Distribution of Macrobiotus shonaicus Stec, Arakawa & Michalczyk, 2018 (Tardigrada: Eutardigrada: Macrobiotidae) in Japan

To date, only seven species of Macrobiotidae (Parachela; Eutardigrada; Tardigrada) have been reported from Japan, including the recently described Macrobiotus shonaicus Stec et al., 2018 from the Shonai region of Japan. This species has flexible filaments on the egg processes and is known to proliferate only through sexual reproduction. Here, we report a multifaceted analysis of nine populations of M. shonaicus found on four Japanese islands. DNA sequencing of the cytochrome oxidase subunit I (COI) and internal transcribed spacer 2 (ITS-2) of the nine populations revealed 8 and 11 haplotypes, respectively. The extensive morphometric analysis showed considerably greater variability in the morphology of eggs than of animals. In addition to the morphological and molecular data, we confirmed the karyotype and found that all populations had a chromosome number of n = 6. Moreover, we observed and filmed mating behaviour between all studied populations of M. shonaicus. Our results clearly indicated that M. shonaicus is widely distributed throughout Japan.

AbstractIn this paper, the Clusiidae (Diptera: Schizophora) is analyzed phylogenetically using morphological and molecular data sets, and then redefined on the basis of derived morphological characters. The biology and distribution of the Clusiidae are also reviewed, a key is provided to the World genera, the status of the genus Craspedochaeta Czerny is reevaluated and the type of Heterochroa pictipennis Wulp is discussed. Molecular data sets include genomic DNA sequences from the mitochondrial genes COI (cytochrome oxidase subunit I) and COII (cytochrome oxidase subunit II), the large ribosomal nuclear subunit 28S, and the nuclear protein-coding carbomoylphosphate synthase (CPS) domain of CAD (or “rudimentary”). Genes were analyzed separately, in combination with each other, and in combination with a morphological data set. Although individual molecular data sets often provided conflicting phylogenetic signals, the topologies of the cladograms produced from each data set alone or in combination were largely similar. Most genus-level relationships and several basal divergences were unresolved, but Apiochaeta was very strongly and consistently supported as Sobarocephalinae, not Clusiinae. The Clusiinae and Sobarocephalinae are subsequently redefined using an adjusted morphological tree — retaining Apiochaeta in the Sobarocephalinae — that is only slightly longer (8.4%, or seven steps) than the most parsimonious tree. Our results illustrate the benefits of multiple independent data sets for phylogenetic reconstruction in order to verify and refine existing classifications.

After reconstructing the phylogenetic relationships among the closely related genera Carbula Stål, Eysarcoris Hahn, and Stagonomus Gorski (tribe Eysarcorini), using the mitochondrial genes: 16S rRNA and cytochrome oxidase subunit I (COI), and incorporating morphological characters to contrast our data, we transfer the species Eysarcoris gibbosus Jakovlev and Eysarcoris venustissimus (Schrank) to the genus Stagonomus, and we propose to place the subgenus Dalleria Mulsant & Rey as a synonym under the nominal subgenus Stagonomus. Additionally, Stagonomus is redefined to include diagnostic characters of the male genitalia.

Chironomids of the genus Pagastia Oliver (Diamesinae, Diamesini) from the mountains of Central Asia are revised using both morphological characters and molecular data. Illustrated descriptions of the adult male Pagastia (P.) caelestomontana sp. nov. from Kirgizstan and Tajikistan, P. (P.) hanseni sp. nov. from Tajikistan, and record of a finding apparently a new species P. (P.) aff. lanceolata (Tokunaga) from Tajikistan as well as an updated a key to the determination of the adult males of all known species of Pagastia are provided. A phylogenetic framework is reconstructed based on two mitochondrial genes cytochrome oxidase subunit I (COI) sequences of 34 samples belonging to 7 species of the genus Pagastia and cytochrome oxidase subunit II (COII) available for most samples. Phylogenetic trees of some known species of the genus Pagastia were reconstructed using the combined dataset and Bayesian inference (BI) and Maximum Likelihood (ML) methods. The interspecific K2P distances between seven Pagastia species including P. (P.) caelestomontana sp. nov., P. (P.) hanseni sp. nov. and undescribed P. (P.) aff. lanceolata (Tokunaga) are 6.3–13.2 which corresponding to species level.

