Determination of Opportunistic Pathogens and Antimicrobial Resistance Characterization Isolated From Rainbow Trout in Turkey

Vibrio (also known as Listonella) species are widely distributed in aquatic environments and are often mentioned as the aquatic equivalent of aeromonads. The aim of this study is to investigate the exposure of rainbow trouts with V. anguillarum, and determination of antimicrobial resistance of the isolated strains. A total of 100 rainbow trout (Oncorhynchus mykiss) samples were collected from various commercial fish hatcheries in the Aegean Region of Turkey in June 2013. Twelve V. anguillarum were isolated from 100 rainbow trout (Oncorhynchus mykiss) samples. V. anguillarum isolates were resistant to cloxacillin, ampicillin, sulfamethoxazol-trimethoprime and erythromycin in the ratio of 100 %. High levels of resistance are considered to be the result of random antibiotic therapies in aquaculture, hence the results of this study indicated that identification of etiological agent and the exact chemotherapy must be applied in order to prevent residual water contamination and resistant strains of V. anguillarum.

Bacterioferritins (Bfrs) are ferritin-like molecules with a hollow spherical 24-mer complex design that are unique to bacterial and archaeal species. They play a critical role in storing iron(III) within the complex at concentrations much higher than the feasible solubility limits of iron(III), thus maintaining iron homeostasis within cells. Here, the crystal structure of bacterioferritin from Achromobacter (Ach Bfr) that crystallized serendipitously during a crystallization attempt of an unrelated mycobacterial protein is reported at 1.95 Å resolution. Notably, Fe atoms were bound to the structure along with a porphyrin ring sandwiched between the subunits of a dimer. Furthermore, the dinuclear ferroxidase center of Ach Bfr has only a single iron bound, in contrast to the two Fe atoms in other Bfrs. The structure of Ach Bfr clearly demonstrates the substitution of a glutamate residue, which is involved in the interaction with the second Fe atom, by a threonine and the consequent absence of another Fe atom there. The iron at the dinuclear center has a tetravalent coordination, while a second iron with a hexavalent coordination was found within the porphyrin ring, generating a heme moiety. Achromobacter spp. are known opportunistic pathogens; this structure enhances the current understanding of their iron metabolism and regulation, and importantly will be useful in the design of small-molecule inhibitors against this protein through a structure-guided approach.

Ureaplasma urealyticum and U. parvum are mollicutes species that colonize the urogenital tract of many asymptomatic persons but are also thought to be associated with symptomatic infections. Using 170 strains isolated between 2016 and 2019 in a German university hospital, resistance was tested by a combination of commercial tests, molecular methods and determination of minimal inhibitory concentrations. Rates of resistance to macrolides, tetracyclines and fluoroquinolones were 0%, 4.1% and 7.1%, respectively.
