Abstract: The objective of this work was to evaluate the use of RAPD and ISSR molecular markers to determine the genetic variability among cashew (Anacardium spp.) genotypes, and to indicate possible promising crosses based on cashew genetic variability and phenotypic performance. Ten hybrids - derived from the crosses CCP 76 x BGC 589, CCP 76 x BRS 226, CCP 76 x HAC 276-1, CCP 76 x Embrapa 51, CCP 76 x BRS 253, CCP 76 x HAC222-4, and BRS226 x Embrapa 51 - and their parents were assessed at the molecular level. The hybrids were evaluated for nut yield, mean nut weight, bored nuts, and powdery mildew on nuts (scale 0-4). The RAPD and ISSR markers were efficient in the determinaton of the genetic variability among cashew genotypes, allowing of the grouping of 21 clusters. Associated with the phenotypic characterization of cashew nut for yield, weight, and health, the used markers can efficiently identify possible combinations with higher genetic variability and higher probability of developing transgressive genotypes in segregating populations.

Genetic diversity of the Greek plum germplasm collection was investigated using a combined RAPD and ISSR molecular markers approach. Twenty-six genotypes held at NAGREF-Naoussa were analyzed, producing in total 150 loci, of which 116 were polymorphic. Both techniques were highly informative and had a discrimination power greater than 0.9. RAPD and ISSR dendrograms were fairly correlated. The accessions were clustered according to ploidy and species. All Prunus domestica genotypes were grouped together and showed greater similarity to P. insititia and P. cerasifera genotypes compared to P. salicina, which was found genetically diverged. Bayesian structural analysis revealed significant admixture among genotypes. Greek varieties P. domestica ‘Goulina’ and ‘Asvestochoriou’ exhibited a distinctive genetic background, differentiating them from foreign varieties. This feature could make them attractive for breeding programs, since they can increase genetic diversity.

The Rheum species are important medicinal plants that are facing extinction due to their unplanned development and overexploitation by pharmaceutical industries. DNA polymorphisms are not prone to environmental modifications, thus they are widely used for the identification and characterization of plants. The use of different molecular markers has enabled the researchers for the valuation of genetic variability and diversity in its natural zone of distribution. The conventional approach may take several years to yield this information. For the estimation of molecular and genetic variations in geographical zone of distribution, various molecular markers technique are available like RAPD (Randomly Amplified Polymorphic DNA), RFLP (Restriction fragment length polymorphism), ISSR (Inter-Simple Sequence Repeats), SSR and AFLP. The uses of different molecular markers for the study of genetic diversity have been discussed in the review.
