ABSTRACT Aromatic rice (Oryza sativa L.) is one type of rice that has aroma and high selling price than non-aromatic rice. The variety development is needed to form superior aromatic rice. The variety development requires information about kinship and diversity between varieties. Kinship analysis with identification of morphological characteristics is an observation based on taxonomic approach. This research aims to determine the morphological characteristics and kinship of 21 aromatic rice varieties. The research was conducted from March to September 2018 with a randomized block design (RBD) of 3 replications. Observation variables used plant height (cm), number of productive tillers (tiller), age of flowering and harvesting (days), weight of 100 grain (gram), shape of grain, color of grain, color of rice and DNA band pattern as molecular data. Morphological data were analyzed by variance (ANOVA) and cluster analysis with UPGMA program (Unweight Pair Group Method with Arithmetic Mean) using DIST Coefficient on NTSYS 2.02 software. Molecular data showed in visualisation of DNA band using Primer OPB-07 based on RAPD marker. The research results showed the differences in morphological characters and kinship levels. Cluster analysis forms 2 clusters and 2 sub-clusters. Keywords: Aromatic Rice, Mophological Diversity, RAPD, Dendrogram ABSTRAK Padi aromatik (Oryza sativa L.) merupakan salah satu jenis padi yang memiliki aroma wangi dan memiliki harga jual yang lebih tinggi dibanding padi biasa. Pengembangan varietas diperlukan untuk membentuk padi aromatik yang lebih unggul. Pengembangan varietas membutuhkan informasi tentang kekerabatan dan keanekaragaman antar varietas. Analisis kekerabatan dengan identifikasi karakteristik morfologi merupakan suatu observasi berdasarkan pendekatan taksonomi. Percobaan ini bertujuan untuk mengetahui karakteristik morfologi dan molekuler 21 varietas padi aromatik. Percobaan dilakukan di mulai Maret - September 2018 dengan Rancangan Acak Kelompok (RAK) 3 ulangan. Variabel pengamatan menggunakan tinggi tanaman (cm), jumlah anakan produktif (batang), umur berbunga dan panen (HSS), bobot 100 gabah, bentuk gabah, warna gabah, dan warna beras serta pola pita DNA sebagai data molekuler. Data morfologi dianalisis dengan ragam (ANOVA) serta dianalisis gerombol dengan program UPGMA (Unweight Pair Group Method with Arithmetic Mean) menggunakan DIST Koefisien pada software NTSYS 2.02. Data molekuler ditunjukkan dalam bentuk visualisasi DNA menggunakan primer OPB-07 berdasarkan penanda RAPD. Hasil percobaan menunjukkan perbedaan karakter morfologi dan tingkat kekerabatan yang berbeda. Analisis gerombol membentuk 2 cluster dan 2 sub-cluster. The availability of molecular marker systems allowed estimating the relationships among various taxa. This study was aimed at assessing the genetic diversity among ten aromatic rice (Oryza sativa L.) pools from Bangladesh by means of randomly amplified polymorphic DNA (RAPD) markers. These varieties were evaluated for polymorphisms after amplification with 10 decamer primers. A total of 60 RAPD fragments were generated among the assessed varieties with a polymorphism percentage of 80. Cluster analysis by the unweighted pair group method of arithmetic means (UPGMA) showed that these 10 varieties could be placed into two groups with a similarity ranging from 65 to 86% depicting adjacent association between Rajbhog and Kalijira‐12, whereas Maloti belongs to a separate group maintaining maximum distance from rest of the varieties. The analysis revealed that the intervarietal genetic relationship of several varieties is related to their center of origin. As expected, most of the varieties have a narrow genetic base. The present results could be used for the selection of possible parents to generate a mapping population and utilized by the breeders for assessing the genetic diversity of rice genotypes.Plant Tissue Cult. & Biotech. 27(2): 217-225, 2017 (December)

Random amplified polymorphic DNA fingerprinting was performed to assess the genetic diversity among rarely cultivated traditional indica rice (Oryza sativa L.) varieties collected from a tribal hamlet of Kerala State, India. A total of 664 DNA bands amplified by 15 primers exhibited 72.9% polymorphism (an average of 32.3 polymorphic bands per primer). The varieties Jeerakasala and Kalladiyaran exhibited the highest percent (50.19%) polymorphism, while Thondi and Adukkan showed the lowest (9.85%). Adukkan (78 bands) and Jeerakasala (56 bands) yielded the highest and the lowest number of amplicons, respectively. Unweighted Pair Group Method with Arithmetic mean analysis using the Dice similarity coefficient showed the highest value of similarity coefficient between the varieties Adukkan and Thondi, both shared higher level of similarity (0.81), followed by Kanali and Thondi (0.88). Of the three subclusters, the varieties of Adukkan, Thondi, Kanali, Mannuveliyan, Thonnuranthondi, and Chennellu grouped together with a similarity of 0.77. The second group represented by Navara, Gandhakasala, and Jeerakasala with a similarity coefficient of 0.76 formed a cohesive group. The variety Kalladiyaran formed an isolated position that joined the second cluster. The Principal Coordinate Analysis also showed separation of Kalladiyaran from the other varieties.

Genetic polymorphism and relationships among 30 commercial varieties of Bangladeshi aromatic rice (Oryza sativa L.) were established using random amplified polymorphic DNA (RAPD) primers. Out of fifty 10-mer RAPD primers screened initially, four were chosen and used in a comparative analysis of different varieties of indigenous Bangladeshi aromatic rice. Of the 33 total RAPD fragments amplified, 7 (21.21%) were found to be shared by individuals of all eight varieties. The remaining 26 fragments were found to be polymorphic (78.79%). Pair-wise estimates of similarity ranged from 0.101 to 0.911. Highest genetic diversity was determined between Radhunipagol and Dubsail varieties (0.911). The amount of genetic diversity within aromatic rice germplasm was quite high as determined by the genetic similarity coefficients between varieties. Genetic similarities obtained from RAPD data were also used to create a cluster diagram. Cluster analysis using an un-weighted pair-group method with arithmetic averages (UPGMA) was used to group the varieties and the 30 aromatic rice varieties were grouped into 6 clusters where cluster I includes the maximum number of varieties (9). Cluster VI includes minimum number of varieties (2). This Study offered a rapid and reliable method for the estimation of variability between different varieties which could be utilized by the breeders for further improvement of the local aromatic rice varieties.

Objetivou-se discernir populações caprinas de cinco ilhas da República de Cabo Verde (n=533) por meio de dados biométricos e análises estatísticas. Foram avaliadas 16 características de fêmeas adultas, através da estatística descritiva simples, análise de variância, teste de multicolinearidade, distância generalizada de Mahalanobis (D²) e algoritmo UPGMA (Unweighted Pair Group Method Arithmetic Mean). Após o teste de multicolinearidade foi identificada e descartada a variável profundidade torácica. As D² foram calculadas com base nas 15 medidas biométricas. O maior valor da D² foi entre as populações das ilhas do Fogo e São Nicolau (22,73), e a menor D² foi entre Santo Antão e São Vicente (3,71). O dendrograma a partir de 15 variáveis em cinco populações colocou as cabras da ilha de Fogo em ramo a parte das demais. Agruparam-se num ramo as cabras das ilhas de Santo Antão e São Vicente. Este resultado está de acordo com a distância geográfica entre as ilhas de Cabo Verde e o histórico recente de intercâmbio de animais entre elas.
