Evaluating the Antifungal Activity of Some Traditional Medicinal Plant Extracts against Alternaria solani (Tomato Early Blight Pathogen) in Ethiopia

Background: Tomato (Lycopersicon esculentum Mill.) belongs to the family Solanaceae. In Ethiopia, control of early blight is largely dependent on fungicidal application. There is a research need to identify effective botanical extracts to control Alternaria solani that cause early blight of tomato and for evaluation of plant extracts through different solvents on the target pathogen. Methods: In vitro experiment was conducted to evaluate the effectiveness of crude extracts of 16 selected medicinal plants against Alternaria solani. Thus, crude extracts were extracted from medicinal plants with different solvents (methanol, ethanol and petroleum at (25%, 50% and 100%) concentrations. The Alternaria solani was isolated from infected tomato leaves showing early blight symptoms. Evaluation of plant extracts was carried out against Alternaria solani using food poisoned technique on PDA. Result: Results showed that most of the methanolic extract plants were showed significant inhibition of the mycelial growth as compared to ethanolic and petroleum ether extracts. A higher rate of mycelial reduction was recorded by ethanol extracts of Allium sativum at all concentrations (100%) followed by methanol extracts of Allium sativum at 25%, 50%, 100% concentration (90.02%, 97.01%, 100% respectively). The effectiveness of extracts against Alternaria solani depends on use at the higher concentrations and various solvents. For crude extracts that have shown higher inhibitory effects against Alternaria solani in vitro conditions, actual chemical compounds should be identified. Furthermore, it is also important to evaluate these plants on other microbes, study to test in vivo and to assess their real potential field condition wherever early blight is an important disease of tomato.

Extraction, Purification and In Vitro Antioxidant Activity Evaluation of Phenolic Compounds in California Olive Pomace

Olive pomace (OP) is a valuable food byproduct that contains natural phenolic compounds with health benefits related to their antioxidant activities. Few investigations have been conducted on OP from the United States while many studies on European OP have been reported. OP of Arbequina, the most common cultivar from California, was collected and extracted by water, 70% methanol and 70% ethanol, followed by purification using macroporous absorbing resin. Results showed that the extractable total phenolic content (TPC) was 36–43 mg gallic acid equivalents (GAE)/g in pitted, drum-dried defatted olive pomace (DOP), with major contributions from hydroxytyrosol, oleuropein, rutin, verbascoside, 4-hydroxyphenyl acetic acid, hydroxytyrosol-glucoside and tyrosol-glucoside. Macroporous resin purification increased TPC by 4.6 times the ethanol crude extracts of DOP, while removing 37.33% total sugar. The antioxidant activities increased 3.7 times Trolox equivalents (TrE) by DPPH and 4.7 times TrE by ferric reducing antioxidant power (FRAP) in the resin purified extracts compared to the ethanol crude extracts. This study provided a new understanding of the extraction of the bioactive compounds from OP which could lead to practical applications as natural antioxidants, preservatives and antimicrobials in clean-label foods in the US.

First report of fatty acids in Mimosadiplotricha bee pollen with in vitro lipase inhibitory activity

Bee pollen (BP) is full of nutrients and phytochemicals, and so it is widely used as a health food and alternative medicine. Its composition and bioactivity mainly depend on the floral pollens. In this work, BP collected by Apis mellifera with different monoculture flowering crops (BP1-6) were used. The types of floral pollen in each BP were initially identified by morphology, and subsequently confirmed using molecular phylogenetic analysis. Data from both approaches were consistent and revealed each BP to be monofloral and derived from the flowers of Camellia sinensis L., Helianthus annuus L., Mimosa diplotricha, Nelumbo nucifera, Xyris complanata, and Ageratum conyzoides for BP1 to BP6, respectively. The crude extracts of all six BPs were prepared by sequential partition with methanol, dichloromethane (DCM), and hexane. The crude extracts were then tested for the in vitro (i) α-amylase inhibitory, (ii) acetylcholinesterase inhibitory (AChEI), and (iii) porcine pancreatic lipase inhibitory (PPLI) activities in terms of the percentage enzyme inhibition and half maximum inhibitory concentration (IC50). The DCM partitioned extract of X. complanata BP (DCMXBP) had the highest active α-amylase inhibitory activity with an IC50 value of 1,792.48 ± 50.56 µg/mL. The DCM partitioned extracts of C. sinensis L. BP (DCMCBP) and M. diplotricha BP (DCMMBP) had the highest PPLI activities with an IC50 value of 458.5 ± 13.4 and 500.8 ± 24.8 µg/mL, respectively), while no crude extract showed any marked AChEI activity. Here, the in vitro PPLI activity was focused on. Unlike C. sinensis L. BP, there has been no previous report of M. diplotricha BP having PPLI activity. Hence, DCMMBP was further fractionated by silica gel 60 column chromatography, pooling fractions with the same thin layer chromatography profile. The pooled fraction of DCMMBP2-1 was found to be the most active (IC50 of 52.6 ± 3.5 µg/mL), while nuclear magnetic resonance analysis revealed the presence of unsaturated free fatty acids. Gas chromatography with flame-ionization detection analysis revealed the major fatty acids included one saturated acid (palmitic acid) and two polyunsaturated acids (linoleic and linolenic acids). In contrast, the pooled fraction of DCMMBP2-2 was inactive but pure, and was identified as naringenin, which has previously been reported to be present in M. pigra L. Thus, it can be concluded that naringenin was compound marker for Mimosa BP. The fatty acids in BP are nutritional and pose potent PPLI activity.

