ISG15 is induced by conceptus-derived interferon-τ in the endometrium on days 15–45 of pregnancy. It was hypothesized that pregnancy induces blood cell ISG15 gene expression and that low blood ISG15 mRNA levels provide an indication of non-pregnant cows on day 18. Blood was collected either on day 18 (n = 78) or on days 15–21, 25, and 32 (n = 21; serial collection) from dairy cows following artificial insemination (AI). Plasma progesterone concentration was determined using RIA. ISG15 mRNA levels were determined using real-time PCR. Pregnancy was diagnosed on day 32 using transrectal ultrasound. ISG15 mRNA levels increased after day 16, peaked at day 20 and then declined to day 16 levels by 32 days following AI. The average pregnancy rate was 43% based on blood cell ISG15 mRNA. The average pregnancy rate was 33% based on the transrectal ultrasound. Lower levels of ISG15 mRNA or progesterone during serial collections were 100% accurate in predicting non-pregnant cows based on day 32 transrectal ultrasound. However, detection of ISG15 mRNA yielded 78% accuracy in predicting pregnant cows, while progesterone yielded 58% accuracy. Average plasma progesterone based on pregnancy status according to ultrasound was consistently higher in pregnant (> 4 ng/ml) when compared with non-pregnant cows from days 15 to 32, except on day 16. It is concluded that detection of low blood ISG15 mRNA levels during serial collection from days 17 to 25 serves as an accurate indicator of cows that are not pregnant, thus allowing re-synchronization and insemination.

The study aimed (1) to verify if artificial insemination (AI), performed in the morning, evening or nighttimehas influence on the pregnancy rate on high production dairy cows, and (2) to determine the repeat breedingrate after the AI at intervals of 01-21, 22-43 and 44-65 days. We analyzed 1980 AI data from HolsteinFriesian cows from years 2004 to 2008. The average milk production on this period was 39 kg/milk/day/cow (305 days of lactation). The AI was performed 12 hours after the first detection of estrus. For the dataanalysis one AI was computed. The AI was done based on conventional method. The incidence of the AI in themorning, evening and night was 29.8; 50.4 and 19.8% respectively; and the pregnancy rate 45.7; 44.9 and48.4%. The frequency of the repeat breeding rate at the interval of 1-21 days on morning, evening and nightafter failed AI was 28.0; 22.3 and 19.8% respectively; at the interval of 22-43 days was 52.0; 52.2 and 55.2%,respectively and at interval of 44-65 days was respectively 20.0; 25.5 and 25.0%. Comparing the three timesof the day each other, the cows showed greater repeat breeding on 22-43 days (P<0.05) after the AI. It wasconcluded that times of the day has no influence on the on pregnancy rate; the highest repeat breeding ratetook place between 22 and 43 days after AI, resulting significant delaying of the open period.

ABSTRACT: The aim of this study was to reduce the waiting voluntary period in dairy cows by administration of two different hormonal protocols for estrus synchronization and fixed-time artificial insemination (FTAI). One hundred twenty-eight Holstein dairy cows were studied. Three groups (G) were carried out: G1 (44 treated), G2 (34 treated) and G3 (50 as control). In the treated cows the following hormonal protocols was applied between 40 and 45 days post partum: G1- Day 0 (D0) - bovine intravaginal device containing progesterone (P4) plus 1.0 mg of estradiol benzoate (EB), D8 - removal of the P4, plus 0.5 mg of cloprostenol plus 400 IU of equine chorionic gonadotropin (eCG), D9 - 1.0 mg of EB and FTAI after 36 hours. G2 - D0 - P4 intravaginal plus 50 mcg of gonadotropin-releasing hormone (GnRH), D7 - removal of the P4 plus cloprostenol plus 400 IU eCG; D9 - 50 mcg of GnRH and FTAI 16 hours after. G3 – Control group inseminated 12 hours after the visual estrus. Results: G1 and G2 were inseminated (FTAI) respectively in the D55.5 and D56.8 after calving, while the G3 in the D84.5 (p<0.01).  The pregnancy rate in the G1, G2 and G3 was respectively 47.0, 46.1 and 35.8% (p>0.05).  In Conclusion: there was significant reduction in the voluntary waiting period in treated cows and higher pregnancy rate although not significant.

This study compared pregnancy rates following 2 timed artificial insemination (TAI) protocols using PGF2α + oestradiol or PGF2α + oestradiol + GnRH in dairy cows. Four hundred fifty-five Holstein dairy cows, 2.2 ± 0.1 in parity, 148.3 ± 3.6 days in milk, with corpus luteum of greater than 20 mm diameter confirmed by ultrasonography (Tringa Linear with 5.0 MHz array transducer; Esaote Pie Medical, Maastricht, the Netherlands) were randomly divided into two treatments: an injection of 500 μg of cloprostenol, PGF2α analogue (Iliren cycle BP®, Intervet International GmbH, Unterschleissheim, Germany) and an injection of 2 mg of oestradiol benzoate (EB, SY Esrone, Samyang, Seoul, Korea) 36 h later (PGF2α + EB group, n = 257), or an injection of 500 μg of cloprostenol, an injection of 2 mg of EB 36 h later and an additional injection of 100 μg of gonadorelin, GnRH analogue (Godorel, Uni-Biotech Co., Ltd., Korea) 24 h later (PGF2α + EB + GnRH group, n = 198). All cows in the two groups received TAI 24 h after the EB injection. Pregnancy was determined at 40 to 50 days after TAI by using both ultrasonography and rectal palpation. Pregnancy rates between the PGF2α + EB and PGF2α + EB + GnRH groups were compared by the chi-square test using the SAS program (version 9.1: SAS Institute Inc., Cary, NC, USA). Pregnancy rate following TAI did not differ between the PGF2α + EB (32.7%) and PGF2α + EB + GnRH groups (34.9%; P > 0.05). In conclusion, addition of GnRH for TAI protocol using PGF2α + oestradiol did not improve pregnancy rate in dairy cows.

In modern dairy cows, poor fertility is an ever-increasing problem. Milk progesterone analysis has revealed that this poor fertility is associated with a high incidence of reproductive cycle problems during the post partum period and with reduced progesterone secretion following mating. One of the likely causes of these problems is the increasing metabolic demand placed on these cows by increasing milk yield. In recent years, the search for an endocrine link between nutritional status and reproductive function has focussed on leptin. The aim of the present study was to determine whether plasma concentrations of leptin differ between cows with and without post partum reproductive problems and to determine if any relationship exists between plasma leptin and post mating plasma progesterone concentration.
