The experiment was conducted to investigate the performance of three different genotypes (BARI Sarisha-6, BARI Sarisha-8, and BARI Sarisha-11) in two different media viz., MS and B5 with different concentrations of phytohormone (2, 4-D) for callus induction from uninucleate stage anthers of Brassica and subsequent plant regeneration in MS media with different concentrations of phytohormone (BAP and NAA). Among the genotypes, BARI Sarisha-8 showed the best performance for all the parameters of callus induction. The performance of BARI Sarisha-6 was poor compared to others. Maximum rate of callus induction (%) was observed in MS + 0.5 mg/L 2, 4-D followed by B5 + 0.5 mg/L 2,4-D. The media combination MS + 1.0 mg/L BAP 0.3 mg/L 2,4-D showed the best performance for maintenance of calli. Significant variations were observed among the genotypes and media composition for shoot regeneration. Among the genotypes, BARI Sarisha-8 showed the best performance for shoot regeneration followed by BARJ Sarisha-l1. The genotype BARI Sarisha-8 produced higher percent of shoots/calli and required minimum days for shoot initiation. Higher percent calli without shoot were produced by the genotype BARI Sarisha-6. The media combination MS + 2.0 mg/L BAP + 0.5 mg/L NAA showed the best performance for shoot regeneration and required maximum days for shoot initiation. The experiment was conducted during May to December 2008 in the Biotechnology Laboratory of Bangladesh Agricultural University, Mymensingh to observe the callus induction, regeneration potentiality and to establish a suitable in vitro plantlet regeneration protocol of Corchorus olitorius. MS medium supplemented with different phytohormone concentrations and combinations were used to observe the callus induction, shoot regeneration and root formation ability of the cotyledon with attached petiole derived explant of three genotypes viz. O-9897, O-72 and OM-1. The highest callus induction (92.85%) was observed in O-9897 followed by O-72 (82.14%) in the MS media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA. Genotype O-9897 in MS media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA produced the highest percentage of shoot regenerants (83.33%) followed by O-72 (75.00%) in the media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA. The root formation from regenerants was the best on halfstrength of MS media supplemented with 0.6 mg/L IBA in genotype O-9897 (45.00%). The in vitro regenerated plantlets from the genotypes O-9897 could be established in the field. Therefore, the genotypes O-9897 of C. olitorius in MS media supplemented with 2.5 mg/L BAP + 0.5 mg/L IAA could be used for callus induction and shoot regeneration. Anther of five varieties of Brassica species, namely BARI Shariaha-7, Tori-7, Agrani, Daulat and Safal were cultured in vitro to observe their regeneration potentiality. Different concentrations and combinations of growth regulators were supplemented in MS medium. The range of callus induction was 12.50-87.50 %. Maximum callus induction (75.00%) was observed on MS +4 mg/L 2, 4-D + 1.0 mg/L BAP. Among the genotypes, BARI Sharisha-7 showed the highest percentage of callus induction (60.42%). Among the treatments, highest percentage of shoot regeneration (75.00%) was observed on MS + 4 mg/L BAP + 1.0 mg/L NAA. BARI Sharisha-7 also showed the highest rate of plant regeneration (66.67%). Root induction was highest (75%) on half strength MS medium supplemented with 1.0 mg/L IBA and 0.5 mg/L NAA. The plantlets with sufficient roots thus obtained were transferred successfully to plastic pots and subsequently to the field. BARI Sharisha-7 and Tori-7 survived easily in the pots as well as in the field but Safal was very poor in survivability both in the pots and in the field. This study was undertaken to establish a regeneration protocol for two scented indica rice varieties, namely Pusa