FQ Columnist Paul Julian Smith explores the latest trends in Mexican cinema, which encompasses such divergent genres as the rom-com and horror. Illustrating the former is the office comedy Mirreyes vs. Godinez (dir. Chava Cartas, 2019), which pits the spoiled offspring of the leisured class against the workers at their family company, a class conflict that predictably resolves through romantic alliances. In stark contrast is Belzebuth (dir. Emilio Portes, 2017), a disturbing film about the Satanic murder of children set in the tense and traumatic territory of Mexico's border with the United States. Finally, Smith looks at two productions—one an independent film, the other a televised sit-com—that use narratives about house shares to explore the theme of national reconciliation.

Asian Euwallacea ambrosia beetles vector Fusarium mutualists. The ambrosial fusaria are all members of the ambrosia Fusarium clade (AFC) within the Fusarium solani species complex (FSSC). Several Euwallacea–Fusarium mutualists have been introduced into nonnative regions and have caused varying degrees of damage to orchard, landscape, and forest trees. Knowledge of symbiont fidelity is limited by current identification methods, which typically requires analysis of DNA sequence data from beetles and the symbionts cultured from their oral mycangia. Here, polymerase chain reaction (PCR)-based diagnostic tools were developed to identify the six Fusarium symbionts of exotic Euwallacea spp. currently known within the United States. Whole-genome sequences were generated for representatives of six AFC species plus F. ambrosium and aligned to the annotated genome of F. euwallaceae. Taxon-specific primer-annealing sites were identified that rapidly distinguish the AFC species currently within the United States. PCR specificity, reliability, and sensitivity were validated using a panel of 72 Fusarium isolates, including 47 reference cultures. Culture-independent multiplex assays accurately identified two AFC fusaria using DNA isolated from heads of their respective beetle partners. The PCR assays were used to show that Euwallacea validus is exclusively associated with AF-4 throughout its sampled range within eastern North America. The rapid assay supports federal and state agency efforts to monitor spread of these invasive pests and mitigate further introductions.

John Cassavetes (b. 1929–d. 1989) was an American actor and filmmaker who wrote, directed, and acted in a catalogue of independent films he made over a forty-year career. Cassavetes directed twelve films—thirteen if one considers Shadows (1958) and Shadows (1959) as distinct works. With a close group of actors and crew, his works often featured Gena Rowlands (b. 1930), Seymour Cassel (b. 1935), Peter Falk (b. 1927–d. 2011), and Ben Gazzara (b. 1930–d. 2012). Despite early praise of Shadows (1958) by writers such as Jonas Mekas, reviewers were often unfavorable, uninterested, and/or unkind to the majority of Cassavetes’s films. Cassavetes won no major awards in the United States, though Rowlands, his chief collaborator and his wife, was nominated for an Academy Award twice, for her performances in his films A Woman Under the Influence (1974) and Gloria (1980). Cassavetes did receive a steady run of accolades near the end of his life. At that point, scholars and critics began to consider his works more expansively in terms of their aesthetics—including oft-cited characteristics of a certain brand of realism, naturalism, and echoes of cinéma vérité. The last three films of his career included two made within the studio system—Gloria (1980) and Big Trouble (1986). Released between these, Love Streams (1984), Cassavetes’s last independent film, was also well received in the United States, and it won first place in the Berlin Film Festival. He died in 1989 of cirrhosis of the liver. For the most part, scholarly writings about the films of John Cassavetes did not appear until the 1980s and 1990s. They have been in large part spearheaded and expanded upon by a small number of authors. Prominent among those writers is Ray Carney. This article began under Carney’s advisorship, and his early input (2011–2012) helped shape its scale and scope. It is not a comprehensive listing and annotation of the writings, but it represents a selection highlighting major trends and directions of response and examination.

Outbreaks of blastomycosis, caused by the fungus Blastomyces dermatitidis, occur in endemic areas of the United States and Canada but the geographic range of blastomycosis is expanding. Previous studies inferred the location of B. dermatitidis through epidemiologic data associated with outbreaks because culture of B. dermatitidis from the environment is often unsuccessful. In this study, we used a culture-independent, PCR-based method to identify B. dermatitidis DNA in environmental samples using the BAD1 promoter region. We tested 250 environmental samples collected in Minnesota, either associated with blastomycosis outbreaks or environmental samples collected from high- and low-endemic regions to determine basal prevalence of B. dermatitidis in the environment. We identified a fifth BAD1 promoter haplotype of B. dermatitidis prevalent in Minnesota. Ecological niche analysis identified latitude, longitude, elevation, and site classification as environmental parameters associated with the presence of B. dermatitidis. Using this analysis, a Random Forest model predicted B. dermatitidis presence in basal environmental samples with 75% accuracy. These data support use of culture-independent, PCR-based environmental sampling to track spread into new regions and to characterize the unknown B. dermatitidis environmental niche.Importance Upon inhalation of spores from the fungus Blastomyces dermatitidis from the environment, humans and animals can develop the disease blastomycosis. Based on disease epidemiology, B. dermatitidis is known to be endemic in the United States and Canada around the Great Lakes and in the Ohio and Mississippi River Valleys but is starting to emerge in other areas. B. dermatitidis is extremely difficult to culture from the environment so little is known about the environmental reservoirs for this pathogen. We used a culture-independent PCR-based assay to identify the presence of B. dermatitidis DNA in soil samples from Minnesota. By combining molecular data with ecological niche modeling, we were able to predict the presence of B. dermatitidis in environmental samples with 75% accuracy and to define characteristics of the B. dermatitidis environmental niche. Importantly, we showed the effectiveness of using a PCR-based assay to identify B. dermatitidis in environmental samples.