Citrus, an important cash crop in India, is adversely affected by Phytophthora nicotianae, P. palmivora, and P. citrophthora. Phytophthora insolita is known to be associated with citrus and reported for the first time in India. It is a rare and poorly characterized Phytophthora species, as its natural host and pathogenic impact are unclear. Previously, it was reported only in Taiwan and China; so to confirm our suspected isolate is P. insolita, regions of internal transcribed spacer, elongation factor, beta-tubulin, and cytochrome oxidase genes were sequenced. This study provides description of the lone Indian P. insolita isolate with respect to molecular identity, morphology, mating behaviour, and pathogenicity.

<em>Cortinarius gueneri</em> (subgen. <em>Telamonia</em> sect. <em>Laeti</em>) is described as a new species from Turkey based on morphological and molecular data. It can be recognized by the somewhat irregular conical to campanulate, translucent-striate, rusty brown pileus; pale yellowish to cinnamon brown broadly attached lamellae; pale salmon to pinkish brown stipe; narrowly amygdaloid to ellipsoid basidiospores; and its putative association with <em>Carpinus orientalis</em>. Full description of the new species is given with field photos, microscopic illustrations, and a short discussion. Phylogenetic analysis of the internal transcribed spacer (ITS) region is also provided.

A new species of fiddler crab, Uca jocelynae sp. nov., belonging to the U. vocans complex, is described from the islands in Western Pacific on the basis of morphological and molecular data. The new species was previously identified with U. neocultrimana (Bott, 1973) (= U. vocans pacificensis Crane, 1975). The two species can be readily distinguished by characters of the male major chelae, carapace features, and the form of the vulvae. The molecular data of parts of the mitochondrial 16S rRNA and cytochrome oxidase I (COI) genes support that both are sister taxa but nevertheless distinct enough to be regarded as separate species. Uca jocelynae is widely distributed from the islands in the Western Pacific, while U. neocultrimana occurs in Fiji and eastwards.

Three new species of the genus Dendronotus are described from the North Pacific waters of Japan and Russia. For the first time since 1949, three new species, D. jamsteci sp. nov., D. zakuro sp. nov. and D. bathyvela sp. nov. are described from the waters of the Japanese Islands. D. zakuro sp. nov. was also discovered in the waters of Russia (Kamchatka). The descriptions of the new species D. jamsteci sp. nov. and D. zakuro sp. nov. are based on morphological and molecular data; D. bathyvela sp. nov. morphologically belongs to the group of D. robustus, D. velifer and D. patricki. This study revises our knowledge of North Pacific species of the genus Dendronotus, considerably expands the number of species worldwide and contributes to the multilevel fine-scale diversity concept.

The taxonomy of Neurachninane has been unstable, with its member genera consisting of Ancistrachne, Calyptochloa, Cleistochloa, Dimorphochloa, Neurachne, Paraneurachne and Thyridolepis, changing since its original circumscription that comprised only the latter three genera. Recent studies on the phylogeny of Neurachninae have focused primarily on molecular data. We analysed the phylogeny of Neurachninae on the basis of molecular data from seven molecular loci (plastid markers: matK, ndhF, rbcL, rpl16, rpoC2 and trnLF, and ribosomal internal transcribed spacer, ITS) and morphological data from 104 morphological characters, including new taxonomically informative micromorphology of upper paleas. We devised an impact assessment scoring (IAS) protocol to aid selection of a tree for inferring the phylogeny of Neurachninae. Combining morphological and molecular data resulted in a well resolved phylogeny with the highest IAS value. Our findings support reinstatement of subtribe Neurachninae in its original sense, Neurachne muelleri and Dimorphochloa rigida. We show that Ancistrachne, Cleistochloa and Dimorphochloa are not monophyletic and Ancistrachne maidenii, Calyptochloa, Cleistochloa and Dimorphochloa form a new group, the cleistogamy group, united by having unique morphology associated with reproductive dimorphism.

Parasola psathyrelloides sp. nov. is described from Kerala State, India, based on both morphological and molecular data. Comprehensive description, photographs, and comparisons with morphologically similar and phylogenetically related species are provided. Sequences of both the nuclear ribosomal internal transcribed spacer region (ITS) and the nuclear ribosomal large subunit (nLSU) region of the new species were obtained and used in BLASTn searches. The phylogenetic study was based on Maximum likelihood (ML) analysis of the ITS sequences.
