Recent studies have reported collagen cross-linking after exposure to riboflavin followed by ultraviolet-A (UVA) exposure. This study is the first to investigate the effect of a riboflavin-containing primer on adhesive interface stability and dentinal matrix metalloproteinase activity. Human dentin was etched with 35% phosphoric acid, treated with 0.1% riboflavin, exposed to UVA for 2 min, and bonded with a two-step etch-and-rinse adhesive. Adhesive was applied to control specimens without riboflavin/UVA. Specimens were subjected to microtensile bond strength tests and pulled to failure after storage for 24 hrs, 6 mos, or 1 yr. Interfacial nanoleakage was evaluated by light and transmission electron microscopy. To investigate dentinal matrix metalloproteinase activity, we performed correlative zymographic assays on protein extracts obtained from phosphoric-acid-etched dentin powder with or without riboflavin/UVA treatment and XP Bond. Ultraviolet-activated riboflavin treatment increased the immediate bond strength to dentin at all aging intervals (p < 0.05 vs. control) and decreased interfacial nanoleakage in aged specimens (1 yr; p < 0.05). Zymograms revealed that riboflavin/UVA pre-treatment inhibited dentinal matrix metalloproteinase activity (especially MMP-9). In conclusion, dentinal collagen cross-linking induced by riboflavin/UVA increased immediate bond strength, stabilized the adhesive interface, and inhibited dentin matrix metalloproteinases, thereby increasing the durability of resin-dentin bonds.

Abstract To investigate the effect of the dimethyl sulfoxide combined with cross-linking agents on microtensile bond strength, silver nitrate penetration and in situ degree of conversion analysis of adhesives to the erosive dentin treatment with Cola-based soft drink. One hundred and sixty-six molars were assigned to 20 groups: (1) Treatment: Sound dentin; Erosive dentin; Erosive dentin treated with primer of dimethyl sulfoxide; Erosive dentin treated with DMSO primer containing proanthocyanidin and rivoflavin; (2) Adhesive systems: iBond Universal and Scotchbond Universal; and (3) adhesive strategy: etch-and-rinse or self-etch strategy. After restoration, specimens were sectioned into sticks to be tested. The data from microtensile bond strength (MPa), silver nitrate penetration (%) and in situ degree of conversion (%) were analyzed by (three- and two-factor ANOVA; Tukey's test α=5%). The application of dimethyl sulfoxide combined of not with cross-linkers improved all properties evaluated when compared to only erosive dentin treatment with Cola-based soft drink. However, only when dimethyl sulfoxide was combined to cross-linkers, the values of the microtensile bond strength, silver nitrate penetration and in situ degree of conversion in erosive dentin treatment with Cola-based soft drink was similar to sound dentin, for both adhesives and adhesive strategies. The application of dimethyl sulfoxide combined with the collagen cross-linking agent contributed to increasing the bond strength and degree of conversion in erosive lesion dentin, at the same time that significantly reduction of nanoleakage in this substrate.

ABSTRACT Aim The aim of this study was to evaluate the effect of chlorhexidine and ethanol application on the push-out bond strength and bond durability of fiber posts cemented with an etch-and-rinse adhesive system/resin cement to intraradicular dentin. Materials and methods Fifty-four bovine roots were shaped for the cementation of a fiberglass post and received the application of 37% phosphoric acid. They were then randomly divided into three groups, according to the type of dentin treatment (n = 18) performed: no treatment (control group), 100% ethanol, or 2% chlorhexidine. Next, the adhesive system (Adper Scotch Bond Multipurpose Plus, 3M ESPE) was applied to the dentin, according to the manufacturer's instructions. Glass fiber posts were cemented with dual resin cement (Rely X ARC, 3M ESPE). After 48 hours, the specimens were serially sectioned for push-out test analysis, providing two slices from each root third (cervical, medium and apical), one of which was tested immediately and the other stored in distilled water for 180 days. The data were analyzed with three-way analysis of variance (ANOVA) for repeated measures and Tukey's test at a 5% significance level. Results Intraradicular treatment with chlorhexidine yielded the highest bond strength means, followed by ethanol treatment. The control group presented the lowest bond strength means. Water storage exerted no effect on bond strength values. Conclusion Both chlorhexidine and ethanol improved push-out bond strength to intraradicular dentin, with the former providing the best results, regardless of the storage time. Clinical significance The application of 2% chlorhexidine or 100% ethanol may be an important step that can be taken to enhance bond strength of fiber posts to intraradicular dentin, when dual resin cements are used. Objective: The purpose of this study was to compare the immediate microtensile bond strength (µTBS) of two-step etch-and-rinse adhesive systems to the dentin of primary and permanent teeth. Study Design: Non-carious human teeth (12 primary molars and 12 premolars) were assigned to 3 groups according to the adhesive system. The adhesive systems were applied to flat superficial coronal dentin surfaces etched with phosphoric acid and composite resin blocks were built up. The teeth were sectioned to produce beam-shaped specimens with 0.81 mm2 cross-sectional area subjected to µTBS testing. µTBS data were analyzed statistically by ANOVA and Tukey’s test (a= 0.05). Results: The adhesive systems produced statistically similar mean µTBS to each other (p&gt;0.05) and no significant differences (p&gt;0.05) were found when the same material was applied to primary or permanent tooth dentin. The mean µTBS values (MPa) obtained were: Prime& Bond NT: 41.7±14.4 (permanent) and 40.8±13.4 (primary); Single Bond: 42.9±8.6 (permanent) and 41.4±11.9 (primary); Excite DSC: 46.3±11.3 (permanent teeth) and 43.4±12.0 (primary). Conclusion:There was no difference in the immediate µTBS of two-step etch-and-rinse adhesive systems when applied to the dentin of primary and permanent teeth.

