To resolve discrepancies between studies on the effects of emotional content on working memory, and to examine changes with age, this study examined the effects of emotional content on working memory and subsequent long-term memory for targets and distracters. Thirty-six younger (ages 18-29) and 36 older adults (ages 65-87) participated in a working memory task in which they viewed two target words intermixed with two distracters followed by a probe word, and responded to whether the probe was a target word. The emotional content (valence and arousal) of targets and distracters was manipulated. Subsequent long-term memory was tested with a free recall task. Results indicated that emotional content of targets facilitated working adults experiencing disruption from positive and neutral distracters, and older adults experiencing interference from negative distracters. Emotional effects in long-term memory were only seen for younger adults.

Background In animal models, neonatal exposure to volatile anesthetics induces neuroapoptosis, leading to memory deficits in adulthood. However, effects of neonatal exposure to desflurane are largely unknown. Methods Six-day-old C57BL/6 mice were exposed to equivalent doses of desflurane, sevoflurane, or isoflurane for 3 or 6 h. Minimum alveolar concentration was determined by the tail-clamp method as a function of anesthesia duration. Apoptosis was evaluated by immunohistochemical staining for activated caspase-3, and by TUNEL. Western blot analysis for cleaved poly-(adenosine diphosphate-ribose) polymerase was performed to examine apoptosis comparatively. The open-field, elevated plus-maze, Y-maze, and fear conditioning tests were performed to evaluate general activity, anxiety-related behavior, working memory, and long-term memory, respectively. Results Minimum alveolar concentrations at 1 h were determined to be 11.5% for desflurane, 3.8% for sevoflurane, and 2.7% for isoflurane in 6-day-old mice. Neonatal exposure to desflurane (8%) induced neuroapoptosis with an anatomic pattern similar to that of sevoflurane or isoflurane; however, desflurane induced significantly greater levels of neuroapoptosis than almost equivalent doses of sevoflurane (3%) or isoflurane (2%). In adulthood, mice treated with these anesthetics had impaired long-term memory, whereas no significant anomalies were detected in the open-field and the elevated plus-maze tests. Although performance in a working memory task was normal in mice exposed neonatally to sevoflurane or isoflurane, mice exposed to desflurane had significantly impaired working memory. Conclusions In an animal model, neonatal desflurane exposure induced more neuroapoptosis than did sevoflurane or isoflurane and impaired working memory, suggesting that desflurane is more neurotoxic than sevoflurane or isoflurane.

In tests of working memory with verbal or spatial materials repeating the same memory sets across trials leads to improved memory performance. This well-established “Hebb repetition effect” could not be shown for visual materials. This absence of the Hebb effect can be explained in two ways: Either persons fail to acquire a long-term memory representation of the repeated memory sets, or they acquire such long-term memory representations, but fail to use them during the working memory task. In two experiments, (N1 = 18 and N2 = 30), we aimed to decide between these two possibilities by manipulating the long-term memory knowledge of some of the memory sets used in a change-detection task. Before the change-detection test, participants learned three arrays of colors to criterion. The subsequent change-detection test contained both previously learned and new color arrays. Change detection performance was better on previously learned compared to new arrays, showing that long-term memory is used in change detection.

