Total One hundred fifty seed samples of Lentil from 18 districts namely Ajmer, Bundi, Barmer, Bharatpur, Bikaner, Churu, Dausa, Dholpur, Hanumangarh, Jaipur, Jalore, Jhunjhnu, Karauli, Kota, Nagaur, Pali, Sirohi, Tonk of Rajasthan were analyzed for the detection of seed-borne mycoflora using standard blotter, agar-plate and deep-freezing methods as suggested by ISTA. A Total of 26 fungal species belonging to 16 genera (SBM) were recorded. Of the three methods used, agar plate method was found to be better for the isolation of fungi from lentil seeds. Studies on the biodiversity of phylloplane and endophytic fungi on the leaf surfaces of the medicinal plant; Piper betle L. was made in our Microbiology laboratory, K.M. Govt. Institute for Postgraduate Studies and Research (Autonomous), Puducherry. The Agar plate method was used to isolate both the leaf surface and sub-surface fungi. During the study period, altogether twelve fungal species of seven genera of phylloplane and five species of endophytes were isolated from Piper betle L. by agar plate method. Penicillium sp., a dominant fungus, was chosen for the synthesis of silver nanoparticles. Silver nanoparticles (AgNPs) were synthesized from Ag+ ions by treating with different extracts of the fungus with AgNO3.The appearance of yellowish-brown color in the conical flasks suggested the formation of AgNPs in dark and light conditions. The AgNPs were characterized by UV-Vis spectroscopy, which has proved to be very useful for the analysis of nanoparticles. Candida albicans was found most susceptible towards the AgNPs of the fungus in comparison to other bacterial strains. Among the bacterial strains, Staphylococcus aureus was more suffered than V. parahaemolyticus and E. coli. It was found in the present study that our process for the synthesis of nanoparticles was easy, safe, and economical to be readily used in the field of biomedicine.

This study was conducted on different species of stored grains obtained from Duhok Province, Kurdistan Region, of Iraq to investigate the occurrence of seed borne fungi. Nine types of grains were collected and two methods were selected (agar plate method and blotter method) for fungal isolation. A total of 19 species assigned to 13 genera were identified. The high frequent genera were Aspergillus (4 species), Penicillium, Alternaria and Fusarium (2 species), while other genera include Rhizopus, Cladosporium, Stemphylium, Ulocladium, Humicola, Bipolaris, Curvularia Phoma and Rhizoctonia were represented only in a single species. The results showed a variation in the fungal species and contamination percentage according to the detection and incubation methods. The lowest occurrence percentage was detected in lentils seeds with only one genus represented by Penicillium spp., while the highest occurrence percentage was detected with Chickpea seeds represent by six genera. The most common fungal genera displayed by agar plate method in wheat and chickpea were Rhizoctonia sp. (34.1%) and Penicillium sp. (42.5%) respectively. The highest fungal detection by blotter method was recorded with barley seeds while the lowest was chickpea seeds. The most common fungal genera recorded by blotter method under room temperature after 7 days and 14 days was Penicillium spp, with a percent 100% from chickpea seeds followed by Aspergillus spp with percent of (66.7%, 47.6%) from barley and raisin seeds respectively. While the highest frequent fungus in seeds incubated at 25°C after 7 and 14 days was Aspergillus parasiticus (88.9%) from mash seeds and Rhizopus sp. (80.1%) from lima bean respectively. Blotter method considered an efficient and economically reliable method.

Markedly increased quantities of lysozyme have been found in the serum and urine (ranging to 2.6 g per day) of ten consecutive cases of monocytic and monomyelocytic leukemia. The enzyme has been isolated from the urine of several cases and physicochemically and immunochemically characterized. It is apparently identical to the lysozyme of normal tears, saliva, leukocytes, and serum, but structurally different from the lysozyme of hen's egg white. The activity of the human enzyme assayed with M. lysodeikticus organisms is 3 to 12 times greater than egg white lysozyme at equivalent concentrations. An agar plate method has been developed for quantitating lysozyme activity in small samples (approximately 25 µl) of serum, urine, or other biological fluids. The range and reproducibility of this method were found to be superior to previously available lysozyme assay procedures. Present evidence indicates that lysozyme is the principal, if not the sole, product of the proliferating monocytes in monocytic and monomyelocytic leukemia, and quantitation of serum and urine lysozyme should be a useful diagnostic procedure for these leukemias.

In this study, quarternary ammonium salts based on triethylammonium and benzyltriethylammonium cations with the anion of chloride are successfully used for giant bamboo (Dendrocalamus Asper) preservation. These salts are new biocides as well as new salts which penetrate bamboo well. The prepared salts with hydrogen and benzyl substituent at the cation and consisted of chloride anion, exhibited fungicidal activity against Aspergillus Flavus. The effective and lethal doses were measured by the agar-plate method. In their activity against bamboo degrading fungi, salt of benzyl-triethyl-ammonium chloride was comparable with commercially available benzalkonium chloride and didecyldimethylammonium. In the process of grape wine fermentation, it has been long known that different flavor, aroma and taste characteristics are usually from various microbes associated grape strains and grape varieties. In the study, we were interested in understanding diversity of microbial niches of fermented glutinous rice ball developed in variety of fruit wine fermentation. Since different raw materials, fruits together with different microbial consortium associated with fruit surface itself affected various dominant culturable bacterial and fungal species. In the study, freshly prepared juices of fruits; mangoes and apples after processing without the pasteurization at 65°C for 30 min revealed dissimilar pH profiles and reducing sugar contents as well as alcohol production. Under microbiological examination as well as serial dilution agar plate technique, diverse dominant bacterial and fungal isolates were detected in the wine sample of the fruits pasteurized. The nutrients originated in apple sample caused more populations of microbes, including dominant bacilli, detected in the fermented broth since they were found in apples than in mangoes (104-1013 and 104-106 cells/ mL, respectively). From the results, it was shown that different nutrient sources played more important roles in stimulation of variations in microbial assort and possibly more complex in fermented juice qualification. The understanding in microflora consortium involving in wine fermentation for each fruit type should be helpful in monitoring and reflecting the concurrent microbial activity present in the large scale of wine production, reducing the risks of existing spoilage species.

Ten media, commonly used for the detection and isolation of streptococci of sanitary significance in water, dairy and other food products, were compared to establish a plating medium for the enumeration of enterococci in dairy products. To make all media suitable for comparison by the agar plate method, agar was added to those media which initially were recommended for use as broths. Criteria used in selecting the medium were high recovery, selectivity but not undue inhibition of enterococci and ease in obtaining and interpreting results. The recovery data of three different platings of enterococcus cultures were statistically analyzed. In this manner, one medium was eliminated on the basis of low recovery. Six of the remaining nine media were eliminated because they permitted the growth of non-enterococcus cultures. Two of the three media then remaining were eliminated because they allowed one S. bovis culture to grow. In addition, these media showed considerable variation in size and color of enterococcus colonies. The medium selected, the Citrate azide medium of Reinbold, Swern and Hussong (13), was modified by increasing the azide concentration. This did not result in undue inhibitory effects. It was further tested by obtaining recovery data for 158 known enterococcus cultures. High selectivity was demonstrated by showing that 408 colonial isolates from plates of raw milk, cheese and butter could be identified as enterococci.
