Plants from Gynura family was used in this study, namely,Gynura procumbensandGynura bicolor.Gynura procumbensis well known for its various medicinal properties such as antihyperglycaemic, antihyperlipidaemic, and antiulcerogenic; meanwhile,G. bicolorremains unexploited. Several nonenzymatic antioxidants methods were utilized to study the antioxidant capacity, which include ferric reducing antioxidant power (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, total flavonoid content, total phenolic content, and ascorbic acid content determination. DPPH assay revealsG. procumbensshoot as the lowest (66.885%) andG. procumbensroot as the highest (93.499%) DPPH radical inhibitor. In FRAP assay, reducing power was not detected inG. procumbensleaf callus (0.000 TEAC mg/g FW) wherebyG. procumbensroot exhibits the highest (1.103 TEAC mg/g FW) ferric reducing power. Total phenolic content and total flavonoid content exhibited similar trend for both the intact plants analysed. In all antioxidant assays,G. procumbenscallus culture exhibits very low antioxidant activity. However,G. procumbensroot exhibited highest phenolic content, flavonoid content, and ascorbic acid content with 4.957 TEAC mg/g FW, 543.529 QEµg/g FW, and 54.723 µg/g FW, respectively. This study reveals thatG. procumbensroot extract is a good source of natural antioxidant.

The antioxidant activity and content of phenolic substances in vegetable broths were determined. Green beans, beetroots, courgettes, onions, parsley, carrots, cabbages, celery, broccoli, spinach, cauliflowers, and tomatoes were subjected to boiling. Fresh vegetables and vegetable broths were analysed for ascorbic acid content, total phenolic content, ORAC and TEAC values. Phenolic acids were quantified using HPLC. The ascorbic acid content of vegetables ranged from 5–109 mg/100 ml, while no ascorbic acids could be detected in vegetable broths. Total phenolic content was between 17–1729 mg GAE/l for all samples. ORAC and TEAC values of vegetable broths were between 0–3 µmol TE/ml and 0–2 µmol TE/ml, respectively. Gallic, chlorogenic, caffeic, p-coumaric, and ferulic acid were detected in both fresh vegetables and vegetable broths. The highest phenolic acid content was observed in water in which beetroots were boiled. It was found that the vegetable broths of beetroots, celery stalks, cabbages, parsley and broccoli harboured remarkable antioxidant activity.

Plant based remedies are of much importance in healthcare due to low side effects. The effective constituents contained in seeds of a papaya have not been utilized efficiently in the production of medicines. The purpose of this study was driven towards determining the total phenolic content, total flavonoid content, antioxidant capacity, anti-inflammatory activity and antibacterial properties of the aqueous seeds extract of a Sri Lankan variety (Red Lady) of papaya (AESP). The AESP prepared according to the method of “Kasaya” in Ayurvedic medicine was used for the investigations according to standard procedures. Total phenolic content and the total flavonoid content of the AESP were 13.5±2.2 mg (pyrogallol equivalence)/g and 315.9±104.6 mg (quercetin equivalence)/g respectively. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of AESP ranged from 12.4% to 81.2% for concentrations of 1.0-20.0 mg/ml; ascorbic acid gave 31.7% - 91.9% activity. The AESP displayed strong reducing power compared to ascorbic acid in the reducing power assay. The hydroxyl radical scavenging activity of the AESP ranged from 6.6% to 69.1% for concentrations ranging from 20.0-95.0 mg/ml; ascorbic acid activity was 37.7%-74.9%. The nitric oxide radical scavenging activity of AESP was 23.9%-62.7% for concentrations of 2.0-20.0 µg/ml; ascorbic acid gave 26.8%-63.5% activity. AESP concentrations ranging from 75.0-150.0 µg/ml exhibited human red blood cell (HRBC) membrane stabilization protection of 15.5%-22.7% compared to 50.8%-58.4% for aspirin. The AESP showed antibacterial activity against Bacillus subtilis and Staphylococcus aureus bacterial species. AESP possess promising antioxidant, anti-inflammatory and antibacterial activity. The aqueous decoction of the discarded seeds of papaya would facilitate remedies for many diseases in which radicals are implicated as well as assist against certain bacterial infections and also has an anti-inflammatory potential.

