Given the advances in science and technology, rapid development of socialist market economy and continuous advance of urbanization, it is necessary to enlarge the scale of engineering construction. As the form of engineering structure becomes more complex, large-scale and high-level projects with deep foundation have appeared in engineering construction. For construction engineering, one of its technologies includes solving the difficulties in construction. It is required to deal with the safety risk of construction in time to guarantee safety construction, timely solve the management difficulties and contradictory problems of the project and ensure both the safety of engineering construction and the rationalization of the institution setting of the safety supervision on the project.

Next-Generation Sequencing (NGS) technologies, by reducing the cost and increasing the throughput of sequencing, have opened doors to generate genomic data in a range of previously poorly studied species. In this study, we propose a method for the rapid development of a large-scale molecular resources for orphan species. We studied as an example the true lavender (Lavandula angustifolia Mill.), a perennial sub-shrub plant native from the Mediterranean region and whose essential oil have numerous applications in cosmetics, pharmaceuticals, and alternative medicines. The heterozygous clone “Maillette” was used as a reference for DNA and RNA sequencing. We first built a reference Unigene, compound of coding sequences, thanks to de novo RNA-seq assembly. Then, we reconstructed the complete genes sequences (with introns and exons) using an Unigene-guided DNA-seq assembly approach. This aimed to maximize the possibilities of finding polymorphism between genetically close individuals despite the lack of a reference genome. Finally, we used these resources for SNP mining within a collection of 16 commercial lavender clones and tested the SNP within the scope of a genetic distance analysis. We obtained a cleaned reference of 8, 030 functionally in silico annotated genes. We found 359K polymorphic sites and observed a high SNP frequency (mean of 1 SNP per 90 bp) and a high level of heterozygosity (more than 60% of heterozygous SNP per genotype). On overall, we found similar genetic distances between pairs of clones, which is probably related to the out-crossing nature of the species and the restricted area of cultivation. The proposed method is transferable to other orphan species, requires little bioinformatics resources and can be realized within a year. This is also the first reported large-scale SNP development on Lavandula angustifolia. All the genomics resources developed herein are publicly available and provide a rich pool of molecular resources to explore and exploit lavender genetic diversity in breeding programs.

UAV assisted project progress, management quality and safety and other aspects, so that managers can timely and efficiently understand the situation of the project site, and effectively improve the management's control over the project construction, indicating the future application prospect of UAV technology in the construction field.

We found similar genetic distances between pairs of clones, related to the out-crossing nature of the species, the restricted area of cultivation and the clonal propagation of the varieties.The method propose is transferable to other orphan species, requires little bioinformatics resources and can be realized within a year. This is the first reported large-scale SNP development on Lavandula angustifolia. All this data provides a rich pool of molecular resource to explore and exploit biodiversity in breeding programs.

AbstractThis paper presents the results of public opinion research conducted in Japan on the willingness of persons to contribute their DNA to a genetic database, and the reasons behind their choices. The results are discussed in light of the Act Concerning Protection of Personal Information issued in 2003, and with reference to genetic database projects that have been conducted in Japan. There appears to be increasing concerns about privacy in Japan, despite a high level of support for science and technology. While 44% said they would agree to blood donation for a DNA bank, 28% were negative to donate, and another 28% answered they “don't know”. The reasons behind these choices and the concepts of storage of genetic databases are partially reflected in the guidelines applied to collection of genetic databases, but greater and more independent community engagement will be important to increase public involvement in large scale genetics projects in Japan.

However, there are also many companies whose own reputation and actual benefits have been affected for poor engineering quality caused by poor construction management. This is exactly what we must pay attention to. This article will analyse and explore the relevant work to improve the engineering quality of different phases of engineering construction and to guide the management of the construction unit in the future from the perspective of the construction unit to strengthen the project quality supervision.

Abstract Safety risk during the construction of a Large Scale Engineering (LSE) project can be avoided through safety education, following a correct procedure and using an engineering method. In order to ensure that LSE works in a safe state, some parameters need to be restricted to a certain range. Besides, in order to apply the former control for the predictive values, a Model Predictive Control (MPC) theory is suggested to be used for safety risk controlling. We share a MPC work flow and detail calculation steps of the rolling optimization, feedback correlation and constraint control, and a mean time gradient decent method is used during the optimization calculation iteration steps. At the end, regarding the resource allocation issue in the ship-lift engineering construction area of “Three Gorges” we verify how MPC works; and the numeric results show MPC’s efficiency. We can see that MPC might have important future in safety risk management of LSE, because as long as the model can be predictive, regardless of an accurate mathematical model of the system, MPC can control a dynamic and uncertain system very well, which is also a characteristic of the safety risk of LSE.

With the rapid development of bridge engineering, the application of supporting and protecting structure that embeds in deep foundation engineering is gradually applied in the design and construction of large-scale suspension bridge. Due to the few amount of stress gauge steel that are pre-installed in the diaphragm wall reinforcement cage in deep foundation engineering and discrete data, it is difficult for the existing frequently-used method to reflect stresses of each part of the wall truly. And for major engineerings, like anchorage foundation, it is necessary to know the stress state of diaphragm wall on time in order to provide basis for decision making for the excavation site. By adopting finite element method, demonstrating the characteristics of its force reversely by using wall deformation and based on the wall deformation data measured in the field, this paper provides strong guarantee for ensuring the safety and reliability of construction.
