Abamectin is a bio-insecticide, derived from the soil bacteria Streptomyces avermitilis. This insecticide is used in public health and agriculture to protect crops. Major adverse impacts of Abamectin are neurological symptoms acting on the peripheral nervous system. The aim of the current study is to reveal the toxic effects of Abamectin on chick embryo Ross 308 including morphological and histological changes and acetyl cholinesterase activity. 120 fresh fertilized eggs were divided into 6 groups; two of them were used as control. After 2 days of incubation, the eggs were injected with 100 µL of Abamectin solution (diluted at concentrations 360, 540, 900 and 1800 ppm) into the yolk sac. The Results showed that the mortality increased significantly in chicks treated with Abamectin, but had a lower weight in comparison to the control groups. Treated chicks started hatching at day 22 but were physically weak with drooping limbs, paralysis and then died after 24 hours of hatching. Some chicks did not normally hatch and needed assistance. They characterized by limb defects, failure retraction of yolk sac with bleeding. Histological examination of the liver showed hepatic cell degeneration, congestion in the central vein, infiltration of inflammatory cells and hepatocytes necrosis. Furthermore, the Acetyl cholinesterase enzyme analysis showed a significant decrease in the enzyme activity which leads to inhibition the activity of the body systems. It is concluded that low and high concentration of Abamectin has adverse impacts on chick embryo by changing some of morphological, histological characteristics and acetyl cholinesterase activity.

Cholinesterases in the chick liver. The role of the endoderm in the appearance of the activity of cholinesterases in the hepatic mesenchyme The histochemical method of Koelle & Friedenwald (1949), as modified by Gerebtzoff (1953), has been used to investigate the distribution of cholinesterases in the chick embryonic and adult liver. Non-specific cholinesterases and, in a lower proportion acetylcholinesterase, have been detected in the endothelial cells of blood sinusoids of both adult and embryonic hepatic tissue. The hepatocytes do not show any cholinesterase activity. Cholinesterases appear precociously in the liver mesenchyme, since they already occur in the septum transversum of the 3-day-old chick embryo. An experimental procedure preventing the invasion of the hepatic mesenchymal Anlage by the endodermic cords has been used. The experimentally isolated hepatic mesenchyme shows an important cholinesterase activity; therefore this activity does not depend on the presence of the hepatocytes. The grafting of the determined hepatic endodern in the somatopleura of the 3-day-old chick embryo results in the development of hepatic tissue in the body wall. In this experimentally produced liver, cholinesterase activities are present in the endothelial cells which have arisen from somatopleura mesenchymal cells, though normally somatopleural mesenchyme does not possess these enzymes. The role of the endoderm in the appearance of this enzymic activity in the somatopleural mesenchyme is discussed.

Otoconia are microscopic geometric structures that cover the sensory epithelia of the utricle and saccule (gravitational receptors) of mammals, and the lagena macula of birds. The importance of otoconia for maintanance of the body balance is evidenced by the abnormal behavior of species with genetic defects of otolith. Although a few reports have dealt with otoconia formation, some basic questions remain unanswered. The chick embryo is desirable for studying otoconial formation because its inner ear structures are easily accessible, and its gestational period is short (21 days of incubation).The results described here are part of an intensive study intended to examine the morphogenesis of the otoconia in the chick embryo (Gallus- domesticus) inner ear. We used chick embryos from the 4th day of incubation until hatching, and examined the specimens with light (LM) and transmission electron microscopy (TEM). The embryos were decapitated, and fixed by immersion with 3% cold glutaraldehyde. The ears and their parts were dissected out under the microscope; no decalcification was used. For LM, the ears were embedded in JB-4 plastic, cut serially at 5 micra and stained with 0.2% toluidine blue and 0.1% basic fuchsin in 25% alcohol.

The article presents the data on the study of the influence of mycotoxins combination (T-2 toxin at the concentration of 0.095 mg/kg and aflatoxin B1 in the concentration of 0.019 mg/kg) on the body of quails and the results of pharmacocorrection of toxicosis with a complex consisting of beet pulp and lecithin. Structural changes in the intestines of quais at fodder mycotoxicosis are described. The use of antitoxic feed additives in poultry led to a weakening of the action of xenobiotics, which was confirmed by an increase in the safety of poultry and increase in body weight of quails, a decrease in the clinical manifestations of intoxication, as well as in positive changes in the structure of the intestine of the poultry during histological examination.