Bioactivity Evaluation of Leucas zeylanica and its Phytochemicals

Leucas zeylanica belongs to the Lamiaceae family has high medicinal value. The phytochemicals screening results showed that terpenoids, flavonoids, tannins, saponins, phenols and alkaloids are present in aerial part of L. zeylanica.Vacuum liquid chromatography (VLC) and column chromatography (CC) techniques were used in fractionation and purification of the crude extracts.

Enhancement of Selected Health Benefits in Fermented Cow and Soy Milk Supplemented with Water Soluble Curcumin

Different formulates of fermented cow and soy milk by Lactobacillus plantarum EMCC 1027 with 50, 100, and 150 mg/100 mL curcumin were investigated for antibacterial, antioxidant, anti-colonic cancer, and anti-inflammation activities. Also, the viability of L. plantarum was monitored during cold storage period. Our results showed that values of antibacterial, antioxidant, anti-colonic cancer, and anti-inflammation activities in crude extracts of fermented soy milk were significantly increased in comparison with crude extracts of fermented cow milk. The addition of different concentrations (100 and 150 mg/100 mL) of curcumin had a significant enhancement effect for all selected health benefits properties. The increase in antioxidant capacity of different crude extracts was in a good correlation with their polyphenols content. Addition of water soluble curcumin did not have any adverse effect on the viability of L. plantarum during fifteen days of cold storage. Therefore, the synergistic effect between fermented cow/soy milk and water soluble curcumin could be recommended. Indeed, extensive research is still needed in order to investigate the molecular mechanisms of health a benefit of different formulates of fermented cow/soy milk and water soluble curcumin.

The Changes of Blood Platelet Reactivity in the Presence of Crude Extracts Isolated from Leaves and Twigs of Elaeagnus Rhamnoides (L.) A. Nelson Measuring in Whole Blood

Uncontrolled blood platelet activation is an important risk factor of cardiovascular disease (CVDs). Various studies on phenolic compounds indicate that they have a protective effect on the cardiovascular system through different mechanisms, including the reduction of blood platelet activation. One of the plants that is particularly rich in phenolic compounds is sea buckthorn (Elaeagnus rhamnoides (L.) A. Nelson). The aim of the present study in vitro was to determine the anti-platelet properties of crude extracts isolated from leaves and twigs of E. rhamnoides (L.) A. Nelson in whole blood using flow cytometric and total thrombus-formation analysis system (T-TAS). The aim of our study was also analyze of blood platelet proteoms in the presence of different sea buckthorn extracts. A significant new finding is a decrease surface expression of P-selectin on blood platelets stimulated by 10 µM ADP and 10 µg/mL collagen, and a decrease surface expression of GPIIb/IIIa active complex on non-activated platelets and platelets stimulated by 10 µM ADP and 10 µg/mL collagen in the presence of sea buckthorn leaf extract (especially at the concentration 50 µg/mL). The twig extract also displayed antiplatelet potential. However, this activity was higher in the leaf extract than in the twig extract in whole blood. In addition, our present findings clearly demonstrate that investigated plant extracts have anticoagulant properties (measured by T-TAS). Therefore, the two tested extracts may be promising candidates for the natural anti-platelet and anticoagulant supplements.

The effect of lichen secondary metabolites on Aspergillus fungi

A systematic review of literature data on the antifungal potential of extracted lichen compounds and individual secondary metabolites against mold species of the genus Aspergillus is provided. Crude extracts from 49 epiphytic, 16 epigeic and 22 epilithic species of lichens and 44 secondary metabolites against 10 species, Aspergillus candidus, A. flavus, A. fumigatus, A. nidulans, A. niger, A. ochraceus, A. parasiticus, A. restrictus, A. stellatus and A. ustus, were analysed. Several measuring techniques were employed for such analyses. Lichen substances were extracted with alcoholic and other organic solvents mainly using the Soxhlet apparatus. Among the three most-studied mold species, the results showed that the crude extracts from the thalli of the lichens Cladonia foliacea, Hypotrachyna cirrhata, Leucodermia leucomelos, Platismatia glauca and Pseudevernia furfuracea against Aspergillus flavus, from C. foliacea, Nephroma arcticum and Parmelia sulcata against A. fumigatus and from Evernia prunastri, Hypogymnia physodes, Umbilicaria cylindrica and Variospora dolomiticola against A. niger have the greatest antifungal potential. The lichen secondary metabolites showed a higher inhibitory potential, e.g. protolichesterinic acid against A. flavus, lecanoric acid against A. fumigatus and orsellinic acid against A. niger; the other seven species of Aspergillus have been poorly studied and require further investigation. A comparison of the inhibitory potential of the tested mixtures of lichen substances and their secondary metabolites shows that they can compete with commonly used antifungal substances, such as ketoconazole and clotrimazole against A. flavus, A. nidulans, A. niger and A. parasiticus and fluconazole in the case of A. fumigatus.

