<p>Kaffir lime (Citrus hystrix) is a native plant of Indonesia. Nowadays the use of plants is limited as ingredients in Indonesia cuisine and aromatherapy oils for industry. Leaves of C. hystrix contain of polyphenols and essential oils thus they were possibly cytotoxic to cancer cell line. The objective of this research was to determine the cytotoxicity effect and apoptosis induced by leaves extract C. hystrix to cervical cancer cell line (HeLa cells). Methods used in this study included sampling of kaffir lime leaves, extractionusing three different solvents (ethanol, ethyl acetate, and hexane), detection of metabolite secondary compound (alkaloids, flavonoids, terpenoids, saponins, and tannins) using thin layer chromatography (TLC), cytotoxicity assay via MTT assay, and apoptosis test with double staining method (ethidium bromide-acrydine orange). Result showed kaffir lime crude extract dose dependently inhibitHeLa cells proliferation. lC50of ethanolicand ethyl acetate extract was 82,034 µg/mL and 57,845 µg/mL, respectively means cytotoxic to HeLa cells. On the other hand lC50 of hexane extract was 203,992 µg/mL which was not cytotoxic. Furthermore ethanolic and ethyl acetate extract were able to induce apoptosis of HeLa cells by increasing the number of apoptotic cells. In conclusion, kaffir lime leaves extract had cytotoxic effect and induced apoptosis. Moreover ethyl acetate extractof kaffir lime was the most potential to induce apoptosis in HeLa cells. </p><p><strong>Keywords</strong>: kaffir lime leaves (Citrus hystrix), HeLa, MTT, cytotoxicity, apoptosis.</p>

Chaihu-shugan-san, as a traditional Chinese herbal formula, is composed of seven different herbs. This medicine can treat cancer due to its antioxidant compounds. In this study, the effect of Chaihu-shugan-san was considered on cytotoxicity induction and PDGF gene expression in cervical cancer cell line HeLa at different concentrations and at different times, by the MTT method. Paclitaxel + cisplatin were used as a control in this study. The expression of the PDGF gene was quantitatively evaluated in treated cells by real-time PCR, and a generalized linear model was used to evaluate the effect of the medicine, and Duncan's multiple range tests were used to evaluate the data. The results of the MTT test showed that Chaihu-shugan-san had antitumor properties in different concentrations, but there was a significant difference between this medicine and paclitaxel +cisplatin. Also, examination of gene expression showed that this medicine reduced the expression of the PDGF gene in the HeLa cancer cell line (P ? 0.04). Therefore, Chaihu-shugan-san could be suggested as an effective factor in preventing the growth of cervical cancer cells and controlling angiogenic factors that play an important role in the metastasis of cancerous tumors.

Alstonia scholaris, a tropical evergreen tree belonging to the family Apocynaceae has traditionally been referred to have anti-cancer activity, but it has not been explored so far. Therefore, the study is aimed to investigate the cytotoxic effect of aqueous and ethanolic bark extracts of Alstonia scholaris against cervical cancer cell line (HeLa Cell line). The bark part of Alstonia scholaris was collected, dried, powdered and made to a coarse powder. Maceration was carried out for the bark of Alstonia Scholaris using two different solvents like ethanol and aqueous solutions. Both the extracts were collected, evaporated to dryness. The extracts were then investigated for the phytochemical screening and cytotoxic effect by MTT Assay using cervical cancer cell line (HeLa cell line). The phytochemical screening of both the extracts showed the presence of various phytoconstituents such as alkaloids, flavonoids, glycosides, gums and mucilage. The IC50 value of Alstonia scholaris of aqueous extract was found to be 13.38µg/ml, whereas the IC50 value of ethanol extract was found to be 14.21µg/ml. According to the results obtained, it has been proved that the aqueous and ethanolic extracts of Alstonia Scholaris bark showed the presence of significant phytoconstituents and also showed significant cytotoxic effect (MTT Assay) against cervical cancer.
