Oral route has been the most convenient and accepted  route of drug delivery. Owing to tremendous curative  benefits of the oral controlled release dosage forms are  being preferred as the interesting topic in pharmaceutical  field to achieved improved therapeutics advantages.  Gastro retentive drug delivery system is novel drug  delivery systems which has an upper hand owing to its  ability of prolonged retaining ability in the stomach and  thereby increase gastric residence time of drugs and also  improves bioavailability of drugs. Concept of novel drug  delivery system arose to overcome the certain aspect  related to physicochemical properties of drug molecule  and the related formulations. In this context, various  gastro retentive drug delivery systems have been used to  improve the therapeutic efficacy of drugs that have a  narrow absorption window, are unstable at alkaline pH,  are soluble in acidic conditions, and are active locally in  the stomach. Concept of novel drug delivery system  arose to overcome the certain aspect related to  physicochemical properties of drug molecule and the  related formulations. Various approaches are currently  used including gastro retentive floating drug delivery  systems, swelling and expanding system, polymeric bio  adhesive systems, modified shape systems, high density  system and other delayed gastric emptying devices.  Moreover, future perspectives on this technology are  discussed to minimize the gastric emptying rate in both  the fasted and fed states. The present review briefly  addresses the physiology of the gastric emptying  process with respect to floating drug delivery systems.  The purpose of this review is to bring together the recent  literature with respect to the method of preparation, and  various parameters affecting the performance and  characterization of floating microspheres. Attempt has  been made to summarize important factors controlling  gastro retentive drug delivery systems. Overall, this  review may inform and guide formulation scientists in  designing the gastro retentive drug delivery system.

The purpose of writing this review on floating drug delivery systems (FDDS) was to compile the recent literature with special focus on the principal mechanism of floatation to achieve gastric retention . This can be achieved by use of various polymeric substances including natural polymers. These polymers are inexpensive, safe and available in a variety of structures with versatile characteristics. Large number of derivatizable groups, wide range of molecular weights, varying chemical composition gel forming nature of these polymers also provide an exciting opportunities in the fascinating arena of applied polymer science and drug delivery technology. All these characteristics make them suitable candidate for design and fabrication of novel gastroretentive drug delivery systems. Various natural polymers have been investigated worldwide by scientific community for their potential as floating drug delivery systems. The present article highlights various recent efforts and advanced approaches exploiting several natural polymers in this technology. The purpose of writing this review on floating drug delivery systems (FDDS) was to compile the recent literature with special focus on the principal mechanism of floatation to achieve gastric retention. It is known that differences in gastric physiology (such as, gastric pH, motility) exhibit both intra- as well as inter-subject variability demonstrating significant impact on gastric retention time and drug delivery behaviour. The recent developments of FDDS including the physiological and formulation variables affecting gastric retention, approaches to design single-unit and multiple-unit floating systems, and their classification and formulation aspects are covered in detail. Conventional chemotherapeutics lack the specificity and controllability, thus may poison healthy cells while attempting to kill cancerous ones. Newly developed nano-drug delivery systems have shown promise in delivering anti-tumor agents with enhanced stability, durability and overall performance; especially when used along with targeting and triggering techniques. This work traces back the history of chemotherapy, addressing the main challenges that have encouraged the medical researchers to seek a sanctuary in nanotechnological-based drug delivery systems that are grafted with appropriate targeting techniques and drug release mechanisms. A special focus will be directed to acoustically triggered liposomes encapsulating doxorubicin.
