Estudou-se, in vitro e in vivo a atividade nematicida dos extratos aquosos de vedélia (Sphagneticola trilobata), erva-de-touro (Tridax procumbens), cravo-de-defunto (Tagetes patula), girassol mexicano (Tithonia diversifolia), botão de ouro (Unxia suffruticosa) e zínia (Zinnia peruviana), sobre Meloidogyne incognita. Os extratos foram preparados na proporção de 1,0g do material seco e triturado para 10mL de água destilada e armazenados por 24h, sendo seguidamente utilizados nos experimentos. Nos testes in vitro, foram depositados 4,0mL do extrato bruto e 2,0mL de uma suspensão aquosa contendo 200 ovos do nematóide em placas de Petri de 5cm de diâmetro e, quinze dias após, procedeu-se a contagem do número de juvenis eclodidos e dos ovos remanescentes para o cálculo das porcentagens de eclosão. Nos testes in vivo, os extratos foram aplicados, separadamente, via pulverização foliar, tratamento de raiz e vertido no solo, semanalmente durante 60 dias. Como testemunha utilizou-se apenas água nos dois experimentos. No ensaio in vitro observou-se que todos os extratos foram eficientes na redução da eclosão de juvenis de M. incognita quando comparados à testemunha, as porcentagens de redução foram 89,96%, 91,13%, 92,48%, 92,72%, 93,2% e 97,48% para erva-de-touro, cravo-de-defunto, girassol mexicano, vedélia, botão de ouro e zínia, respectivamente, e no ensaio in vivo, que os tratamentos não exerceram nenhum efeito sobre o peso do sistema radicular dos tomateiros, no entanto, observou-se que os resultados diferiram entre as espécies utilizadas e a forma de aplicação do extrato na avaliação do peso fresco da parte aérea das plantas. Quanto ao fator de reprodução, observou-se que nenhum dos extratos apresentou diferença estatística em relação à testemunha, no entanto, quando se compara as diferentes formas de aplicação dos mesmos, observa-se que houve diferença estatística quando os extratos de erva de touro e girassol mexicano foram aplicados via pulverização foliar e no tratamento de raiz, contudo, não houve diferença quando estes extratos foram aplicados em forma de rega no solo.

Abstract1,2-dehydropyrrolizidine alkaloids (PAs) represent a class of secondary plant compounds that are active in defence against herbivory. They are present in Chromolaena odorata, one of the most invasive weeds of Asia and Africa. In vitro studies demonstrate that pure PAs from C. odorata roots have nematicidal effects on the root-knot nematode Meloidogyne incognita, even at concentrations of 70-350 ppm. In vivo experiments show that mulch or aqueous crude extracts from C. odorata roots reduce the infection of lettuce by M. incognita. Thus, the use of PA-containing plants appears to be a valuable element for integrated nematode management.

Lysobacter antibioticus HS124 is an antagonistic bacterial strain that was previously isolated from the rhizosphere soil of pepper and showed an enhanced ability to produce lytic enzymes as well as an antibiotic that was identified as 4-hydroxyphenylacetic acid (4-HPAA). In the present study, nematicidal activity of the strain and 4-HPAA against the root-knot nematode, Meloidogyne incognita, causing disease in tomato was investigated in both in vitro and in vivo conditions. For this purpose, adding different concentrations of culture filtrate, crude extract collected from extraction with ethyl acetate and 4-HPAA, in 24-well plates containing ca 500 eggs or 300 second-stage juveniles (J2), significantly decreased the rate of nematode hatch and caused higher mortality of J2 compared with the control treatments. Nematicidal activity of the bacterial strain was further confirmed by conducting pot experiments in which tomato plants were inoculated with M. incognita and the HS124 culture (BC). The control pots were treated with commercial nematicide (CN, 5% Ethoprophos), tap water (TW) or the non-inoculated bacterial culture medium (BCM). In these pot experiments, results demonstrated a strong antagonistic potential of L. antibioticus HS124 against M. incognita where the disease was significantly reduced in the pots treated with BC as compared to TW or BCM. Furthermore, the shoot fresh weight was also increased significantly, which may be attributed to the disease control ability of the strain. Hence, L. antibioticus HS124 may be further developed as a potential biocontrol of root knot nematode in the field.

Summary Avermectins and spinosyns are structurally related natural products of microbial origin and belong to a new family of macrolides which are active against a vast array of invertebrate pests. In the present study, the effects of four members of macrolides; abamectin (ABM), emamectin benzoate (EMB), spinosad (SPI) and spinetoram (SPIT), on Meloidogyne incognita were investigated under in vitro and in vivo conditions. All compounds reduced egg hatching and led to high mortality of the nematode second-stage juveniles (J2). ABM showed the maximum rate of egg hatching inhibition and J2 mortality while SPIT recorded the minimum. All treatments reduced the number of galls, egg masses, eggs/egg mass in roots and J2 in the soil when compared to the control. Based on the 10 folds of the 24 h-LC50 values of J2 mortality in vitro, EMB and ABM exhibited higher percent reduction in galls (79.68 and 71.45%), egg masses (75.19 and 70.54%), eggs/egg mass (60.49 and 40.91%) and J2 in the soil (90.31 and 86.54%), respectively, compared to SPI and SPIT. Significant increase in tomato shoot height occurred in all biopesticides (10 folds) and SPIT (20 folds). SPI at 10 folds of the 24 h-LC50 values of J2 mortality in vitro, significantly increased root length while ABM at 50 folds and SPIT at 20 folds decreased root length by 5.15% and 5.88%, respectively, compared to the untreated inoculated plants. In all treatments, the dry shoot and root weights increased, compared to the untreated control. Our findings suggest that these macrolides have the ability to regulate nematode population densities and may be an alternative to classical nematicides.

