Abstract Immunoglobulins (Ig) are essential components in the colostrum of bovine species that enable passive immunization of newborn calves. Concentrations of fat and protein are greater in colostrum compared with mature milk and represent a vital source of energy and nutrients. Colostral IgG was shown to vary between individual dairy cows, but comparative data on different breeds and performance levels are scarce. The objective of the present field study was to investigate the contents of total IgG, fat, protein, and lactose in colostrum in different Swiss and German dairy and dual-purpose breeds. We collected colostrum samples of 458 cows of 13 different breeds (dairy breeds: Brown Swiss, Swiss and German Holstein Friesian, and New Zealand Holstein; dual-purpose breeds: German Fleckvieh, Holstein Friesian × Montbéliarde, Montbéliarde, Murnau-Werdenfels, Original Braunvieh, Pinzgauer, Rhetic Gray, and Simmental; and beef-type crossbred: Charolais × Holstein Friesian). Colostrum samples were obtained between 5 and 900 min after calving and analyzed for total IgG, fat protein, and lactose contents. Immunoglobulin G concentrations varied between 12.7 and 204.0 mg/mL. No effect of breeding purpose (i.e., dairy or dual-purpose) nor of previous lactation yield on IgG content was observed. However, milking of cows for the first time later than 12 h after parturition resulted in lower colostrum IgG concentrations compared with colostrum harvest within 9 h after calving (P < 0.05). Multiparous cows had a higher colostral IgG concentration than primiparous cows (P < 0.0001). Overall, concentrations of IgG and other constituents in colostrum varied widely in the different cattle breeds. High-yielding dairy cows did not have poorer colostrum quality compared with lower-yielding animals or beef and dual-purpose breeds, which suggests an individually different transfer of circulating IgG into colostrum.

Abstract Neonatal calf survival and health is predominantly dependent on sufficient consumption of immunoglobulin G (IgG) and the resulting transfer of passive immunity (TPI). In this study we investigate the potential for continued IgG secretion and temporal kinetics of mammary IgG output in sequential milkings performed at 0, 4, 16, 28, 40 and 52 hours post-calving in Holstein dairy cows. For colostrum (0 hour), we also scrutinize the relationships between IgG concentration, volume, refractometer readings (˚Bx values, Brix ®) and concentration of sugars (lactose and glucose). Mammary transcripts postpartum (0 hour) indicated that active IgG secretion continues beyond the first milking (colostrum; n=4-5). IgG measurements at the different timepoints indicated that colostrum represents only 25.1% of the total IgG produced across the six sequential milking timepoints, with a substantial 48.9% being secreted into transition milk over the next three timepoints (4-, 6- and 28-hour) combined. The differences on the basis of IgG concentrations across 0-, 4- and 16-hour milking timepoints were not statistically significant (p=0.1522; n=9). For colostrum, volume remained highly variable, even with induced let-down prior to milking (n=27). Nonetheless, colostrum IgG secretion was significantly co-regulated with volume (R 2=0.915; p<0.001; n=18), an association that was stronger than that measured for lactose (R 2=0.803; p<0.001; n=18) and glucose (R 2=0.467; p=0.002; n=17). Comparing colostrum ˚Bx values to absolute IgG concentrations showed no correlation (R 2=0.127; p=0.07; n=27); biochemical separation of colostrum components indicated that both proteins and non-protein solutes could affect ˚Bx values (p<0.0001 for both; n=5). This suggests that ˚Bx values do not reasonably indicate IgG concentration to serve as a measure of “colostrum quality.” Additionally, our finding that early transition milk (4-, 6- and 28-hour) can contribute substantially more IgG than colostrum forces a rethink of existing feeding paradigms and means to maximize TPI in calves. Collectively, our results reveal the remarkable value of early transition milk and caveats to colostrum assessments that could advance application in enhancing neonatal calf health.

Subacute sclerosing panencephalitis (SSPE) is a slowly progressing disease of the CNS in children which is caused by measles virus. Ferrets immunized with measles virus prior to inoculation with the cell associated, syncytiogenic D.R. strain of SSPE virus exhibit characteristics very similar to the human disease. Measles virus nucleocapsids are present, high measles antibody titers are found in the sera and inflammatory lesions are prominent in the brains. Measles virus specific immunoglobulin G (IgG) is present in the brain,and IgG/ albumin ratios indicate that the antibodies are synthesized within the CNS.

Individual and combined effects of several isomers of retinoic acid (RA) and 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) on interferon-gamma (IFN-gamma) secretion by blood mononuclear leukocytes (MNL) from nulliparous and postparturient Holstein cattle were evaluated in vitro. In the first experiment, effects on incubation period (24 to 72 hours) and time of supplementation (0 to 32 hours) with all-trans, 9-cis, 13-cis-, and 9,13-dicis-RAs (0 to 100 nM) on IFN-gamma secretion by pokeweed mitogen (PWM)-stimulated (0 and 10 mug/ml) MNL from nulliparous cattle were evaluated. In the second experiment, MNL from postparturient cows (bled at 0, 2, 4, and 16 days postpartum) were stimulated with PWM (0 and 10 mug/ml) in the presence of RA isomers (9-cis- or 9,13-dicis-RA; 0 to 100 nM), 1,25-(OH)2D3 (0 to 100 nM), or with combinations of these metabolites. The results show that individual isomers of RA had no effect on IFN-gamma secretion by PWM-stimulated MNL from nulliparous or postparturient cows. Furthermore 1,25-dihydroxyvitamin D3 inhibited IFN-gamma secretion by MNL from nulliparous and postparturient dairy cows; however, the degree of inhibition was greater when 9-cis- and 9,13-dicis-RA were also present in the cultures. Finally mononuclear leukocytes from postparturient dairy cows produced substantially less IFN-gamma than did MNL from nulliparous cattle. It is concluded that retinoic acids individually did not affect the capacity of leukocytes from dairy cattle to secrete IFN-gamma. This result is in marked contrast to studies in monogastric species indicating that RAs inhibit IFN-gamma secretion by peripheral blood T cells. Inhibition of IFN-gamma secretion by 1,25-(OH)2D3 was potentiated by 9-cis- and 9,13-di-cis-retinoics acids, suggesting that an excess of dietary vitamins A and D may compromise further the naturally immunosuppressed postparturient dairy cow. Additional research is necessary to determine if the combined effects of these metabolites on IFN-gamma secretion represent an increased susceptibility of the dairy cow to infectious diseases during the periparturient period. Lower secretion of IFN-gamma by MNL from postpartutient dairy cows, relative to nulliparous cattle, suggests that recently-calved cows are naturally immunosuppressed.
