorption in the far IR spectral range, absorption and refraction in THz diapason, dynamic light scattering in the UV–near IR spectral range for the blinded samples of homeopathic-like preparations (HLP) of several parent substances and hidden/apparent controls, and statistical analysis on the significance of distinctions in spectral data between ensembles of HLP of each parent substance and ensembles of hidden/apparent controls. The analysis of nine independent blind studies of aqueous HLP of several parent substances gave statistically significant spectral differences in some preparations with an apparent control (25 comparisons of 35) and a hidden control (11 comparisons of 40). The revealed dominance in the occurrence frequency of differences in any HLP with an apparent control can be treated as involving most of these changes to the samples by the spectral measuring process. This allows interpreting the main mechanism of manifestation of the spectral changes found as the “observer effect”. The therapeutic effect of HLP may be assumed as a combination of the “observer effect” from the physician side and a “placebo effect” from the patient side.

A procedure is described for the isolation of nucleic acids from apple bark tissue using dimethyl sulfoxide. Nucleic acids isolated by this method are high in yield, and exhibit a high degree of purity as evidenced by their spectra. Experimental evidence is given to show that extraction of the tissue with dimethyl sulfoxide before the extraction of nucleic acids removes protein and pigments. Little or no ribonuclease activity could be detected in apple bark tissue after extraction with dimethyl sulfoxide. Ribonuclease activity was shown to be strongly inhibited by high concentrations of dimethyl sulfoxide.

Aim. Study of effect of heat-labile (HLT) and thermostable (HST) lethal toxins of Yersinia pseudotuberculosis on the development of embryos of sea urchin Strongylocentrotus intermedius, processes of biosynthesis of nucleic acids and protein in embryo cells and activity of nucleoside-kinases of sea urchin. Materials and methods. Y. pseudotuberculosis strains 2517 (pYV-) and 512 (pYV48MD, pYV82MD) were used for isolation of HLT and HST. Gametes and embryos of sea urchin S. intermedius were used to carry out the experiments and isolate nucleoside-kinases. Results. Both of the studied toxins of Y. pseudotuberculosis possessed spermiotoxic effect and reduced fertilizing ability of sea urchin spermies. HLT LD50 was 1 (ig/ml, and HST - 2 pg/ml. Toxins affected the development of embryos of sea urchin resulting in severe morphologic damages, cessation of the development of embryos at early stages of embryogenesis, destruction of cells and death of embryos. Wherein, damaging effect of HLT was observed at lower concentrations compared with HST. HLT inhibited DNA and RNA biosynthesis at concentrations of 1-2 pg/ml. HST did not affect biosynthesis of nucleic acids even at high concentrations, but inhibited protein biosynthesis in sea urchin embryos. HLT did not reduce the level of inclusion of labeled amino acids into embryo cells. HLT had inhibiting effect on the activity of thymidine- and uridine-kinase of sea urchin, whereas HST did not affect these enzymes. Conclusion. Both of Y. pseudotuberculosis protein toxins affect the development of sea urchin embryos, however, mechanisms of action of HLT and HST on embryos and processes occurring in them differ.