Antibaterial activity of the extracts of pineapple and pomelo against five different pathogenic bacterial isolates

To combat the infections caused by antibiotic resistant bacteria, natural candidates are being studied to find out antibacterial activity against the drug-resistant microorganisms. Among the variety of natural candidates of plant origin, many fruits have been proved to have potent antibacterial activity. In the current study, we chose pineapple (Ananas comosus), and pomelo (Citrus maxima) to determine their efficacy against some clinical isolates. Fruit samples were subjected to prepare crude, ethanol, methanol and aqueous extract to determine their antibacterial potency. Clinical isolates were used to determine the antibacterial activity of the extracts against them. The isolates were found to be multi-drug resistant. Out of twenty-eight antibiotics, Pseudomonas aeruginosa was resistant to ten antibiotics and Salmonella spp. was resistant to nine antibiotics. Rather than the crude extracts of the fruits, ethanol and methanol extracts showed antibacterial activity towards multi-drug resistant pathogenic bacteria. Aqueous extract did not show any significant antibacterial activity at all. Extracts of pomelo fruit exhibited the highest results whereas pomelo skin and pineapple peel crude extracts were the least effective compared to the other extracts. Ethanol extract of pineapple fruit (against all isolates but Staphylococcus aureus) and methanol extract of pomelo fruit (against all isolates) showed the lowest MIC (minimum inhibitory concentration) of 187.5 μg/ml. MBC (minimum bactericidal concentration) was found (within the range of 500 μg/ml to 1000 μg/ml) only with ethanol and methanol extracts of pomelo and pineapple. As the clinical isolates were found to be inhibited by the extracts, they can be used as an alternative for treating infections caused by these bacteria. Comparative Hypolipidemic Evaluation of Aframomum Melegueta Seeds and Moringa Oleifera Leaves

Aims: The study was carried out to compare the lipid lowering effects of both crude drugs as well as deducing the extracts with the best lipid lowering property; and the fractions. Study Design: The research was conducted with an experimental design solely based on laboratory trials which involved the use of ninety-six (96) male albino wistar rats to compare the hypolipidemic effects of both crude drugs and respective fractions. Place and Duration of Study: Department of Pharmacognosy Laboratory, Faculty of Pharmacy, Delta State University, Abraka, Nigeria and Department of Pharmacology and Toxicology Laboratory, Faculty of Pharmaceutical Sciences, Nnamdi Azikiwe University, Agulu, Anambra State, Nigeria. The research was carried out from March to August, 2021. Methodology: Ethanol extracts of both plants were prepared using soxhlet extraction. Each extract was then subjected to VLC fractionation using four solvents: n-hexane, chloroform, ethyl acetate and methanol. The fractions were bulked together after conducting thin layer chromatographic procedures and each extract was bulked into four fractions. The acute toxicity studies (LD50) of both extracts were determined in the rats using Lorke’s method. The crude extracts were screened for the presence and quantity of phytoconstituents using standard methods. The antilipidemic study was carried out using sixty-eight (68) rats randomized into seventeen (17) groups of four (4) animals each. Lipid profile was determined using spectrophotometer. Liver function tests and histology was also carried out using standard procedures. Results: Administration of various treatments (both crude extracts and fractions) evoked a significant (p<0.05) reduction of TC, TG, and LDL-C as well as significant (p<0.05) elevation of HDL-C when compared with the negative control. With a percentage serum lipid reduction of 45.11%TC, 48.23%TG, 63.39% LDL-C and 174.69% elevation of HDL-C, the group treated with the combination of 500 mg/kg Aframomum melegueta and 500 mg/kg Moringa oleifera produced the best hypolipidemic effect. This is closely followed by fraction MO4. Comparatively, Moringa oleifera extracts exerts a better antilipidemic effect than Aframomum melegueta seed extract. The liver function test showed that both plants has no toxic effect on the liver cells at doses of 250 mg/kg and 500 mg/kg, hence confirming the hepatoprotective effect of both crude drugs at the doses administered. Conclusion: In conclusion, results from this study suggests that ethanol extract of Moringa oleifera leaves is more effective than ethanol extract of Aframomum melegueta seeds as a hypolipidemic agent, however, combination of both crude drugs as a lipid lowering agent has proved to be more effective and reliable when compared to each crude drug administered independently.