Background: Oral infection caused by Candida spp. is a major healthcare problem in dental and oral care. Treatment failure has been reported in cases of oral candidiasis as a result of resistance to common antifungals. Aim and Objective: In this study, the in vitro and in vivo activities of extract of Tithonia diversifolia against virulence factor-borne and antifungal resistant-Candida albicans were investigated. Candida albicans was isolated from the saliva of patients attending a tertiary hospital in Ekiti State. Methodology: Standard methods were used to determine the presence of virulence factors in the isolates. In vitro and in vivo anti-candidal activities of the hydro-ethanolic extract of T. diversifolia were also tested on the test fungus. Results: The virulence factors have varying percentage of occurrence in all the isolates with catalase having the highest. Itraconazole and nystatin were not effective against the isolates. Out of the six isolates selected (based on antifungal resistance) only three produced strong biofilm. The reduction in the population of the test organisms by the extract was time and concentration dependent. At the end of candidal challenge and treatment assays, extract of T. diversifolia has lower anti-candidal property compared to nystatin. Conclusion: This study has shown that C. albicans associated with the mouth carries virulence factors and are resistant to common antifungals. In this work, we noticed antifungal effects of hydro-alcoholic extract of T. diversifolia on C. albicans associated with oral infections.

O objetivo deste trabalho foi avaliar o controle de Meloidogyne incognita e a ativação de mecanismos de defesa de plantas em função de diferentes métodos de aplicação de biofertilizante de esterco bovino (BIO). Para isto foi conduzido um experimento para avaliar a porcentagem de eclosão (PE) do nematoide com dois tratamentos, água e BIO, com dez repetições. Um segundo experimento foi conduzido em casa de vegetação em esquema fatorial 2 x 3 + 1 com seis repetições. O fator A (água e BIO) e fator B (aplicação via solo (S), pulverizado (PUL) e a combinação S + PUL), além da testemunha (sem inoculação do patógeno) como tratamento adicional. Foram avaliados o número de ovos (NO) e juvenis (NJ) de M. incognita na raiz, atividade específica de catalase (CAT), peroxidase (POX) e polifenoloxidase (PFO) no sistema radicular e parte aérea da cultura. A PE de M. incognita foi 65% inferior em solução com BIO, in vitro. In vivo, a aplicação de BIO reduziu o NO e NJ. Ao aplicar o BIO via S, observou-se incremento de até 1,8 vezes da CAT e 35 vezes da POX em folhas. A POX foi incrementada em até 1,7 vezes no sistema radicular. O biofertilizante bovino apresentou potencial no controle M. incognita.

Phytobiocides are a good alternative to chemicals in managing bacterial diseases including bacterial wilt of tomato caused by Ralstonia solanacearum. In the present research study, finely ground dried powders of seven widely available medicinal plants/weeds species viz., Peganum harmala (esfand or wild rue), Calotropis procera (sodom apple), Melia azedarach (white cedar), Allium sativum (garlic), Adhatoda vasica (malabar nut), Tagetes patula (marigold) and Nerium oleander (oleander) were assessed for their anti-microbial activity, both in-vitro (10% w/v) and in-vivo (10, 20, 30, and 40 g/kg of potted soil) against R. solanacearum. Aqueous extracts (prepared as 10% w/v, soaking for 48-72 h and filtering) of C. procera, A. vasica, and T. patula inhibited the in-vitro growth of the bacterial pathogen over 60% of that produced by the standard antibiotic streptomycin. A. sativum, N. oleander and P. harmala aqueous extracts were less effective while M. azedarach showed no effect against R. solanacearum. The higher dose (40 g/kg of soil) of C. procera, A. vasica and T. patula decreased disease severity quite effectively and increased yield and plant growth characters as much as the standard antibiotic did. No phytotoxicity of medicinal plants powder was observed on tomato plants. Alkaloids, flavonoids, tannins, saponins and terpenoids were detected in the aqueous extracts of T. patula and A. vasica whereas C. procera was found to have only alkaloids, flavonoids, tannins and saponins. Our data suggest that dried powders of T. patula, C. procera and A. vasica (40 g/kg of soil) could be used as an effective component in the integrated disease management programs against bacterial wilt of tomato.

Plant root-knot nematode disease is a great agricultural problem and commercially available nematicides have the disadvantages of high toxicity and limited usage; thus, it is urgent to develop new nematicides derived from nature substances. In this study, a novel fluorinated derivative was synthesized by modifying chitosan oligosaccharide (COS) using the strategy of multiple functions. The derivatives were characterized by FTIR, NMR, elemental analysis, and TG/DTG. The activity assays show that the derivatives can effectively kill the second instar larvae of Meloidogyne incognita in vitro, among them, chitosan-thiadiazole-trifluorobutene (COSSZFB) perform high eggs hatching inhibitory activity. The derivatives can regulate plant growth (photosynthetic pigment), improve immunity (chitinase and β-1,3-glucanase), and show low cytotoxicity and phytotoxicity. According to the multi-functional activity, the derivatives exhibit a good control effect on plant root-knot nematode disease in vivo. The results demonstrate that the COS derivatives (especially fluorinated derivative) perform multiple activities and show the potential to be further evaluated as nematicides.
