Cherry yellow virus, isolates Y.2 and Y.4, and necrotic ring spot virus, isolates N.4 and N.5, purified either from cucumber leaves or from sour cherry petals, were characterized by antigenically related particles that sedimented at approximately 72 S. Some preparations of each virus derived from either host also contained either a 35 S or a 22 S component usually having a low nucleic acid content. Such preparations were shown by the agar gel double-diffusion technique to contain two antigens, V and Q, that were only distantly, if at all, related. The 72 S component is associated with the V antigen, whereas the 22 S and 35 S components are tentatively considered to be two forms of the Q antigen. The Q antigen could also be detected in clarified expressed sap of infected cucumber tissue, but not in that of healthy cucumber nor in extracts prepared from healthy sources by methods used to purify the viruses. The Q antigen is thus associated with infection, but its origin has not yet been satisfactorily determined.

Purified preparations of several isolates of tomato ring-spot virus were shown by rate-zonal centrifugation in sucrose and equilibrium centrifugation in CsCl to be composed of two individual nucleoprotein components. Acrylamide-gel electrophoresis showed that the lighter (middle) component contained a nucleic acid (RNA 2) that was distinct from the species (RNA 1) contained in the heavier (bottom) component. The bottom was more infectious than the middle component and infectivity was enhanced by mixing the components, indicating that the virus genome is divided between component types. Similar results were obtained from infectivity tests on the two nucleic acids. The nucleic acid contents of the middle and bottom components were about 40 and 41%, respectively. The average molecular weights of RNA 2 and RNA 1 from three virus isolates, as determined by acrylamide-gel electrophoresis, were 2.5 and 2.6 × 106, respectively. Molecular complexing between the RNA species during electrophoresis was prevented with the use of formamide. The single protein subunit from the same three isolates had an average molecular weight of about 58 000. Serological comparisons of five tomato ring-spot isolates associated with diseases of fruit trees and grapevines indicated that only the grape yellow vein strain was antigenically distinct. These and other properties indicate that this virus is similar to other members of the nepovirus group.

Faba bean necrotic yellow virus (FBNYV) was not detected during 1994 to 1996 field surveys of faba beans (Vicia faba L.) in Spain (1). In 1997, however, one sample with symptoms of necrosis, collected in Baleares, was tested using ELISA (enzyme-linked immunosorbent assay) and was positive for both Tomato spotted wilt virus (TSWV) and FBNYV. FBNYV is a single-strand DNA virus that is transmitted by aphids and is the main virus disease of broad bean in North Africa and West Asia (2). During 1997 to 1999, faba bean plants with symptoms of necrosis, yellowing, small leaves, and stunting were collected from several fields in the Murcia Region (Spain) and were analyzed using ELISA. To detect FBNYV, we used monoclonal 2E9 supplied by H. J. Vetten (Institute of Plant Virology, Microbiology and Biosafety, BBA, Braunschweig, Germany). Of 700 samples analyzed, 34 were positive for FBNYV. Of the 34 positive samples, 12 tested positive, using commercial antiserum from Loewe, Inc. (Munich) for mixed infections with TSWV. FBNYV was transmitted to healthy faba bean plants by aphids (Acyrthosiphon pisum) in greenhouse experiments and was confirmed by ELISA. Preliminary epidemiological data showed a gradual increase in the number of plants infected with time in the same field. Aphid transmission of FBNYV to faba beans has established the disease in Spain and is a potential threat to other leguminous crops. This is the first report of a nanovirus in Europe.
