We investigate the recent history and stock identity of beluga whales (Delphinapterus leucas) in Kotzebue Sound in the Chukchi Sea, a region long frequented by large numbers of belugas in summer until their near disappearance in the 1980s. Wide variation in numbers since then suggests a complex recent history that hinders recovery efforts. Analysis of teeth sampled during the historical (pre-decline) era using ancient DNA (aDNA) methods found that the original Kotzebue Sound whales were differentiated for mitochondrial DNA (mtDNA) from other summering concentrations across the Pacific Arctic revealing a demographically distinct subpopulation where long-established migratory culture likely facilitated population divergence. Analysis of microsatellite (nDNA) and mtDNA markers in belugas from the contemporary (post-decline) era revealed that whales from other stocks likely visited Kotzebue Sound, including during two low ice years when relatively large numbers of whales were present. Some mtDNA lineages were found only in Kotzebue Sound, with one recorded in both the historical and contemporary eras. Exclusion tests found a number of whales in Kotzebue Sound during the contemporary era that had nDNA genotypes unlikely to arise in other contemporary stocks in the Pacific Arctic. Our findings indicate that the Kotzebue Sound belugas comprised a unique stock of which a few remnants likely still co-occur with belugas from other larger stocks. We recommend that the US government work through the co-management process to greatly reduce or eliminate the taking of belugas, especially adult females, likely to belong to the Kotzebue Sound stock, until they recover.

The eastern Chukchi Sea (ECS) stock of beluga whales is one of three stocks in western Alaska that are co-managed by the National Marine Fisheries Service and the Alaska Beluga Whale Committee. Abundance of this stock was estimated as 3710 in 1991 from incomplete data. Analysis of data from satellite-linked time-depth recorders (SDRs) attached to belugas in summer concentration areas of the ECS and Beaufort Sea (BS) stocks provided an overview of beluga distribution and movements and allowed the identification of an area (140˚ W to 157˚ W in the BS) and a time period (19 July – 20 August) in which the distributions of the two stocks do not overlap. Aerial survey data were collected by the Aerial Surveys of Arctic Marine Mammals (ASAMM) project in that region and time period in 2012. We used those data in a line transect analysis that estimated there were 5547 (CV = 0.22) surface-visible belugas in the study area. Data from SDRs were used to develop correction factors to account for animals that were missed because they were either outside of the study area or diving too deep to be seen, resulting in a total abundance estimate of 20 752 (CV = 0.70). The average annual Alaska Native subsistence harvest from the ECS stock (57) is about 0.3% of the population estimate. Without data collected by the ASAMM project and from satellite-linked tags, this analysis would not have been possible. Additional surveys and tagging of ECS belugas are warranted.

Cytokines are fundamental for a functioning immune system, and thus potentially serve as important indicators of animal health. Quantitation of mRNA using quantitative reverse transcription polymerase chain reaction (qRT-PCR) is an established immunological technique. It is particularly suitable for detecting the expression of proteins against which monoclonal antibodies are not available. In this study, we developed a probe-based quantitative gene expression assay for immunological assessment of captive beluga whales (Delphinapterus leucas) that is one of the most common cetacean species on display in aquariums worldwide. Six immunologically relevant genes (IL-2Rα, -4, -10, -12, TNFα, and IFNγ) were selected for analysis, and two validated housekeeping genes (PGK1 and RPL4) with stable expression were used as reference genes. Sixteen blood samples were obtained from four animals with different health conditions and stored in RNAlater™ solution. These samples were used for RNA extraction followed by qRT-PCR analysis. Analysis of gene transcripts was performed by relative quantitation using the comparative Cq method with the integration of amplification efficiency and two reference genes. The expression levels of each gene in the samples from clinically healthy animals were normally distributed. Transcript outliers for IL-2Rα, IL-4, IL-12, TNFα, and IFNγ were noticed in four samples collected from two clinically unhealthy animals. This assay has the potential to identify immune system deviation from normal state, which is caused by health problems. Furthermore, knowing the immune status of captive cetaceans could help both trainers and veterinarians in implementing preventive approaches prior to disease onset.
