The rs1277306 Variant of the REST Gene Confers Susceptibility to Cognitive Aging in an Elderly Taiwanese Population

Background/Aims: There is growing evidence that the RE1-silencing transcription factor (REST) gene may contribute to cognitive aging and Alzheimer diseases. In this replication study, we reassessed whether single nucleotide polymorphisms (SNPs) within the REST gene are linked with cognitive aging independently and/or through complex interactions in an older Taiwanese population. Methods: A total of 634 Taiwanese subjects aged over 60 years from the Taiwan Biobank were analyzed. Mini-Mental State Examination (MMSE) scores were performed for all subjects to weigh cognitive functions. Results: Our data showed that the REST rs1277306 SNP was significantly associated with cognitive aging among all subjects (p = 0.0052). Furthermore, the association remained significant for individuals without APOE ε4 allele (p = 0.0092), but not for individuals with at least 1 APOE ε4 allele. This association remained significant after Bonferroni correction. Additionally, we found the interactions between the rs1713985 and rs1277306 SNPs on cognitive aging (p = 0.016). However, the 3-marker haplotype derived from the rs1713985, rs3796529, and rs7680734 SNPs in the REST gene demonstrated no association with cognitive aging. Conclusion: Our study indicates that the REST gene may contribute to susceptibility to cognitive aging independently as well as through SNP-SNP and APOE-REST interactions.

Η παρούσα έρευνα αποσκοπούσε να εξετάσει την άμεση επίδραση του άγχους εξέτασης στη Δοκιμασία Σύντομης Γνωστικής Εκτίμησης (ΔΣΓΕ, MMSE Mini Mental State Examination), καθώς και την τυχόν έμμεση επίδραση της ηλικίας και βασικών βιοδεικτών που συνδέονται με το γνωστικό γήρας στη ΔΣΓΕ, μέσω του άγχους εξέτασης. Στην έρευνα συμμετείχαν 60 άτομα και των δύο φύλων, ηλικίας άνω των 55 ετών. Οι συμμετέχοντες κλήθηκαν να απαντήσουν αρχικά σε μια σειρά ερωτήσεων που αφορούσαν τα ατομικάδημογραφικά τους χαρακτηριστικά καθώς και το αν έχουν διαγνωστεί ή μη ως υπερτασικοί, υπερχοληστεριναιμικοί ή/και διαβητικοί. Στη συνέχεια, τους ζητήθηκε να απαντήσουν σε ερωτηματολόγιο σχετικό με το άγχος που γενικά νιώθουν πριν, κατά τη διάρκεια και μετά από μια εξέταση των γνωστικών τους ικανοτήτων. Οι γνωστικές τους ικανότητες εξετάστηκαν μέσω της ΔΣΓΕ. Η στατιστική ανάλυση των δεδομένων που συγκεντρώθηκαν, έδειξε ότι το άγχος εξέτασης επιδρά στην επίδοση στη ΔΣΓΕ. Ειδικότερα, βρέθηκε ότι από τις δύο βασικές διαστάσεις του άγχους εξέτασης μόνον η συναισθηματικότητα επιδρά άμεσα κι αρνητικά στην επίδοση στη ΔΣΓΕ, ενώ η ανησυχία όχι. Η ηλικία και ο βιοδείκτης «διαβήτης» φάνηκε ότι επιδρούν επίσης αρνητικά στην επίδοση στη ΔΣΓΕ. <br />στόσο η επίδρασή τους είναι μόνον άμεση και όχι έμμεση, μέσω του άγχους εξέτασης. Μάλιστα, ο διαβήτης επιδρά άμεσα τόσο στη ΔΣΓΕ όσο και στη συναισθηματικότητα, εξαλείφοντας την επίδραση της συναισθηματικότητας στη ΔΣΓΕ.

We examined 510 subjects representing 83.2% of all citizens of a Finnish city aged 85 years or over. Mini-Mental State Examination (MMSE) scores, diagnosis of dementia by DSM-III-R criteria, and Apo-E genotype were determined. The prevalence of dementia was 38.6%. The odds ratio (OR) of the Apo-E ε4 carriers (with the reference population of people with the genotype ε3/ε3) for dementia was 2.36 (95% CI 1.58–3.58). There was a significant sex difference: The OR in women was 3.23 (95% CI 2.02–5.17) whereas among men it was insignificant. The mean MMSE score (± SD) among the Apo-E ε4 carriers (15.0 ± 10.0) and noncarriers (18.7 ± 8.6) (p < .001) differed among the whole population, but not within the demented or nondemented subjects analyzed separately. This study does not support the hypothesis that the Apo-E ε4 allele impairs cognitive functions of nondemented elderly, at least in those surviving to very old age.

