The primordial germ cells (PGCs) are the precursors for both the oocytes and spermatogonia. Recently, a novel culture system was established for chicken PGCs, isolated from embryonic blood. The possibility of PGC long-term cultivation issues a new advance in germ cell preservation, biotechnology, and cell biology. We investigated the consequence of gga-miR-302b-5P (5P), gga-miR-302b-3P (3P) and dual inhibition (5P/3P) in two male and two female chicken PGC lines. In treated and control cell cultures, the cell number was calculated every four hours for three days by the XLS Imaging system. Comparing the cell number of control and treated lines on the first day, we found that male lines had a higher proliferation rate independently from the treatments. Compared to the untreated ones, the proliferation rate and the number of apoptotic cells were considerably reduced at gga-miR-302b-5P inhibition in all PGC lines on the third day of the cultivation. The control PGC lines showed a significantly higher proliferation rate than 3P inhibited lines on Day 3 in all PGC lines. Dual inhibition of gga-miR-302b mature miRNAs caused a slight reduction in proliferation rate, but the number of apoptotic cells increased dramatically. The information gathered by examining the factors affecting cell proliferation of PGCs can lead to new data in stem cell biology.

Ayam leher gundul adalah ayam asli/lokal Indonesia yang belum banyak diketahui informasinya dalam halsejarah, potensi, dan karakteristik; sehingga belum dimanfaatkan secara optimal. Ciri yang paling menonjoldari ayam leher gundul adalah tidak mempunyai bulu dibagian lehernya. Untuk mempertahankannya, metode konservasi yang telah dilakukan oleh Balai Penelitian Ternak (Balitnak) adalah dengan memelihara ternak hidup, akan tetapi dengan keterbatasan sarana dan prasarana tidak dapat dilanjutkan. Data performan ayam leher gundul yang dipelihara di Balitnak sudah terkumpul dan ditabulasi. Alternatif konservasi yang dilakukan oleh Balitnak untuk mempertahankan plasma nutfah ayam leher gundul adalah berupa primordial germ cell (PGC) yang dibekukan pada suhu -196 oC yang sewaktu-waktu dibutuhkan dapat ditransfer ke embrio resipien. Sumber PGC yang di koleksi adalah dari darah dan gonad embrio. Berdasarkan potensi yang dimilikinya, ayam leher gundul memiliki performan yang baik, sehingga dapat dimanfaatkan sebagai ayam lokal penghasil daging dan telur. Tulisan ini diharapkan dapat bermanfaat sebagai data pendukung bagi upaya pelestarian dan pengembangan ayam-ayam lokal yang ada di Indonesia, khususnya ayam leher gundul dan umumnya ayam-ayam lokal Indonesia lainnya.

Müllerian inhibiting substance (MIS) is a glycoprotein belonging to the TGF-β superfamily. In mammals, MIS is responsible for the regression of Müllerian ducts in the male fetus. However, the role of MIS in gonadal sex differentiation of teleost fish, which have no Müllerian ducts, has yet to be clarified. In the present study, we examined the expression pattern of mis and mis type 2 receptor (misr2) mRNAs and the function of MIS signaling in early gonadal differentiation in medaka (teleost, Oryzias latipes). In situ hybridization showed that both mis and misr2 mRNAs were expressed in the somatic cells surrounding the germ cells of both sexes during early sex differentiation. Loss-of-function of either MIS or MIS type II receptor (MISRII) in medaka resulted in suppression of germ cell proliferation during sex differentiation. These results were supported by cell proliferation assay using 5-bromo-2′-deoxyuridine labeling analysis. Treatment of tissue fragments containing germ cells with recombinant eel MIS significantly induced germ cell proliferation in both sexes compared with the untreated control. On the other hand, culture of tissue fragments from the MIS- or MISRII-defective embryos inhibited proliferation of germ cells in both sexes. Moreover, treatment with recombinant eel MIS in the MIS-defective embryos dose-dependently increased germ cell number in both sexes, whereas in the MISRII-defective embryos, it did not permit proliferation of germ cells. These results suggest that in medaka, MIS indirectly stimulates germ cell proliferation through MISRII, expressed in the somatic cells immediately after they reach the gonadal primordium.