Abstract Background: Nymphaea capensis is an aquatic flowering plant which is included in the family of Nymphaeaceae. Literature review on the plants of Nymphaeaceae family exhibited significant medicinal activities. Therefore, the objective of the present study is to evaluate possible anti-oxidant activity of crude methanol extract of N. capensis leaf.Method: In antioxidant study, methanolic plant extract was evaluated for 1,1-diphenyl,2-picrylhydrazyl (DPPH) and reducing power capacity. Moreover, total phenolic and total flavonoid content of plant extracts were determined and expressed in mg of gallic acid equivalent per gram of dry sample (mg GAE/g dry weight).Result: In the DPPH free radical scavenging assay, methanol extract showed concentration dependent inhibition of the free radicals. IC50 of Ascorbic acid was 14.84 µg/ml whereas N. capensis was 130.94 µg/ml. In case of reducing capacity, at conc. 62.5, 125, 250, 500, 1000 ug/ml, the absorbances of Ascorbic acid were 0.65, 1.12, 1.45, 1.78 and 1.89 respectively. In case of N. capensis, the absorbances were 0.46, 0.75, 1.04, 1.27 and 1.50 respectively. The extract displayed a concentration dependent increase in reducing power. In the case of total phenolic content, the methanol extract of N. capensis contained a considerable amount of phenolic contents which was 215±7 of GAE mg/g. In the case of total flavonoid content, methanol extract of N. capensis contained significant amount of flavonoid contents which was 184.75±6.78 of GAE mg/g.Conclusion: These results suggested that the methanol extract of N. capensis possess considerable anti-oxidant activity.

Phenolic compounds are a large group of phytochemical compounds in plants which have antioxidant activity. Hibiscus species were used in traditional medicine in North-America with biological activities. In this study, total phenolic content, total flavonoid content and antioxidant activities of Chaba maple (Hibiscus acetosella) flower extract (CFE) in Thailand were investigated. The total phenolic and flavonoid contents were determined by Folin-Ciocalteu assay and aluminum chloride colorimetric method, respectively. Antioxidant activities of CFE were examined by DPPH radical scavenging, ferric reducing antioxidant power (FRAP) and nitric oxide (NO) radical scavenging ability methods. The results showed that total phenolic content and total flavonoid content were 91.22±0.68 mg gallic acid equivalent/g fresh matter and 0.31±0.01 mg quercetin equivalent/g fresh matter, respectively. For antioxidant activities, IC50 of DPPH radical scavenging was 57.93±0.37 μg/ml, the power of ferric reducing was 1,707.18±1.99 μM and IC50 of NO radical scavenging was 229.30±0.71 μg/ml. In conclusion, CFE could be potentially used as a new natural bioactive ingredient for nutritional supplements and cosmetics application.

In the present study leaves and seeds of two different varieties of Fenugreek plant viz. Trigonella foenum gracium and Trigonella corniculata, collected from Bahawalpur, Pakistan were subjected to the biochemical, phytochemical and antioxidant analysis. The content of total sugars, non reducing sugars and reducing sugars in selected parts of these varities were found to be 5.007±0.101 to 8.133±0.223, 4.255±0.150 to 7.099±0.316 and 0.751±0.064 to 1.410±0.071 g/100g of dry weight respectively. Water soluble, salt soluble and total salt soluble protein content range from 1.172±0.001 to 6.015±0.132, 1.517±0.038 to 1.174±0.006 and 0.013±0.001 to 0.018±0.001 g/100 g of dry weight respectively and the total free amino acids ranged from 1.198±0.014 to 4.554±0.006 g/100 g of dry weight. The range of tannins and saponin contents in analyzed parts of these varities was found to be 0.057±0.009 to 0.117±0.025 and 0.083±0.012 to 0.0151±0.045 g /100g of dry weight respectively. Similarly, flavonoids and hydrogen cyanide content ranged from 0.021±0.000 to 0.055±0.004 and 0.0234±0.008 to 0.307±0.009 g/100g of dry weight respectively; while ascorbic acid content was found to be in the range of 0.068±0.08 to 0.267±0.031 g/100g of dry weight. Total phenolic acid content of the selected parts of these varities were estimated on the basis of difference in dipole moment and its range was observed from 0.0539±0.024 to 0.856±0.064 g/100g of dry weight. The total antioxidants analysis showed that ascorbic acid and trolox contents (mg Eqv./100g of extract) were found to be lower in Trigonella corniculata seeds (0.066±0.001 and 0.047±0.001 mg Eqv./100g respectively) and higher in Trigonella corniculata leaves (0.069±0.001 and 0.050±0.001 mg Eqv./100g respectively).The DPPH radical scavenging activity and reducing power were found to be in the range of 31.736±2.430 to 39.590±2.432 and 0.974±0.231 to 2.301±0.068 respectively. The different parts of studied varities of fenugreek plant, having higher values of %DPPH inhibition and reducing power showed least count of IC50 and vice versa.

