This paper attempts to study and optimize the affinity partitioning conditions of papain in an aqueous two-phase system (ATPS). The effect of the amount of ionic liquids (ILs), the concentration of K2HPO4, temperature, pH, and the volume of papain solution were discussed concretely. The optimum conditions were determined as ionic liquid was 1.4 g and K2HPO4was 1.4 g, the extraction efficiency of papain could reach 98.33% with pH unadjusted. The temperature and the pH of the solution are major parameters that influence the partitioning of protein in ILs-based ATPSs. The partition of papain to the IL-rich phase was enhanced by increasing the amount of ILs, the concentration of K2HPO4, and temperature, especially at its isoelectric point.

The extraction of Zn(II) in an aqueous PEG (1550) - (NH4)2SO4 two-phase system as a function of several experimental parameters was studied. PEG-based aqueous two-phase systems are composed of two immiscible phases: a polymer-rich phase and a salt-rich phase, which can be used for extraction experiments. In the absence of a suitable extracting agent, for the system consisting of a mixture of equal volumes of 40 mass% PEG and 40 mass% (NH4)2SO4 aqueous solutions, Zn(II) remained predominantly in the salt-rich phase. Variation of the pH of the salt stock solution did not change very much the extraction efficiency. By adding chloride ions, an enhancement of the Zn(II) extraction was observed. The Zn(II) extraction efficiency in presence of Cl- depends on the acidity of the salt stock solution and on the concentration of chloride ions added into the system.

Ionic liquid aqueous two-phase system (ILATPS) was applied in the extraction and separation of hydrosoluble antibiotics. The partitioning behavior of cephalexin (CEX) in 1-butyl-3-methylimidazolium tetrafluoroborate ([Bmim]BF4)-ZnSO4aqueous two-phase system was studied by the partitioning parameter of the extraction efficiency. The effect of the volume of [Bmim]BF4, the concentration of ZnSO4, temperature, pH, and the volume of ZnSO4solution was discussed concretely. When the volume of [Bmim]BF4was 2 mL and the concentration of ZnSO4was 35%, the extraction efficiency of CEX could reach 92.64% with pH unadjusted. The effect of the volume of [Bmim]BF4on the extraction efficiency was higher than that of the concentration of ZnSO4. The temperature influenced not only the formation of aqueous two-phase system but also the extraction efficiency of CEX. The target was found to be preferentially extracted to the [Bmim]BF4-rich phase at the pH below 4.3. The partition of CEX to the top phase was enhanced by increasing the volume of [Bmim]BF4, the concentration of ZnSO4, and temperature; however, the partition of CEX to the top phase increased by decreasing the pH.

In this article, the partitioning behavior of C-phycocyanin (C-PC) in aqueous two-phase systems (ATPS) with different component was investigated to evaluate the applicability of ATPS for extracting high purity of C-PC without chromatography. Under the optimized conditions (pH = 6; PEG-2000, 16 %, w/w; potassium sodium tartrate 21 %, w/w), a maximum purity of 5.01 was achieved after the whole separation process and the yield of the C-PC was 76.94 %.

This work presents the experimental study results of Fe(III) and Zn(II) extraction by ionic liquids based on trioctylmethylammonium chloride and rhodanide in an aqueous two-phase system based on polypropylene glycol 425 and sodium chloride. Based on the results obtained, it was concluded that these extractants are effective for the extraction of metals from aqueous solutions, and aqueous two-phase systems represent a “green” alternative to traditional extraction systems.

An aqueous two-phase system (ATPS) was presented with hydrophilic ionic liquid 1-Butyl-3-methylimid-azolium tetrafluoroborate ([BmiBF4) and salts for the extraction of penicillin G in this paper. The phase forming abilities of different salts were investigated to choose a suitable salt.The partitioning behavior of penicillin G in the ATPS was investigated.Concentrations of (NH4)2SO4, penicillin G, and [BmiBF4 were evaluated to determine their effects on the extraction yield of penicillin G. It was found that extraction yield strongly depended on the concentration of [BmiBF4, penicillin and (NH4)2SO4. A high extraction yield around 93% was achieved with the following parameters: (NH4)2SO4 20% (wt%), penicillin 40000 ~ 45000 U/mL, [BmiBF4 20% (wt%).
