Programas de gerenciamento de rebanhos leiteiros têm utilizado o escore de células somáticas (ECS) para registrar os valores médios da contagem de células somáticas (CCS). Trata-se da conversão da CCS em dez categorias, sendo utilizado desde 1983, auxiliando a avaliação da qualidade do leite, permitindo mensurar a prevalência da mastite no rebanho. A presente pesquisa avaliou o comportamento do ECS em amostras de leite de tanques provenientes de indústrias de laticínios do Estado do Paraná, analisadas pelo Laboratório de Análise da Qualidade do Leite (LQL) do Programa de Análise de Rebanhos Leiteiros do Paraná (PARLPR) da Associação Paranaense de Criadores de Bovinos da Raça Holandesa (APCBRH), fruto do convênio entre UFPR, McGill University e APCBRH. Foram analisadas 1.950.034 amostras de leite de tanques, obtidas no período de janeiro de 2005 a abril de 2012, em dez regiões do Estado. Os efeitos de meio, entre eles, mês e ano de análise, região e idade da amostra foram estudados. Estimaram-se os componentes de variância para a característica e analisou-se a distribuição das amostras nas classes de ECS. A média estimada e desvio-padrão do ECS foi de 4,83 ± 1,52. Todos os efeitos incluídos no modelo foram significativos (P<0,01). Ocorreram variações do ECS entre os meses de análise, destacando-se maior média para o mês de fevereiro (4,87). Da mesma forma, o ano de análise apresentou variações significativas, em que a maior média ocorreu no ano de 2010 (4,85 ± 0,0031). Ocorreram significativas variações também entre regiões, em que a maior média e seu erro-padrão foi de 5,28 ± 0,0023 (Centro Sul – Guarapuava). O efeito de idade da amostra apresentou redução de 4,75 para 4,66 do primeiro ao quinto dia. Das amostras analisadas, 65,95% apresentaram escore cinco ou mais.

Subclinical mastitis, diagnosed by elevated somatic cell count (SCC) in milk, is an important monitoring parameter of dairy cows’ udder health, related to their productivity and welfare. The present retrospective study aims to evaluate the epidemiology of subclinical mastitis (SCM) among the 37 herds of the Holstein Association of Greece participating in the milk quality recording system “ΙΩ”, from the start of 2015 until the end of 2018. The herds’ inclusion criterion was the consistency of monthly SCC recording throughout at least one full year between 2015 and 2018, with a maximum interval of 61 days between two consecutive monthly SCC recordings. Twenty-six herds (8630 cows) in 2015, thirty herds (10763 cows) in 2016, thirty herds (10945 cows) in 2017 and twenty-six herds (9597 cows) in 2018 were included. The prevalence of SCM and chronic SCM, the incidence rate of new cases of SCM, as well as the average somatic cell score and bulk tank milk SCC were determined for each of the four years. The results indicate a progressive deterioration of udder health from the onset of the cow’s productive life until culling. A year-over-year increase in the number of cows with subclinical mastitis led to an overall SCM prevalence of 34.5%, chronic SCM prevalence of 26.9% and a bulk tank milk SCC of 463000 cells/mL, in 2018. The average somatic cell score, a base 2logarithm of individual cow’s SCC, was found persistently above the subclinical mastitis indicative cut-off in all four years, with a peak in 2018. At herd level, the incidence rate of new SCM cases was 12 new cases / 100 cows / month; the highest incidence rate was observed in the early lactation stage group (1-60 days-in-milk), in all four years, reaching a peak of 31 new cases / 100 cows / month, in 2018. In 2018, prevalence of heifers’ SCM and chronic SCM was23.4% and 16.9%, respectively. Despite the adequate average 305-days milk yield (9608 kg in 2018), the results were indicative of poor udder health status, pointed out by reduced duration of cows’ productive life (less than 3 lactations)and lower milk quality (elevated SCC). The severity and wide spreading of subclinical mastitis in Greek dairy herds highlights the necessity of a national mastitis control program, aiming to improve the productive efficacy, management decisions accuracy and quality of produced milk.