Abstract Introduction It is widely accepted that “offline” processes during sleep contributes to memory. Working Memory (WM) capacity, which reflects “online” memory processing, is an important factor influencing cognitive functioning, which declines with age. In younger individuals, a positive association is reported between WM-capacity and declarative memory improvement. Methods We examined the relation between WM and long-term memory consolidation, among younger [N=105, 18-25yr] and older adults (N=119, 60-85yr). Subjects completed an OSPAN WM task, encoded a Word-Paired Association (WPA) task in the morning (Test1), and were tested on the WPA in the afternoon (Test2) after a 90-minute polysomnographically-recorded nap or wake. Half of the subjects were exposed to negatively valenced word-pairs (EWPA) while the other half were exposed to neutral word-pairs (NWPA). Subjects rated valence of the word-pairs at Test1 and Test2. We compared the four groups (young-EWPA, young-NWPA, old-EWPA and old-NWPA) on WM and WPA in both wake and sleep. Results In both wake and sleep, in the WPA, ageXword-condition interaction was found (p=.004). Post-hoc analysis revealed that in wake, younger-EWPA had higher performance (p=.03) than younger-NWPA, however, older-EWPA had lower performance (p=.03) than older-NWPA. Additionally, we found an ageXword-condition interaction whereby youngers showed no change in ratings, while older adults rated word-pairs more positively both in wake (p=.03) and sleep (p=.002) at Test 2. Youngers had higher WM performance (p=.007), also their WM performance was positively associated with WPA both for Neutral (p=.03) and Emotional (p=.01). WM and WPA among older adults was not related. In younger-EWPA, Stage2-sleep-minutes was positively associated to WPA improvement (p=.03) where this association was negative among older-EWPA (p=.02). In older-NWPA, Stage2-sleep-minutes was positively associated with WPA (p=.004). Conclusion Our findings indicate an association between WM and emotionally-salient memory formation that is modulated by age. Older adults, but not younger, showed the emotional bias previously reported. WM was higher in younger adults related to memory improvement. Stage2-sleep was related to memory improvement in both groups, but in opposite directions. In sum, the role of sleep in memory consolidation changes with aging and WM may play a role in this process. Visual working memory is the cognitive system that holds visual information active to make it resistant to interference from new perceptual input. Information about simple stimuli – colors, orientations – is encoded into working memory rapidly: in under 100ms, working memory ‘fills up’, revealing a stark capacity limit. However, for real-world objects, the same behavioral limits do not hold: with increasing encoding time, people store more real-world objects and do so with more detail. This boost in performance for real-world objects is generally assumed to reflect the use of a separate episodic long-term memory system, rather than working memory. Here we show that this behavioral increase in capacity with real-world objects is not solely due to the use of separate episodic long-term memory systems. In particular, we show that this increase is a result of active storage in working memory, as shown by directly measuring neural activity during the delay period of a working memory task using EEG. These data challenge fixed capacity working memory models, and demonstrate that working memory and its capacity limitations are dependent upon our existing knowledge.

AbstractHumans have remarkable visual long-term memory abilities, capable of storing thousands of objects with significant detail. However, it remains unknown how such memory is utilized during the short-term maintenance of information. Specifically, if people have a previously encoded memory for an item, how does this affect subsequent working memory for that same item? Here, we demonstrate people can quickly and accurately make use of visual long-term memories and therefore maintain less perceptual information actively in working memory. We assessed how much perceptual information is actively maintained in working memory by measuring neural activity during the delay period of a working memory task using electroencephalography. We find that despite maintaining less perceptual information in working memory when long-term memory representations are available, there is no decrement in memory performance. This suggests under certain circumstances people can dynamically disengage working memory maintenance and instead use long-term memories when available. However, this does not mean participants always utilize long-term memory. In a follow-up experiment, we introduced additional perceptual interference into working memory and found participants actively maintained items in working memory even when they had existing long-term memories available. These results clarify the kinds of conditions under which long-term and working memory operate. Specifically, working memory is engaged when new information is encountered or perceptual interference is high. Visual long-term memory may otherwise be rapidly accessed and utilized in lieu of active perceptual maintenance. These data demonstrate the interactions between working memory and long-term memory are more dynamic and fluid than previously thought.

Older adults show a pronounced decline in long-term memory (LTM), but the source of this deficit is still debated. The present study investigated whether deficient engagement in refreshing and elaboration at the level of working memory (WM) causes this deficit. Our results show that the benefit of refreshing in WM was unaffected by age. Refreshing had no effect on LTM in both young and older adults. Elaboration benefited LTM in young adults, but not in older adults. Therefore, the LTM deficit of older adults might arise at least in part from a deficit in the process of elaboration.