<p><strong>Objective</strong>:<strong> </strong>To find out the effect of rotatory vestibular stimulation in cognition in rats through examining the behavioural patterns, the alterations in dendritic arborization and changes in AChE activity.</p><p><strong>Methods</strong>:<strong> </strong>Rotatory vestibular stimulation was provided in a rotatory vestibular apparatus at a rate of 50 rpm for 5 min, for 30 d for rats. 0.3 mg/kg of physostigmine also administered to rats of another group as a standard drug. No rotatory vestibular stimulation or physostigmine is provided to the control rats. Behavioural analysis, Neuromorphological and biochemical studies were done after vestibular stimulation.</p><p><strong>Results</strong>:<strong> </strong>No. of trails for acquisition and retention reduced significantly in treated rats when compared with the control rats. In all the treated rats the dendritic arborization increased significantly, and activity of AChE decreased significantly when compare with the control.</p><p><strong>Conclusion</strong>:<strong> </strong>Rotatory vestibular stimulation enhances learning and memory <em>via</em> increasing dendritic arborization and inhibiting acetyl-cholinesterase activity in rats. </p>

The present study was aimed to investigate the mechanism underlying the influence of maternal low-protein (LP) diet on offspring growth in the chicken. One hundred and twenty Chinese inbred Langshan breeder hens were allocated randomly into two groups fed diets containing low (10 %, LP) or normal (15 %) crude protein levels. Low dietary protein did not affect the body weight of hens, but significantly decreased the laying rate and egg weight. The yolk leptin content was significantly lower in eggs laid by LP hens, while no differences were detected for yolk contents of corticosterone, tri-iodothyronine (T3) or thyroxine. Despite significantly lower hatch weight, the LP offspring demonstrated obviously higher serum T3 concentration, which is in accordance with the faster post-hatch growth rate achieving significantly heavier body weight and pectoralis major muscle weight 4 weeks post-hatching. Expression of 20-hydroxysteroid dehydrogenase (20-HSD) mRNA in the yolk-sac membrane was significantly down-regulated at embryonic day 14, whereas that of transthyretin and leptin receptor (LepR) was not altered. Moreover, hypothalamic expression of 20-HSD, glucocorticoid receptors, thyrotropin-releasing hormone and LepR mRNA was significantly up-regulated in the LP group compared with their control counterparts. In the pectoralis major muscle, significantly higher expression of insulin-like growth factor (IGF)-I and IGF-I receptor mRNA was observed in LP embryos. The present study provides evidence that maternal LP diet programmes post-hatch growth of the offspring. The associated alterations in yolk leptin deposition as well as in yolk-sac membrane, fetal hypothalamus and muscle gene expression may be involved in mediating such programming effect in the chicken.

A significant acetylcholinesterase inhibitory activity was observed for the Ethanolic extract from the leaves of Calophyllum polyanthum by using TLC bioautographic method. Further bioassay-guided isolation of this extract using TLC bioautographic method resulted in obtaining a pyranochromanone, apetalic acid (1). The structure of 1 was identified by comparison of it’s spectral characteristics with previous reports. The concentration required for 50% inhibition of 1 was 0.95 mM, determined by a microplate assay. The anti-acetylcholinesterase aity of compound 1 was weak, but it was the first pyranochromanone which have anti-acetyl cholinesterase activity. As a new leading compound, it can be modified and transformed to obtain more potently active compounds.

The present work was aimed to identify phytochemicals in C. uredinicola methanol extract from qualitative, TLC and GC-MS method and evaluated for antioxidant, anti-HIV, anti-diabetes, anti-cholinesterase activity in vitro and in silico. The C. uredinicola extract showed flavonoids, tannins, alkaloids, glycosides, phenols, terpenoids, and coumarins presence in qualitative method. From GC-MS analysis, identified seven different phytochemicals and out of seven, four (coumarin, coumarilic acid, hymecromone, alloisoimperatorin) are coumarins. The C. uredinicola extract have shown significant antioxidant activity in DPPH (73) and FRAP (1359) method. The HIV-1 RT (83.81+2.14), gp 120 (80.24+2.31), integrase (79.43+3.14) and protease (77.63+2.14), DPPIV, β-glucosidase and acetyl cholinesterase activity was significantly reduced by the extract. The 2-diphenylmethyleneamino methyl ester had shown significant interaction with oxidant and HIV-1 proteins whereas alloisoimperatorin have interacted with diabetes and cholinesterase proteins followed by hymecromone with high binding energy. These three phytochemicals are non-carcinogens, non-toxic, readily degradable and have drug likeliness properties. The C. uredinicola phytochemicals are responsible for management of diabetes, HIV-1 and Alzheimer. Further in vivo work is needed to justify our research.
