Type 2 diabetes mellitus (T2DM) belongs to the diseases with hereditary predisposition, so both environmental and genetic factors contribute to its development. Recent studies have demonstrated that the skeleton realizes systemic regulation of energy metabolism through the secretion of osteocalcin (OCN). Thus, the association analysis between HindIII single nucleotide polymorphism of OCN gene (BGLAP) promoter region and T2DM development in Ukrainian population was carried out. 153 individuals diagnosed with T2DM and 311 control individuals were enrolled in the study. The genotyping was performed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The lack of association between BGLAP HindIII single nucleotide polymorphism (SNP) and T2DM development among Ukrainians was found. Further studies with extended groups of comparison are needed to confirm the obtained results.

ABSTRACT Background The literature suggests that periodontal disease and diabetes mellitus share a two-way relationship. The aim of this study was to evaluate and compare the levels of adrenomedullin (ADM) in gingival crevicular fluid (GCF) of periodontally healthy and periodontitis patients with or without type 2 diabetes with different glycemic controls. Methods Ninety patients were included in the study and were divided into five groups based on CPI scores and ADA classification of diabetes. Probing pocket depth (PPD) and clinical attachment level (CAL) were measured in all the subjects. GCF was collected from all the participants using micropipettes. ADM levels were measured in GCF samples by enzyme-linked immunosorbent assay. Results The results showed higher levels of ADM in patients with periodontitis as compared to healthy group. Significant correlation was present between PPD and CAL and ADM levels in all periodontitis patients with or without type 2 diabetes. Conclusion Increase in GCF levels of ADM from periodontal health to disease and in periodontitis patients with type 2 diabetes with the worsening of glycemic control underlines the possible role of ADM in mounting a protective response to worsening disease state. Type 2 diabetes mellitus (T2DM) is one of the threatening disorders in the world. It affects people of all ages. Type 2 diabetes mellitus is a condition in which the glucose level in the blood is elevated due to improper function of the secretion of insulin from beta cells of the pancreas. It is a multifactorial disease because it is caused by both environmental and hereditary factors. One of the genes which play an important role in type 2 diabetes mellitus is SLC30A8 which encodes for zinc transporter ZnT8. The common polymorphic site for SLC30A8 is rs13266634. This single-nucleotide polymorphism leads to type 2 diabetes mellitus by replacing the arginine residue with tryptophan residue. This review mainly focuses on the polymorphic studies in the gene SLC30A8 and its association with type 2 diabetes mellitus.
