A vertebrate disk membrane is composed of 40 % lipids and 60 % proteins. Its fracture faces have been classed into the plasmic (PF) and exoplasmic faces (EF), complementary with each other, like those of most other types of cell membranes. The hypothesis assuming the PF particles as representing membrane-associated proteins has been challenged by serious questions if they in fact emerge from the crystalline formation or decoration effects during freezing and shadowing processes. This problem seems to be yet unanswered, despite the remarkable case of the purple membrane of Halobacterium, partly because most observations have been made on the replicas from a single face of specimen, and partly because, in the case of photoreceptor membranes, the conformation of a rhodopsin and its relatives remains yet uncertain. The former defect seems to be partially fulfilled with complementary replica methods.

Abstract Background Immune checkpoint inhibitors (ICIs) are a novel class of oncological agents which are used to treat a number of malignancies. To date seven agents have been approved by the Food and Drug Administration (FDA) to treat both solid and haematological malignancies. Despite their efficacy they have been associated with a number of endocrinopathies. We report a unique case of hypophysitis, thyroiditis, severe hypercalcaemia and pancreatitis following combined ICI therapy. Case presentation A 46-year old Caucasian female with a background history of malignant melanoma and lung metastases presented to the emergency department with lethargy, nausea, palpitations and tremors. She had been started on a combination of nivolumab and ipilimumab 24 weeks earlier. Initial investigations revealed thyrotoxicosis with a thyroid stimulating hormone (TSH) of < 0.01 (0.38–5.33) mIU/L, free T4 of 66.9 (7–16) pmol/.L. TSH receptor and thyroperoxidase antibodies were negative. She was diagnosed with thyroiditis and treated with a beta blocker. Six weeks later she represented with polyuria and polydipsia. A corrected calcium of 3.54 (2.2–2.5) mmol/l and parathyroid hormone (PTH) of 9 (10–65) pg/ml confirmed a diagnosis of non-PTH mediated hypercalcaemia. PTH-related peptide and 1, 25-dihydroxycholecalciferol levels were within the normal range. Cross-sectional imaging and a bone scan out ruled bone metastases but did reveal an incidental finding of acute pancreatitis – both glucose and amylase levels were normal. The patient was treated with intravenous hydration and zoledronic acid. Assessment of the hypothalamic-pituitary-adrenal (HPA) axis uncovered adrenocorticotrophic hormone (ACTH) deficiency with a morning cortisol of 17 nmol/L. A pituitary Magnetic Resonance Image (MRI) was unremarkable. Given her excellent response to ICI therapy she remained on ipilimumab and nivolumab. On follow-up this patient’s thyrotoxicosis had resolved without anti-thyroid mediations – consistent with a diagnosis of thyroiditis secondary to nivolumab use. Calcium levels normalised rapidly and remained normal. ACTH deficiency persisted, and she is maintained on oral prednisolone. Conclusion This is a remarkable case in which ACTH deficiency due to hypophysitis; thyroiditis; hypercalcaemia and pancreatitis developed in the same patient on ipilimumab and nivolumab combination therapy. We postulate that hypercalcaemia in this case was secondary to a combination of hyperthyroidism and secondary adrenal insufficiency.

Abstract Three-dimensional (3D) digital technology is an essential conservation method that complements the traditional restoration technique of cultural artifacts. In this study, 3D scanning, virtual restoration modeling, and 3D printing were used as a noncontact approach for restoring a damaged stone-seated Bodhisattva (stone Buddha statue). First, a 3D model with an average point density of 0.2 mm was created by integrating the fixed high-precision scanning of the exterior and the handheld mid-precision scanning of the interior excavated hole. Using a 3D deterioration map of the stone Buddha statue, the area of the missing parts was measured to be 400.1 cm2 (5.5% of the total area). Moreover, 257.1 cm2 (64.2% of the missing part area) of four parts, including the head, surrounding area of the Baekho, right ear, and right eye, for which symmetry was applicable for modeling or there could be ascertainable historical evidence for the total missing parts, was selected for restoration. The virtual restoration of the missing parts of the stone Buddha statue was performed using a haptic modeling system in the following order. First, the location of the three fragments detached from the head was determined. Next, a reference model was selected, and its symmetrization and modification with respect to the original model were conducted. Further, estimation modeling and outer shape description were achieved through historical research and consultation with experts. The heuristic-based assembly suitability of the created virtual restoration model (461 cm3) was verified by design mockup printing and digital–analog simulation. In particular, to address assembly interference, the interface surface was modified and reprocessed several times. Accordingly, the volume of the final design mockup decreased by 5.2% (437 cm3). Photopolymerization 3D printing technology was used for the actual restoration of the stone Buddha statue, and considering the surface roughness, the layer thickness of the material used for restoration was set at 0.10 mm. Finally, the surface of the printed output was colored to prevent yellowing and joined to the missing parts of the stone Buddha statue. This study presents a remarkable case of shifting from the traditional manual-contact method to the contactless digital method for restoring artifacts and is expected to largely contribute to increasing the usability of digital technologies in the restoration of cultural artifacts.

In this paper, we investigate some mathematical structures underlying the physics of linear optical passive (LOP) devices. We show, in particular, that with the class of LOP transformations on N optical modes one can associate a unitary representation of U (N) in the N-mode Fock space, representation which can be decomposed into irreducible sub-representations living in the subspaces characterized by a fixed number of photons. These (sub-)representations can be classified using the theory of representations of semi-simple Lie algebras. The remarkable case where N = 3 is studied in detail.
