Although it is well known that the biological and physical characteristics of skin mucus in fishes are strongly affected by changes in environmental conditions, the influence of photoperiod and time-dependent bacterial endotoxin stimulation is not well documented. In the present study, we determined the diel variations in the basal activities of secretory proteins with known defense functions in the skin mucus of Nile tilapia (Oreochromis niloticus) maintained under two photic environments: equal length of day and night (12L:12D, LD) or total darkness (0L:24D, DD). A second experiment was conducted to determine how time-dependent (i.e., day versus night) lipopolysaccharide (LPS) challenge could influence these skin mucosal defenses. The results revealed that LD signal differentially modulated the activities of mucosal immune molecules. Fish subjected to LD regime showed significantly higher levels of skin mucus lysozyme and protease at nighttime than at daytime. This distinct feature was not observed in fish under DD. There was no general mucosal response patterns to time-dependent LPS challenge. Nonetheless, protease and lysozyme, which were identified to be at elevated levels at night, were significantly modulated when the endotoxin was administered at nighttime. Ceruloplasmin was the only molecule that responded to LPS challenge at daytime, where its activity significantly increased at 8 h post-stimulation. Collectively, the results revealed that photoperiod cues influenced the activities of mucosal defenses and this may play, at least in part, in the temporal sensitivity to bacterial endotoxin.

Fish meal (FM) is no longer a sustainable source for the increasing aquaculture industry. Animal proteins from insects may be used as a FM alternative source as long as they do not create adverse effects in fish. Black soldier fly larvae meal (BSFLM) was tested in a 12-week experiment on Nile tilapia (Oreochromis niloticus). Four hundred and twenty (14.77 ± 2.09 g) fish were divided into seven groups and were fed seven diets: control (0% BSFLM-100% FM), and FM replaced by BSFLM at rates of 10%, 20%, 40%, 60%, 80% and 100%. Growth indexes, feed utilization efficiency indices, feed intake, and survival rate were not significantly different (p > 0.05) between FM and BSFLM fed fish. Values of red blood cell, white blood cells, hemoglobin, hematocrit, mean corpuscular volume and hemoglobin, mean corpuscular hemoglobin concentration, red blood cell distribution width, and platelet values were not affected by BSFLM. Skin, mucus lysozyme, and peroxidase activities were improved in BSFLM fed fish. BSFLM can be used as a substitution for FM in the Nile tilapia (O. niloticus) diet at up to a 100% rate with no adverse effects.

This study aims to apply vaccination against bacterial disease. The purpose of vaccination is to trigger the immune respone both non-specific and specific of fish against bacteria Motile Aeromonas Septicaemia (MAS) caused by Aeromonas hydrophila. The vaccination for fish with different ages: 2-3 weeks and 5 weeks were done using immersion method. The fish were re-vaccinated (booster) after two weeks of the first vaccination. The survival rate was < 50 % for juveniles 2-3 weeks and > 50% for juveniles 5 weeks. Survival rate for juveniles 5 weeks was higher than juveniles of 2-3 weeks. This shows that organs of juveniles of 5 weeks were more complete than the 2-3 weeks juveniles. The age of fish is one of the important factors for successfully vaccination. Penelitian ini bertujuan untuk melakukan penerapan vaksinasi terhadap penyakit bakterial. Vaksinasi ditujukan untuk merangsang respon kekebalan non- spesifik dan spesifik pada tubuh ikan terhadap penyakit Motile Aeromonas Septicaemia (MAS) yang disebabkan oleh bakteri Aeromonas hydrophila. Vaksin diberikan pada benih ikan nila yang berbeda umur yaitu 2-3 minggu dan 5 minggu dengan menggunakan metode perendaman. Ikan divaksinasi ulang (booster) setelah 2 (dua) minggu dari vaksinasi yang pertama. Prosentase kelangsungan hidup < 50 % untuk benih umur 2-3 minggu dan > 50% untuk benih umur 5 minggu. Jumlah kematian benih umur 2-3 minggu lebih tinggi dari 5 minggu. Hal ini menunjukkan bahwa fungsi organ benih umur 5 minggu telah lebih lengkap dari pada benih umur 2-3 minggu. Umur ikan merupakan salah satu faktor penting penentu keberhasilan suatu kegiatan vaksinasi.
