Mercury is one of the detrimental toxicants that can be found in the environment and exists naturally in different forms; inorganic and organic. Human exposure to inorganic mercury, such as mercury chloride, occurs through air pollution, absorption of food or water, and personal care products. This study aimed to investigate the effect of HgCl2 on cell viability, cell cycle, apoptotic pathway, and alters of the transcriptome profiles in human non-small cell lung cancer cells, H1299. Our data show that HgCl2 treatment causes inhibition of cell growth via cell cycle arrest at G0/G1- and S-phase. In addition, HgCl2 induces apoptotic cell death through the caspase-3-independent pathway. Comprehensive transcriptome analysis using RNA-seq indicated that cellular nitrogen compound metabolic process, cellular metabolism, and translation for biological processes-related gene sets were significantly up- and downregulated by HgCl2 treatment. Interestingly, comparative gene expression patterns by RNA-seq indicated that mitochondrial ribosomal proteins were markedly altered by low-dose of HgCl2 treatment. Altogether, these data show that HgCl2 induces apoptotic cell death through the dysfunction of mitochondria.

Current scenario of newer diseases with multiple causes has drawn the attention of the researchers in the field of therapeutics and they are now inclined to identify molecules effective for targeted therapy. Objective: Quinoline (1-azanaphthalene); belongs to heterocyclic aromatic nitrogen compound. Some quinoline-based derivatives are also known for their anti-tumor activity. The study was planned to evaluate the cytotoxic potential of quinoline derivatives. Methods: Berberine; a quinoline compound was made part of study to make structural analogs which were docked against potential target proteins. Cytotoxic profiling of all derivatives was done using MTT cytotoxicity assay. Results: The pharmacoinformatic and structure activity relationship studies of analogs were done. The cytotoxic profiles were elucidated by comparing viability rates of analogs treated hepatic cancerous cell line with untreated hepatic cells and untreated mesenchymal stem cells as standards. Marked cytotoxicity was seen in all molecules at low doses than reported in past studies with relevance to parent compound. Conclusions: The results will be further confirmed through various other cell culture assays targeting different marker proteins, pharmacoinformatics tools and structure activity relationship studies.

Abstract This study focused on evaluating different support media for COD and nitrogen compound removal from an Upflow Anaerobic Sludge Blanket (UASB) reactor fed with swine wastewater. Maximum specific nitrification (MSNA) and denitrification (MSDA) activity tests were performed in two fixed-film systems with (1) polyurethane foam (R1) and (2) polyethylene rings (R2). The results showed that the R2 system performed more efficiently than R1, reaching organic matter removal of 77 ± 8% and nitrogen of 98 ± 4%, attributed to higher specific denitrifying activity recorded (5.3 ± 0.34 g NO3--N/g VTS ∙h). In this sense, MSDA tests indicated that the suspended biomass was responsible for at least 70% of nitrogen removal in the form of ammonium compared with 20% attributed to biomass in the form of biofilm. On the other hand, 40 ± 5% of initial nitrogen could not be quantified in the system effluents, but 10 ± 1% was attributed to loss by volatilization. According to the analyses, the previous information infers the development of simultaneous nitrification-denitrification (SND) routes. Respect to the analyses of microbial diversity and abundance in the biofilm of R2 rings, the presence of the genus Pseudomonas dominated the prokaryotic community of the system in 54.4%.
