Pearl millet is an important food and fodder crop cultivated in the arid and semi-arid regions of Africa and Asia, and is now expanding to other regions for forage purpose. This study was conducted to better understand the forage quantity and quality traits to enhance the feed value of this crop. Two sets of pearl millet hybrids (80 single cross hybrids in Set-I and 50 top cross hybrids in Set-II) along with their parents evaluated multi-locationally for the forage-linked traits under multi-cut (two cuts) system revealed significant variability for the forage traits in the hybrids and parents. The mean better parent heterosis (BPH) for total dry forage yield (TDFY) was 136% across all the single cross hybrids and 57% across all the top cross hybrids. The mean BPH for in vitro organic matter digestibility (IVOMD) varied from −11 to 7% in the single cross hybrids and −13 to 11% in the top cross hybrids across cuts. The findings of TDFY and IVOMD heterosis in these sets indicated the potential of improvement of the hybrid cultivars for forage quantity and quality in forage pearl millet. The parental lines single cross parent (SCP)-L02, SCP-L06, and top cross parent (TCP)-T08 found superior in the forage quantity and quality traits can be utilized in the future breeding programs. Most of the forage traits were found to be controlled by using the non-additive gene action. A diverse panel of 105 forage-type hybrid parents (Set-III) genotyped following genotyping by sequencing (GBS) and phenotyped for crude protein (CP) and IVOMD under multi-cuts for 2 years identified one stable significant single nucleotide polymorphism (SNP) on LG4 for CP, and nine SNPs for IVOMD distributed across all the linkage groups except on LG2. The identified loci, once validated, then could be used for the forage quality traits improvement in pearl millet through marker-assisted selection.

Single-cross hybrids of orchard grass (Dactylis glomerata L.) were evaluated for yield, in vitro digestibility (IVD), and relative maturity in three trials. All trials were planted broadcast, using seed produced on male-sterile clones. All trials were harvested for 2 yr. The average yield of the 48 hybrids tested was 98% of that for the cultivar Frode in the first cut each year, and 103% over the season. The most productive hybrid produced 28% more forage than Frode in the first cut, and 15% more over the season. For IVD, there was no evidence of hybrid superiority. The results indicate that the use of hybrids can result in an increase in forage yield.

Genetic diversity of 130 forage-type hybrid parents of pearl millet was investigated based on multiple season data of morphological traits and two type of markers: SSRs (Simple sequence repeats) and GBS identified SNPs (Genotyping by sequencing-Single nucleotide polymorphism). Most of the seed and pollinator parents clustered into two clear-cut separate groups based on marker based genetic distance. Significant variations were found for forage related morphological traits at different cutting intervals (first and second cut) in hybrid parents. Across two cuts, crude protein (CP) varied from 11% to 15%, while in vitro organic matter digestibility (IVOMD) varied from 51% to 56%. Eighty hybrids evaluated in multi-location trial along with their parents for forage traits showed that significant heterosis can be realized for forage traits. A low but positive significant correlation found between SSR based genetic distance (GD between parents of hybrid) and heterosis for most of the forage traits indicated that SSR-based GD can be used for predicting heterosis for GFY, DFY and CP in pearl millet. An attempt was made to associate marker-based clusters with forage quality traits, to enable breeders select parents for crossing purposes in forage breeding programs.

Pollen grains from three inbreds (H49, H55, and H50) and two of their single cross hybrids (H49 × H55 and H49 × H50) were cultured on an artificial medium containing 15% sucrose and 0.6% bactoagar supplemented with all possible combinations of two levels (0.00 and 0.03%) of calcium nitrate (Ca) and two levels (0.00 and 0.01%) of boric acid (B). For all characteristics measured (germination percentage, germination with more than one pollen tube percentage, rupturing percentage, length and growth rate of the pollen tubes), significant differences between inbreds were obtained on most combinations of Ca and B. Since the inbreds were presumably homozygous and, as a result, each produced pollen grains of only one genotype, these differences in germination characteristics were apparently produced by the genetic differences between the inbreds. Therefore, the in vitro germination characteristics of pollen grains are associated with pollen genotype. The response of the hybrids could not be accurately predicted from the behavior of their inbred parents. In general, the hybrid mean was in the range between the means for the two inbred parents. However, the value of the mean for the hybrid relative to the values of the parental means depended on which hybrid combination was involved. The results indicated that the germination characteristics of pollen grains on various combinations of Ca and B are influenced in large part by pollen genotype but that the genetic expression is quite complex. Vigor of the pollen source per se was not a factor.

Forty-five single-cross hybrids developed (in rabi 2014) from ten inbred lines of quality protein maize through diallel mating design along with four checks viz., Pratap QPM Hybrid- 1, Vivek QPM- 9, HQPM- 1 and HQPM-5 were evaluated in randomized block design with three replications for yield and quality traits during kharif-2014, to estimate the gca (general combining ability) of the parents and sca (specific combining ability) of hybrids considered for the development of high yielding varieties. The analysis of variance for combining ability revealed significant mean sum of squares due to gca and sca for all the traits, except mean sum of square due to gca for num-ber of grain rows per ear. Ratio of σ2 sca / σ 2 gca was greater than one for all the traits, thereby indicating the preponderance of non-additive gene effects in the expression of these traits. Inbred line P8 and P5 has been found good general combiner with highest magnitude of gca effects 10.46 and 8.89, respectively and high per se i.e. 52.33 g and 44.67 g, respectively for grain yield per plant and majority of traits. Hybrids P6xP8, P5xP8, P3xP5, P5xP7 and P1xP8 showed higher significant positive sca effects ranged from 25.66 to 34.59 along with good per se ranged from 98.00 to 107.67 g for grain yield per plant. These hybrids also exhibited significant positive sca effects for most of the yield and quality traits under study, indicating potential and may be used for exploiting hybrid vigour in in QPM hybrid breeding programmes.

SummaryPlatelets were isolated from blood of baboons and treated with neuraminidase to remove platelet membrane sialic acid, a process which artificially ages the platelets. The platelets were then labelled with 111In and their mean life span, in vivo distribution and sites of Sequestration were measured. The effect of removal of sialic acid on the attachment of immunoglobulin to platelets were investigated and related to the Sequestration of the platelets by the spleen, liver, and bone marrow. Removal of sialic acid by neuraminidase did not affect the aggregation of platelets by agonists in vitro, nor their sites of Sequestration. The removal of 0.51 (median, range 0.01 to 2.10) nmol sialic acid/108 platelets shortened their life span by 75 h (median, range 0 to 132) h (n = 19, p <0.001), and there was an exponential correlation between the shortening of the mean platelet life span and the amount of sialic acid removed. The increase in platelet-associated IgG was 0.112 (median, range 0.007 to 0.309) fg/platelet (n = 25, p <0.001) after 0.79 (median, range 0.00 to 6.70) nmol sialic acid/108 platelets was removed (p <0.001). There was an exponential correlation between the shortening of mean platelet life span after the removal of sialic acid and the increase in platelet-associated IgG. The results suggest that platelet membrane sialic acid influences ageing of circulating platelets, and that the loss of sialic acid may have exposed a senescent cell antigen that binds IgG on the platelet membrane. The antibody-antigen complex may then provide a signal to the macrophages that the platelet is old, and can be phagocytosed and destroyed.