Background: Both leisure activities and the ε4 allele of the apolipoprotein E (APOE ε4) have been shown to affect cognitive health. We aimed to determine whether engagement in leisure activities protects against APOE ε4-related cognitive decline.Methods: We used the cohort data from the Chinese Longitudinal Healthy Longevity Survey. A total of 3,017 participants (mean age of 77.0 years, SD = 9.0; 49.3% female) from 23 provinces of China were recruited in 2008 and were reinterviewed in 2014. We assessed cognitive function using the Mini-Mental State Examination (MMSE). We calculated cognitive decline using subtraction of the MMSE score of each participant in 2008 and 2014. We genotyped a number of APOE ε4 alleles for each participant at baseline and determined the Index of Leisure Activities (ILAs) by summing up the frequency of nine types of typical activities in productive, social, and physical domains. We used ordinal logistic regression models to estimate the effects of leisure activities, APOE ε4, and their interaction on cognitive decline, statistically adjusted for a range of potential confounders.Results: There were significant associations between APOE ε4 and faster cognitive decline, independent of potential confounders, and between leisure activities and mitigated cognitive decline. The odds ratios were 1.25 (95% CI: 1.03, 1.53) and 0.93 (95% CI: 0.89, 0.97), respectively. We found significant interactions of APOE ε4 with leisure activities with a P-value of 0.018. We also observed interactive effects of subtypes of leisure activities: participants who regularly engaged in productive activities were more likely to reduce the risk of APOE ε4-related cognitive decline.Conclusion: Our findings provide support for the indication that participating in leisure activities reduces the risk of APOE ε4-related cognitive decline.

Education, sex, and the APOE-rs405509 variant are associated with Alzheimer’s disease and cognitive performance. We investigated if the rs405509 TT, TG, and GG genotypes modulate the effect of sex and education on cognitive impairment in Taiwanese adults. Data on cognitive health (defined by Mini-Mental State Examination (MMSE) scores) and rs405509 were from Taiwan Biobank. Participants included 2105 men and 2027 women with a mean age of 64 years. Education below university level was significantly associated with lower MMSE scores. The odds ratios (ORs) were 1.82; 95% confidence interval (CI) 1.38–2.41 for senior high school, 3.39; 95% CI 2.50–4.59 for junior high school, and 11.94; 95% CI 9.91–15.50 for elementary school and below (p-trend < 0.05). The association between MMSE score and sex was significant only in the lowest educational group (elementary and below), with lower odds of having a low MMSE score in men compared to women (OR = 0.51; 95% CI 0.34–0.77). After stratification by rs405509 genotypes, this association was significant only among TT genotype carriers (OR = 0.481; CI = 0.253–0.915). In conclusion, a significant association between MMSE score and sex was observed in the lowest educational group, especially among carriers of rs405509 TT genotypes.

      RELATIONSHIP OF MARITAL STATUS, APOE ε4, AND TYPES OF PHYSICAL ACTIVITIES ON COGNITIVE DECLINE IN ELDERLY WOMENABSTRACTIntroduction: The number of elderly population in Indonesia continues to increase every year, especially women. Elderly women are at higher risk of experiencing decreased cognitive function.Aims: To determine the relationship between the characteristics  of the subject, vascular risk factors, APOE ε4 genotype, and the type of physical activity with the decline of cognitive function in elderly women.Methods: A cognitive cohort study of 114 elderly women who had been observed for a mean of ±2.5 years, as part of the Active Aging study at Atma Jaya Catholic University. The independent variables of this study included were age, education, marital status, vascular risk factors (hypertension, diabetes, dyslipidemia, and BMI), APOE ε4, and types of physical activity. Cognitive function is measured using Mini Mental State Examination (MMSE).Results: There were 114 subjects with mean age was 71.22±7.297 and 33.3% experienced a decline in cognitive function. Variabel bebas penelitian ini meliputi usia, pendidikan, status perkawinan, faktor risiko vaskuler (hipertensi, diabetes, dislipidemia, dan IMT), APOE ε4, dan jenis aktivitas fisik. Fungsi kognitif diukur menggunakan Mini Mental State Examination (MMSE).Hasil: Didapatkan 114 subjek dengan rerata usia 71,2±7,3 dan 33,3% mengalami penurunan fungsi kognitif. Analisis multivariat menunjukkan usia, status perkawinan (single) , dan jenis aktivitas fisik (memasak) memiliki hubungan bermakna dengan risiko relatif (RR) masing-masing 4,46; 0,12; dan 0,334.Diskusi: Faktor risiko gangguan fungsi kognitif pada lansia perempuan adalah usia.