ABSTRACT The rapidly growing supplement industry operates without a formal premarket approval process. Consumers rely on product labels to be accurate and true. Those products containing live microbials report both identity and viability on most product labels. This study used next-generation sequencing technology as an analytical tool in conjunction with classic culture methods to examine the validity of the labels on supplement products containing live microbials found in the United States marketplace. Our results show the importance of testing these products for identity, viability, and potential contaminants, as well as introduce a new culture-independent diagnostic approach for testing these products. Millions of people consume dietary supplements either following a doctor’s recommendation or at their own discretion to improve their overall health and well-being. This is a rapidly growing trend, with an associated and expanding manufacturing industry to meet the demand for new health-related products. In this study, we examined the contents and microbial viability of several popular probiotic products on the United States market. Culture-independent methods are proving ideal for fast and efficient analysis of foodborne pathogens and their associated microbial communities but may also be relevant for analyzing probiotics containing mixed microbial constituents. These products were subjected to next-generation whole-genome sequencing and analyzed by a custom in-house-developed k-mer counting method to validate manufacturer label information. In addition, the batch variability of respective products was examined to determine if any changes in their formulations and/or the manufacturing process occurred. Overall, the products we tested adhered to the ingredient claims and lot-to-lot differences were minimal. However, there were a few discrepancies in the naming of closely related Lactobacillus and Bifidobacterium species, whereas one product contained an apparent Enterococcus contaminant in two of its three lots. With the microbial contents of the products identified, we used traditional PCR and colony counting methods to comparatively assess our results and verify the viability of the microbes in these products with regard to the labeling claims. Of all the supplements examined, only one was found to be inaccurate in viability. Our use of next-generation sequencing as an analytical tool clearly demonstrated its utility for quickly analyzing commercially available products containing multiple microbes to ensure consumer safety. Othello (1989) is a little-known independent film directed and produced by Ted Lange. Released at the end of the 1980s, a decade of mounting racial tension in the United States, the film offers an interpretation of Shakespeare from a minority point of view. This examination of the director’s motives and directorial choices reveals the strategies employed by Lange to make Shakespeare relevant and more available to a non-traditional audience, especially to African-Americans. In addition, delving into the acceptance and distribution difficulties that Ted Lange faced gives insight into the issue of discriminatory assumptions related to applying a minority point of view to a traditionally highbrow domain.

Prostate adenocarcinoma is the most common malignant tumor of men in the United States and is the third leading cause of death in men. Despite attempts at early detection, there will be 244,000 new cases and 44,000 deaths from the disease in the United States in 1995. Therapeutic progress against this disease is hindered by an incomplete understanding of prostate epithelial cell biology, the availability of human tissues for in vitro experimentation, slow dissemination of information between prostate cancer research teams and the increasing pressure to “ stretch” research dollars at the same time staff reductions are occurring.To meet these challenges, we have used the correlative microscopy (CM) and client/server (C/S) computing to increase productivity while decreasing costs. Critical elements of our program are as follows:1) Establishing the Western Pennsylvania Genitourinary (GU) Tissue Bank which includes >100 prostates from patients with prostate adenocarcinoma as well as >20 normal prostates from transplant organ donors.

In recent years it became apparent that we needed to improve productivity and efficiency in the Microscopy Laboratories in GE Plastics. It was realized that digital image acquisition, archiving, processing, analysis, and transmission over a network would be the best way to achieve this goal. Also, the capabilities of quantitative image analysis, image transmission etc. available with this approach would help us to increase our efficiency. Although the advantages of digital image acquisition, processing, archiving, etc. have been described and are being practiced in many SEM, laboratories, they have not been generally applied in microscopy laboratories (TEM, Optical, SEM and others) and impact on increased productivity has not been yet exploited as well.In order to attain our objective we have acquired a SEMICAPS imaging workstation for each of the GE Plastic sites in the United States. We have integrated the workstation with the microscopes and their peripherals as shown in Figure 1.