This research studied somatic cell count in bulk tank milk samples (BTSCC) from the state of Paraná, Brazil, at the Milk Quality Laboratory of the Dairy Herd Analysis Service of the Holstein Association of Paraná, the result of technical and scientific cooperation between UFPR and McGill University of Canada. A total of 1,950,034 bulk tank milk samples from ten regions of the state of Paraná were analyzed between January 2005 and April 2012 and were studied using PROC GLM (SAS, version 9.3). Fixed effects were the month and year of analysis, region and age of the sample. Means and standard deviations of BTSCC were 553,519 ± 545,532 cells/ml, respectively. All fixed effects were statistically significant (P<0.01). Highest values for BTSCC are observed in the month of February (554,000 cells/ml ± 1.45) and lowest values in September (450,000 cells/ml ± 1.47). Similarly, the highest values were found in the year 2010 (567,000 cells/ml ± 1.16), the lowest BTSCC was found in 2012 (444,000 cells/ml ± 1.57). The region effect was also significant with the highest values found in the South Center/ Guarapuava (668,000 cells/ml ± 0.87) and the lowest in the Southwest/ Francisco Beltrão (359,000 cells/ml ± 2.00). Both variables showed a reduction of their values with increasing age of the sample, from 518,000 ± 1.08 to 472,000 cells/ml ± 2.14 between the first and the seventh day, for the BTSCC. Coefficient of variation for BTSCC was 96.10%. The R² was 0.39 for BTSCC.

Objectives were to investigate somatic cell counts (SCC) and total bacterial counts (TBC) in the raw bulk-tank milk of sheep flocks in Greece, to study factors potentially influencing increased SCC and TBC in the bulk-tank milk of sheep and to evaluate possible associations of SCC and TBC with milk content. Throughout Greece, 325 dairy sheep flocks were visited for collection of milk sampling for somatic cell counting, microbiological examination and composition measurement. Geometric mean SCC were 0.488 × 106 cells mL−1; geometric mean TBC were 398 × 103 cfu mL−1; 228 staphylococcal isolates were recovered form 206 flocks (63.4%). Multivariable analyses revealed annual incidence risk of clinical mastitis, age of the farmer and month into lactation period (among 53 variables) to be significant for SCC > 1.0 × 106 cells mL−1 and month into lactation period at sampling and availability of mechanical ventilators (among 58 variables) to be significant for TBC > 1500 × 103 cfu mL−1. Negative correlation of SCC with fat, total protein and lactose and positive correlation of SCC with added water were found. With SCC > 1.0 × 106 cells mL−1, significant reduction of protein content (2%) was observed, whilst in flocks with SCC > 1.5 × 106 cells mL−1, significantly lower annual milk production per ewe (42.9%) was recorded.

One hundred and seventy-seven (177) bulk tank milk samples were analyzed with a commercially available real-time polymerase chain reaction kit and 11 (6.21%), 41 (23.16%), and 58 (32.77%) tested positive for Mycoplasma bovis, Staphylococcus aureus, and Streptococcus agalactiae, respectively. Statistical analysis revealed a significant relationship between the presence of S. aureus and S. agalactiae. Enumeration of somatic cells was performed in the same samples by flow cytometry. The somatic cell counts were found higher in S. aureus and S. agalactiae positive samples. No association was found between M. bovis presence and somatic cells counts. Low internal assay control Ct values were found to be related with high somatic cell counts. Noticeably, this is the first report for the presence of M. bovis in Cyprus. Therefore, its presence was confirmed by bulk tank milk culture, conventional PCR, and next generation sequencing. Furthermore, M. bovis was typed with multilocus sequencing typing and was allocated to sequence type 29 (ST 29). Real-time PCR in bulk tank milk samples is a useful tool to detect mammary infections, especially for neglected pathogens such as M. bovis.

