Summary The queen and worker bees have the same genetic makeup. However, the queen differs dramatically from the workers in anatomy, physiology, behavior, and lifespan. Three genes (sir2, ash2, and hdac1) have been shown to be associated with histone methylation and acetylation as well as longevity in worms and flies. The relative expression level of these genes was examined in the heads of queens and workers at different developmental stages. The sir2, ash2, and hdac1 expression levels in newly emerged queens, egg-laying queens, and egg-laying workers were significantly higher than those in newly emerged workers, nurses, and foragers. We conclude that these genes are possibly “queen-like” genes.

AbstractQueenless, caged, newly emerged worker bees (Apis mellifera L.) were fed honey, 22 and 40% pollen in honey, and 22 and 40% royal jelly in honey for 14 days. Workers fed royal jelly, pollen, and honey had large, medium, and small ovaries, respectively. Royal jelly had higher nutritive value for workers’ ovarian development than did pollen, possibly because royal jelly is predigested by nurse bees and easily used by adult and larval bees. These results suggest that nurse bees could mediate workers’ ovarian development in colonies via trophallactic exchange of royal jelly. Six levels of royal jelly in honey, 0, 20, 40, 60, 80, and 100% (royal jelly without honey), were tested for their effects on workers’ ovarian development and mortality for 10 days. High levels of royal jelly increased ovarian development, but also increased worker mortality. All caged bees treated with 100% royal jelly died within 3 days. When workers were incubated at 20, 27, and 34 °C for 10 days, only bees at 34 °C developed ovaries. These findings suggest that nurse bees functioning as units which digest pollen and produce royal jelly may feed some potentially egg-laying workers in a brood chamber with royal jelly when a queen is lost in a colony. Feeding workers a diet of 50% royal jelly in honey and incubating at 34 °C for 10 days is recommended for tests of ovarian development.

Abstract The recent alarming loss of honey bee colonies around the world is believed to be related to the presence of viruses. The aim of this study was to detect two major viral diseases, Apis mellifera Filamentous virus (AmFV) and Israeli Acute Paralysis Virus (IAPV) using Reverse Transcription - Polymerase Chain Reaction RT-PCR, in honey bees in Mexico. Adult and larvae honey bee samples were collected from asymptomatic colonies of six major beekeeping regions in the state of Chihuahua, Mexico. Both viruses were detected in both developmental stages of honey bees, IAPV at a higher prevalence (23.5%) as compared to AmFV, only in 0.9% of samples. However, this is the first report on AmFV infection in Mexican apiaries. Further studies are required to understand the AmFV and IAPV impact on colony loss in Mexico and to develop strategies for enhancing the control of viral diseases.

AbstractWe examined the factors that might influence ovary development in worker honey bees, Apis mellifera L. Queenless workers at different ages (≤ 12 h, and 4, 8, and 21 d) were tested in cages for ovarian development. Newly emerged, 4- and 8-d-old, and 21-d-old workers had medium-, large-, and small-sized ovaries, respectively, suggesting that of the worker ages tested only 4- and 8-d-old workers are likely to become egg layers in a queenless colony. Also, we compared ovarian development of newly emerged workers that were caged for 14 d and allowed to consume either pollen or royal jelly to that of another group of workers similarly caged but screened so that they could only obtain food via trophallaxis from young bees. Ovaries of newly emerged workers that received food from young bees were as well developed as those of newly emerged workers allowed to take pollen or royal jelly directly. Screened workers also had lower but still elevated vitellogenin levels compared with bees having direct access to food. These results indicate that nurse-age bees functioning as pollen-digesting units affect the ovarian development of other workers and to a lesser extent vitellogenesis via food exchange. We compared the influence of group sizes of 25, 125, and 600 bees per cage on ovarian development for 14 d. The two groups of 25 and 125 bees had similar mean ovary scores, and higher scores than a group of 600 bees. Our findings suggest that nurse-age bees could play an important role in mediating worker fertility via trophallaxis, possibly by differentiating worker dominance status, and generally only young workers become fertile when a queen is lost in a colony. Vitellogenin is a more sensitive parameter to measure bee fertility, and might be a useful tool to further explore ovary development and egg laying in worker social insects. We recommend measuring haemolymph vitellogenin titres and (or) oocyte length of workers in a group of 25 bees per cage, supplied with 50% royal jelly in honey as a standard method to assess honey bee worker fertility in future experiments.