Basmati1 and Kalanamak. Callus culture in Pusa Basmati1 was initiated in MS containing 2.0 mg/l of 2,4-D. Optimum requirement of growth hormones for callus induction in Kalanamak was 1.5 mg/l of 2,4-D supplemented with 0.1 mg/l each of NAA and BAP in MS. Shoot regeneration in Pusa Basmati1 initiated in MS containing 2.0 mg/l BAP, 0.5 mg/l each of NAA and Kn. There was no shoot initiation in MS with same composition in Kalanamak. The shoot regeneration was successfully initiated and achieved in Dl medium supplemented with 2.0 mg/l BAP, 0.2 mg/l each of IAA, NAA and Kn, 500 mg/l each of proline and glutamine and 800 mg/l casein hydrolysate. The calli derived from mature seed embryo produced fertile green plants. The plants were successfully transferred to field with normal flowering. This study was carried out to improve an efficient protocol for in vitro callus induction and plant regeneration using Bangladeshi barley genotypes collected from BARI, Gazipur, Bangladesh. After sterilization embryos were separated carefully from mature seeds of six barley genotypes (BB-1, BB-2, BB-3, BB-4, BB-5 and BB-6) and cultured them in MS medium supplemented with various concentration and combination of PGRs for callus induction and regeneration. Out of six genotypes BB-6 showed highest (38.17%) callus induction in MS + 4.0 mg/l 2,4-D + 200 mg/l L-proline + 300 mg/l casein hydrolysate; whereas, BB-4 and BB-5 showed no callus induction in the same medium. For plant regeneration from embryogenic calli the same genotype (BB-6) also performed the best results (19.25%) in MS medium supplemented with 1.5 mg/l BAP + 30 g/l sucrose. In vitro selection of Foeniculum vulgare for salt tolerance was undertaken by the use of somaclonal variation. In this idea, explants of root, hypocotyl and cotyledon of sterilized seedling were transferred to callus and regeneration media with concentrations of 0, 50, 100 and 150 mM of NaCl. After 4 weeks, calli induction, regeneration frequency and calli fresh and dry weights, in both control and stress conditions, were measured. The results showed that salinity caused a significant decrease in the callus induction and shoot regeneration of fennel. However, in the presence of 100 and 150 mM NaCl, the highest frequency of callus induction in hypocotyl and cotyledon explants was recorded on the media supplemented with 1 mg l-1 IAA (Indol-3-Acetic Acid) plus 1 mg l-1 2,4-D (2, 4- dichlorophenoxyacetic acid) and 2 mg l-1 kinetin. Among different growth regulator treatments, the combination of 2 mg l-1 NAA (Naphtaleneacetic acid) and 0.5 mg l-1 kinetin was found to be the most effective for shoot regeneration under stress condition. The highest dose of NaCl (150 mM) inhibited callus induction and shoot regeneration compared to control with 41% and 96% respectively. The calli fresh and dry weights of all explants were decreased with the increas of NaCl concentration. The highest and the lowest of dry and fresh weight of calli were observed in 0 and 150 mM respectively.

Petiole of six genotypes of oilseed Brassica viz. Tori-7, Sampad, Kallyania, BARI Sarisha-7, BARI Sarisha-8, and MM 20-3 were cultured in MS medium with different concentrations of BAP, NAA, and AgNO3 for callus induction and subsequent plant regeneration. The highest percentage of callus induction (91.43%) was observed in Tori-7 in the media supplemented with 2 mg/L BAP, 0.1 mg/L NAA and 2.0 mg/L AgNO3. Calli were maintained in order to get sufficient number of regenerants. With the increased concentration of BAP, the highest percentage (57.14) of regenerants were found in Tori-7 followed by Sampad (33.13%) and BARI Sarisha-8 (31.42%) in MS media supplemented with 2.5 mg/L BAP, 0.1 mg/L NAA and 2.0 mg/L AgNO3. Root formation from the regenerants was found best in half MS medium supplemented with 0.5 mg/L NAA in genotype Tori-7. Lychnis cognate Maxim and Lychnis fulgens Fish. Ex Spreng are two valued ornamental plants in Korea. Soaking of seeds in GA3 solution remarkably promoted germination up to 60%, but the control (0 mg/l) was not effective (> 5%). To select an adequate temperature for seed germination, seeds, previously soaked in a 1000 mg/l GA3 for 24 hrs, were incubated at 15, 20, 25, and 30°C. Seed germination of over 20% was obtained at 15, 20, and 25°C, but only 10% at 30°C. These results indicate that the seeds of L. cognate and L. fulgens are in a such dormant state that they hardly germinate even by dormancy breaker (GA3) and low (15 ? 