The aim of the study was to investigate the effects of season, cow cleanliness and milking routine on bacterial and somatic cell counts of bulk tank milk. A total of 22 dairy farms in Lombardy (Italy) were visited three times in a year in different seasons. During each visit, samples of bulk tank milk were taken for bacterial and somatic cell counts; swabs from the teat surface of a group of cows were collected after teat cleaning and before milking. Cow cleanliness was assessed by scoring udder, flanks and legs of all milking cows using a 4-point scale system. Season affected cow cleanliness with a significantly higher percentage of non-clean (NC) cows during Cold compared with Mild season. Standard plate count (SPC), laboratory pasteurization count (LPC), coliform count (CC) and somatic cell count, expressed as linear score (LS), in milk significantly increased in Hot compared with Cold season. Coagulase-positive staphylococci on teat swabs showed higher counts in Cold season in comparison with the other ones. The effect of cow cleanliness was significant for SPC, psychrotrophic bacterial count (PBC), CC and Escherichia coli in bulk tank milk. Somatic cell count showed a relationship with udder hygiene score. Milking operation routine strongly affected bacterial counts and LS of bulk tank milk: farms that accomplished a comprehensive milking scheme including two or more operations among forestripping, pre-dipping and post-dipping had lower teat contamination and lower milk SPC, PBC, LPC, CC and LS than farms that did not carry out any operation.

Milk somatic cell count (SCC) is the gold standard in diagnosis of subclinical mastitis, and is also an important parameter in quality programmes of dairy cooperatives. As routine SCC analysis is usually restricted to central laboratories, much effort has been invested in the search for alternative biomarkers of mastitis and milk quality, including the presence in the milk of the acute phase proteins (APP), haptoglobin (Hp) and serum amyloid A (SAA). The aim of this study was to investigate relationships between Hp, SAA and SCC in quarter, cow composite, and bulk tank milk samples. Cows (n=165), without any clinical signs of disease or abnormalities in the milk or udder, from three different dairy farms, were used. Cow composite milk samples from all cows delivering milk at the sampling occasion were taken once in each herd. In one of the farms, representative quarter milk samples (n=103) from 26 cows were also collected. In addition, bulk tank milk samples from 96 dairy farms were included in the study. Samples were analysed for Hp, SAA and SCC, and relationships between the parameters were evaluated at quarter, cow and tank milk levels using Chi-square analysis. Milk samples were categorized according to their SCC, and the presence, or no presence, of SAA and Hp, based on the detection limits of the screening methods (0·3 mg/l and 1·0 mg/l for SAA and Hp, respectively). Hp and SAA were found in milk at quarter, cow composite and bulk tank levels. A large proportion (53%) of the animals had detectable milk concentrations of APP, and SAA was detected more frequently, and at higher concentrations than Hp, regardless of sample type. SAA was detected in as many as 82% of the bulk tank milk samples. Significant relationships were found between Hp, SAA and SCC at quarter and cow composite milk levels, but only between SAA and SCC at bulk tank milk level. Detectable levels of APP were more common at high SCC.

AbstractThe objectives of this work were (a) to determine the presence of streptococci in samples from small ruminant dairy farms (bulk-tank milk and, where possible, teatcup swabs), (b) to investigate the potential adverse effects of streptococci on milk quality and (c) to investigate the importance of some husbandry factors for the isolation of streptococci. Bulk-tank milk samples and teatcups swab samples were examined bacteriologically for the presence of streptococci. Somatic cell counting and milk composition measurements were also performed. The husbandry factors present in each farm were assessed for potential associations with the isolation of streptococci. Streptococci were isolated from milk samples from 31.4% of sheep and 17.4% of goat farms and from 4.8% of sheep and 5.9% of goat teatcups. Streptococci were isolated more frequently from the upper part than the lower part of teatcups: 5.0% vs. 1.9%. Most isolates (57.9%) were identified as Streptococcus uberis. Most isolates (68.4%) were slime-producing; slime-production was more frequent among isolates from teatcups (83.3%) than from bulk-tank milk (55.0%). Somatic cell counts and milk composition did not differ between farms in which streptococci were or were not isolated. Machine-milking was associated with the isolation of streptococci from bulk-tank milk samples. The initial stage of the milking period (first two months) was found to be associated with the isolation of streptococci from milking machine teatcups in sheep farms only.
