AbstractAuxin affects the growth and development of in vitro plantlets including root induction. An experiment was conducted to determine the combination and concentration of auxin for rooting of oil palm plantlets in liquid medium.Unrooted plantlets of oil palm MK 649 clone with height 6 – 7 cm and 2 – 3 leaves were used as material source. The plantlets were cultured in de Fossard liquid medium. The treatments used were combinations of NAA and IBA at 0, 5,10 and 20 μM. The results show that 10 μM NAA combined with 20 μM IBA gave the highest percentage of rooting of oil palm plantlets (73.3%) in 10 weeks. NAA and IBA concentration influenced significantly rooting percentageand root quality and there was a significant interaction between the two auxins. Root initiation response of oil palm plantlets to NAA was higher than to IBA. The best of oil palm root class which indicates root quality was obtained in a medium with 10 μM NAA + 20 μM IBA. The aerial parts of the plantlets grew well in term of shoot height, leaf number and shoot diameter especially in a medium with 10 μM NAA + 20 μM IBA. AbstrakAuksin berpengaruh terhadap pertumbuhan dan perkembangan planlet in vitro, termasuk terhadap induksi akar. Penelitian ini bertujuan untuk menentukan kombinasi dan konsentrasi auksin yang tepat dalam pembentukan akarplanlet kelapa sawit in vitro dalam medium cair. Bahan yang digunakan berupa planlet kelapa sawit klon MK 649 tanpa akar dengan tinggi 6 – 7 cm dan jumlah daun 2 – 3 helai. Planlet dikulturkan dalam medium de Fossard cair. Perlakuan yang digunakan adalah kombinasi NAA dan IBA dengan konsentrasi 0, 5, 10 dan 20 μM. Hasil penelitian menunjukkan bahwa perlakuan NAA 10 μM dikombinasikan dengan IBA 20 μM menghasilkan persentase pembentukan akar planlet kelapa sawit tertinggi yaitu 73,3% dalam waktu 10 minggu. Konsentrasi NAA dan IBA secara nyata mempengaruhi persentase pembentukan dan kualitas akar serta terdapat interaksi yang nyata antara kedua perlakuan auksin. Respons induksi akar kelapa sawit terhadap NAA lebih tinggi daripada IBA. Kelas akar planlet kelapa sawit terbaik yang menunjukkan kualitas perakaran, juga diperoleh pada NAA 10 μM dan IBA 20 μM. Pertumbuhan dan perkembangan organ bagian atas yang meliputi tinggi tunas, jumlah daun dan diameter tunas menunjukkan peningkatan yang cukup baik terutama pada perlakuan NAA 10 μM + IBA 20 μM.

Oil palm (Elaeis guineensis Jacq.) leaves (OPL) are widely available at zero cost in Southeast Asia countries, especially in Malaysia and Indonesia due to large scale oil palm plantations. OPLs contain a large amount of flavonoids in particular flavonoid C-glycosides, which are known to possess useful biological properties including antioxidant and wound healing properties. The present study aimed at evaluating the wound healing efficacy of OPL in various solvent extracts and flavonoid enriched fractions and to determine the contribution of flavonoid C-glycosides (orientin, isoorientin, vitexin and isovitexin) using in-vitro scratch assay on 3T3 fibroblast cells. Solvent crude extracts with different polarity were screened and the most active extract was subjected to acid hydrolysis. The crude and acid hydrolysed extracts were further enriched using macroporous resins, XAD7HP. UHPLC-UV/PDA and LC-MS/MS analysis were applied for identification and confirmation of flavonoid C-glycosides. The wound healing properties comprised of cell viability, cell proliferation and cell migration were studied. Allantoin was used as a positive control to compare the efficacy among the tested samples. The results revealed all OPL crude extracts, flavonoid enriched fractions and flavonoid C-glycosides were non-toxic at concentrations below 25 µg/mL and showed better cell proliferation and migration activities at low concentrations than higher concentrations.. This study also demonstrated orientin, isoorientin, vitexin and isovitexin presented in OPL extracts and flavonoid enriched fractions stimulated proliferation and migration of 3T3 fibroblast cells. Hence, these findings may pose potential therapeutic bioactive agents for wound healing by enhancing fibroblast proliferation and migration.

ABSTRACT Large-scale oil palm propagation (Elaeis guineensis Jacq.) is difficult due to its unique apical meristem. In this context, micropropagation allows the multiplication of seedlings in vitro and the storage of germplasm elites. This study aimed to induce embryogenic calluses from leaves of oil palm plants in low concentrations of auxins and to observe the maintenance of these characteristics during in vitro cultivation. Calluses were induced in 0.5 cm leaf explants in Y3 culture medium supplemented with Picloram (4-Amino-3,5,6-trichloro-2-pyridinecarboxylic acid) or 2,4-D (2,4-dichlorophenoxyacetic acid), at concentrations of 0, 1, 3, 6, and 9 mg L-1. The callus with embryogenic appearance was subcultured and evaluated regarding maintenance of embryogenic characteristics by cytochemical analyses. The best treatment for induction of calluses was composed of 1mg.L-1 of Picloram, which led to 30% callus formation. The calluses were classified into4 types, based on color and morphology. The cells of calluses with nodular and beige appearance have embryogenic characteristics, and the embryogenic potential of the cell masses was maintained over the 20 months of cultivation. This differentiated adaptation to the protocol can allow the advance in the mass propagation of oil palm through tissue culture, indicating the importance of investigating the topics proposed by the research.

This study reported in vitro embryogenesis of oil palm using young leaves as explants. Explants were grown in solid modified MS or Eeuwens medium containing different concentrations of NAA and 2,4-D, i.e. media C1, C2, C3, C4 and C5, M1, M2, M3 and M4, to induce embryogenic calli. Compact and pearly-white, globular calli were obtained from the youngest leaf explants 28 weeks after culture.C1 media (MS medium + 107.41 µM of NAA + 100 mg.L-1 of asparagine + 100 mg.L of glutamine-1) produced the highest percentage of calli formation (30.56%), whereas C4 media (C1 supplemented with 67.86 µM of 2,-D ) was the optimal media for embryogenic callus induction. Direct embryoids were obtained from slightly older leaf explants on the C3 media containing NAA after 36 weeks of culture. However, four subcultures using the same medium with gradual reduction of auxin concentration were not successful to develop embryogenic callus and embryoid cells during the course of this study.