25°C) temperature treatment. The highest callus induction was observed in the leaf explants of the seedlings on MS containing specific concentrations of 3.0 mg/l BA and 1.0 mg/l NAA. The adventitious shoot was formed < 90% of calli on 1/2 WPM medium. The height of in vitro propagated plantlet was no different media used for regeneration. High frequency plant regeneration protocol has been standardized from banana cultivar Musa paradisiaca cv. Karibale Monthan, an endemic cultivar of Malnad region of Karnataka. The fruits are used as glomerular protective to solve kidney problems. To minimize the microbial contamination and to promote healthy growth, explants were treated with 70 % absolute alcohol for 6 min, 0.1 % Mercuric chloride for 10 min and 0.2 % for 10 min, 1 % Sodium hypochlorite for 15 min, 0.1 % Cefotaxime for 5 min and 0.05 % Gentamicin for 5 min. The high frequency shoot initiation (93.33 %) was recorded at 5 mg/l BAP. The synergetic effect of BAP (4 to 6 mg/l), TDZ (0.1 to 1.2 mg/l) and coconut water (0.1 to 0.9 ml/l) induced multiple shoot buds and it was optimized at the concentration of 5 mg/l BAP, 0.5 mg/l TDZ and 0.5 ml/l coconut water with 15.90 ± 1.66 frequency of shoots per propagule. Supplementation of 1.0 mg/l IBA induced 5.33 ± 1.21 numbers of roots with a mean root length of 7.50 ± 1.87 roots. The 99% of plantlets with distinct roots and shoots were successfully acclimatized in the green house and transferred to the field to evaluate the agro-morphological variations. The weight of the bunch (kg), number of hands in a bunch, number of fingers in a hand, length of the finger (cm), girth of the finger (cm) and girth of the pseudostem (cm) exhibited by in vitro plants were higher than the in vivo plants.Int J Appl Sci Biotechnol, Vol 3(2): 202-209 DOI: http://dx.doi.org/10.3126/ijasbt.v3i2.12536

ABSTRACT<br /><br />The breeding of rainfed rice tolerant to drought can be accomplished using anther culture. The objectives of this research were to determine regeneration abilities of six F1 anther culture and its acclimatization ability. The experiment was arranged in completely randomized design with 14 replications. The treatments consisted of six F1 derived from crossing:  INPARI 18 x IR83140-B-11-B (G1), INPARI 18 x B12825E-TB-1-25 (G2), INPARI 18 x IR87705-14-11-B-SKI-12 (G3), INPARI 22 x IR83140-B-11-B (G4), Bio-R81 x O18b-1 (G5), Bio-R82-2 x O18b-1 (G6). Media for callus induction was based on N6 medium + 2.0 mg L-1 NAA + 0.5 mg L-1 kinetin + 1.0 mM putresin + 60 g L-1 sucrosa, media for regeneration was based on MS + 0.5 mg L-1 NAA + 2.0 mg L-1 kinetin + 1.0 mM  putresin, and media for rooting was based on  MS + 0.5 mg L-1 IBA + 30 g L-1 sucrosa. The result indicated that all six F1 had different ability in anther culture. Bio-R82-2 x O18-b1 (G6) and  Bio-R81 x O18-b1 (G5) F1 genotype had good response both of callus induction and plant regeneration. These two F1 genotypes also gave the highest ratio of green planlet production to number of anther inoculated (GP:AI) were 5.50% and 4.65%,  respectively. In this research, there were identified doubled haploid plants were developed from 4 F1 derived cross namely G2 (2 plants), G3 (4 plants),  G5 (21 plants), and G6 (26 plants).<br /><br />Keywords: Callus induction, doubled haploid, rice<br /><br />
