Fractionation processes based on physical separation are a good strategy to produce enriched cereal flours. Therefore, the aim of this work is to evaluate the suitability of sieving of buckwheat flours to produce protein and phenolic (especially rutin) enriched fractions. Because of that, dehulled whole buckwheat flour (GSTQ) was sieved obtaining fractions with a particle size of 215 µm, 160 µm, 85 µm, and 45 µm (GS215, GS160, GS85, and GS45). For that purpose, the determination of protein, ash, and total starch content and free and bound phenolic compounds was carried out. The highest content of total phenolic compounds was obtained in GS215 (3118.84 mg kg−1 d.w.), followed by GS160 (2499.11 mg kg−1 d.w.), GS85 (989.46 mg kg−1 d.w.), GSTQ (983.15 mg kg−1 d.w.), and GS45 (481.31 mg kg−1 d.w.). Therefore, the phenolic content decreased with the particle size decrease from 215 µm to 45 µm. Besides, there were no significant differences between the total phenolic content in GS85 and GSTQ. The fraction with 215 µm reported the highest protein and mineral salt content and presented rutin amounts four times higher than GSTQ.

Neither the original Mannich–Lenz procedure nor its recently proposed revisions proved suitable for the polarimetric determination of starch in gluten. The principal defect lay in the means of removing or correcting for proteins dissolved along with the starch and there also was a need for improvements in technique of starch dispersion.In the present method, the starch of the gluten sample is dispersed by 15 min. boiling in a calcium chloride solution having a specific gravity of 1.30 and a pH of approximately 2.5. Frothing is controlled by the addition of one or two drops of n-octyl alcohol, constant volume being maintained during boiling by the addition of water. The proteins dissolved by this treatment are precipitated with 20% stannic chloride prior to filtration and polarization. This rapid procedure underestimates the starch content but the application of a factor to correct for the constant ratio of underestimation allows the real starch content to be determined with an accuracy of ±0.5%.

A quantitative method of determination by photometric measurement in the iodine-starch complex is described. Determinations were made within 0-250 μg range of starch content in 1 ml of solution. Starch was treated with 0.01N J in 1% KJ and extinction was read at wavelength 610 nm in a 1 cm thick layer of solution, with a "Specol" photocolorimeter. Statistical analysis of measurements showed a positive correlation between the starch amount and extinction.

Abstract A method is presented for extracting and determining the starch content of tobacco. Dried tobacco is ground to pass a 60 mesh screen, interfering colored materials are removed from the tobacco by a methanol-water extraction, and the starch is extracted from the tobacco by a perchloric acid treatment. Final quantitative measurement of the starch is made on a colored starch–iodine complex. The absorbance of this colored complex at 600 nm is proportional to the concentration of the starch in solution and can be related to the starch content of the original tobacco sample. Also included is a procedure for the isolation of pure tobacco starch. Such material was prepared for use as a calibration standard needed for the calculation of starch content of tobacco samples from the colorimetric data.