The vitellogenin receptor (VgR) belongs to the low-density lipoprotein receptor (LDLR) family, responsible for mediating the endocytosis of vitellogenin (Vg) into the ovaries to promote ovarian growth and oviposition. Here, we cloned and measured VgR gene expression characteristics in the bumblebee Bombus lantschouensis. RNA interference was used to validate VgR function. The results showed that the full length of the BLVgR cDNA was 5519 bp, which included a 5280 bp open reading frame encoding 1759 amino acids (AAs). Sequence alignment revealed that the protein contained 12 LDLa, 5 EGF, 2 EGF-CA and 10 LY domains. Phylogenetic analysis showed that BLVgR and the VgR of Bombus terrestris clustered together and the tree of bumblebees (Bombus) appeared as one clade next to honeybees (Apis). Transcript expression analysis showed that BLVgR was expressed in all tested tissues and showed the highest abundance in the ovaries. BLVgR expression was present in all developmental stages. However, the expression level in larvae was extremely low. In addition, the expression of BLVgR was significantly upregulated after egg laying in both workers and queens. In new emerging workers injected with 5 µg of VgR dsRNA, the expression level of BLVgR was 4-fold lower than that in the GFP dsRNA-injected group after 72 h. Furthermore, BLVgR silencing significantly reduced the number of eggs laid (3.67 ± 1.96 eggs) and delayed the first egg-laying time (16.31 ± 2.07 days) in worker microcolonies when compared to dsGFP (37.31 ± 4.09 eggs, 8.15 ± 0.22 days) and DEPC-treated water injected controls (16.42 ± 2.24 eggs, 10.00 ± 0.37 days). In conclusion, the BLVgR gene and its reproductive function were explored in the bumblebee B. lantschouensis. This gene plays an important role in egg laying time and egg number.

We compared nutrient compositions of honey bee (Apis mellifera) drones of different developmental stages from two different populations—the Italian honey bee reared in Korea and Buckfast bees from Denmark. Analyses included amino acid, fatty acid, and mineral content as well as evaluations of antioxidant properties and haemolysis activities. The compositions of total amino acids, and thus protein content of the insects, increased with development. A similar trend was observed for minerals presumably due to the consumption of food in the adult stage. In contrast, total fatty acid amounts decreased with development. Altogether, seventeen amino acids, including all the essential ones, except tryptophan, were determined. Saturated fatty acids dominated over monounsaturated fatty acids in the pupae, but the reverse held true for the adults. Drones were found to be rich in minerals and the particularly high iron as well as K/Na ratio was indicative of the nutritional value of these insects. Among the three developmental stages, adult Buckfast drones exhibited the highest antioxidant activity. Bearing in mind the overall high nutritional value, i.e., high amino acids, minerals and less fatty acids, late pupae and adult drones can be useful for human consumption while the larvae or early pupal stage can be recommended as feed. However, owing to their relatively high haemolysis activity, we advocate processing prior to the consumption of these insects.

As social insects, honey bees (Apis mellifera) rely on the coordinated performance of various behaviors to ensure that the needs of the colony are met. One of the most critical of these behaviors is the feeding and care of egg laying honey bee queens by non-fecund female worker attendants. These behaviors are crucial to honey bee reproduction and are known to be elicited by the queen’s pheromone blend. The degree to which workers respond to this blend can vary depending on their physiological status, but little is known regarding the impacts of developmental exposure to agrochemicals on this behavior. This work investigated how exposing workers during larval development to chronic sublethal doses of insect growth disruptors affected their development time, weight, longevity, and queen pheromone responsiveness as adult worker honey bees. Exposure to the juvenile hormone analog pyriproxyfen consistently shortened the duration of pupation, and pyriproxyfen and diflubenzuron inconsistently reduced the survivorship of adult bees. Finally, pyriproxyfen and methoxyfenozide treated bees were found to be less responsive to queen pheromone relative to other treatment groups. Here, we describe these results and discuss their possible physiological underpinnings as well as their potential impacts on honey bee reproduction and colony performance.