Drought stress is a serious threat for cocoa production. One of the plant mechanisms to survive from the drought stress is through producing some biochemical compounds. The objective of this research was to know biochemical changes in three cocoa clones during drought stress. The research was conducted at the green house of Indonesian Coffee and Cocoa Research Institute from January – December 2017. The experiment was designed by using Randomized Complete Block Design (RCBD) with 2 factors and with 3 replications. The first factor was clones of scion that consisting KW 641, Sulawesi 1 and ICS 60 which each of its grafted to Scavina 6 clone as rootstock. The second factor was watering intervals which consisting; every 2 days / regularly watering (adequate of water), every 5 days (moderate drought), and every 8 days (severe drought). The result of this research showed that drought stress decreased chlorophyll content, increased hydrogen peroxide content (ROS), increased the activity of superoxide dismutation (SOD), increased ascorbic acid content, increased total phenolic content, and increased proline content. Sulawesi 1 and KW 641 clones showed lower hydrogen peroxide (ROS) content, while chlorophyll content, SOD activity, total phenolic content, ascorbic acid content, and proline content were higher compared with ICS 60 clone. It showed that Sulawesi 1 and KW 641 clones had more drought-resistance compared with ICS 60 clone.

Objectives: The aim of the current study was to perform phytochemical screening and determination of total phenolics, total flavonoids and antioxidant activity of various crude extracts of Alternanthera sessilis red leaves.Methods: Determination of antioxidant nature of Alternanthera sessilis red was carried out by DPPH radical scavenging method. GC-MS analysis of various crude extracts resulted in the presence of different types of low and high molecular weight compounds consisting of carbohydrates, fatty acid and vitamins.Results: Among all the extracts, methanol extracts exhibited higher DPPH radical scavenging activity with IC50 value of 0.183 mg/ml. BHT used as the positive control showed IC50 value of 0.089 mg/mL. The results suggested that the crudes extracts of Alternanthera sessilis red exhibit moderate antioxidant activity. The total phenolic content (279.8 ± 0.02 mg GAE/g) and total flavonoid content (250.7 ± 0.03 mg QE/g) in methanol was estimated higher as compared to other solvents.Conclusion: The total flavonoid content and total phenolic content were  found to be higher in methanolic extract followed by ethyl acetate  and hexane extracts.

The flowers of Rhododendron arboreum Smith is a source of polyphenolic compounds. A flavonol, quercetin, was isolated from ethyl acetate fraction of the methanol extract of flower petals of R. arboreum by repeated Sephadex LH-20 column chromatography. Isolated quercetin was characterized by comparing melting point, Rf values, UV and IR spectra with authentic quercetin. The isolated quercetin was used as a standard for the estimation of total flavonoids.  Total phenolic and total flavonoid content in different parts of R. arboreum was carried out spectrophotometrically using Folin-Ciocalteu reagent and Aluminium chloride reagent respectively. Gallic acid and quercetin were used as standard for the construction of calibration curve of phenolic and flavonoid respectively. The results showed that the highest total phenolic content was detected in the 70% acetone extract of the flowers (600 mg GAE/g extract) and petals (600 mg GAE/g extract) and the lowest amount was detected in methanol extract of stem (188 mg GAE/g extract). Similarly, the highest total flavonoid was detected in the 70% acetone extract of the twigs (170 mg QE/g extract) and the lowest amount was detected in the methanol extract of stem (45 mg QE/g extract).  The antioxidant activity of the methanol extracts obtained from different parts of R. arboreum was determined by 2, 2- diphenyl-1-picryl hydrazyl (DPPH) assay and the radical scavenging activity (IC50) was calculated. The highest free radical scavenging effect was observed in leaves with IC50=8.34 ?g/ml and lowest was observed in stem with IC50=67.83 ?g/ml. The IC50 values, total phenolic and total flavonoid content (correlation coefficient R2= 0.923 for phenolic, R2= 0.965 for flavonoid) were correlated which showed strong correlation indicating that the major components responsible for antioxidant activity is phenolics. The highest the phenolic content, the lowest the IC50 value observed. The result indicated that R. arboreum is a rich source of high value polyphenols as natural antioxidant to use in preventive medicine as well as in food and pharmaceutical industry.Scientific World, Vol. 12, No. 12, September 2014, page 34-40