SUMMARY The aim of this study was to evaluate the effect of a chlorhexidine and/or ethanol application on the bond strength of an etch-and-rinse, hydrophobic adhesive system either under in vitro aging or in situ cariogenic challenge. The dentin surface of 36 human third molars were flattened and allocated into four groups to be treated with chlorhexidine, ethanol, or chlorhexidine + ethanol or left unexposed to any solution (control) (n=9). Then, a resin composite restoration was made on the dentin surface and longitudinal sticks were obtained. Sticks from each tooth were assigned to three test conditions: stored in water in vitro for 24 hours, stored in water in vitro for 6 months, or worn in situ for 14 days. During in situ wear time, a high-cariogenic challenge condition was simulated. Specimens were tested for microtensile bond strength (μTBS). Multivariate analysis of variance and Tukey's test showed that chlorhexidine, ethanol, or chlorhexidine + ethanol did not affect the μTBS. The in vitro μTBS values were significantly lower for the specimens stored for 6 months than for those stored for 24 hours. Intermediate μTBS values were shown by the specimens worn in situ. Thus, use of chlorhexidine and/or ethanol was incapable of containing the degradation at the bond interface in the in vitro model. The in situ model was capable of reducing bond strength similarly to the in vitro/6 months model. Despite this, the in situ bond strength was still similar to that of the in vitro/24-hour model.

<p class="Corpo"><strong>Objective</strong>: the aim of the present study was to evaluate the influence of adding different concentrations of chitosan to an experimental two-step etch-and-rinse adhesive system on the bond strength and failure mode to dentin. <strong>Material</strong> <strong>and</strong> <strong>Methods</strong>: thirty-two flat dentin surfaces were obtained from extracted human third molars and divided into four groups  (n=8) for application of the adhesive systems: AD - conventional two-step adhesive system (Adper Single Bond 2); EXP – experimental two-step etch-and-rinse adhesive system; Chi0.2% - EXP with addition of 0.2% Chitosan; Chi0.5% - EXP with addition of 0.5% Chitosan. Resin composite build-ups were made and the composite/dentin specimens were sectioned to obtain rectangular beams with a bond area of approximately 1mm<sup>2</sup>. After 24 hours, the sticks were submitted to microtensile bond strength tests in a universal test machine. The fracture pattern was evaluated under a stereoscopic loupe at 40X magnification. <strong>Results</strong>: one-way analysis of variance showed that the type of adhesive system had no significant effect on the bond strength values (p = 0.142), showing the mean bond strength values (standard deviation), in MPa, for the groups as follows: AD=20.1 (5.4); EXP=16.6 (2.3); Chi0.2%=16.1 (2.8); Chi0.5%=16.9 (2.3). In all the groups there was predominance of cohesive fractures in dentin, representing 68 to 82% of the failure modes. <strong>Conclusion</strong>: the addition of 0.2 or 0.5% of chitosan had no influence on the bond strength and failure mode of an experimental two-step etch-and-rinse adhesive system to dentin.</p><p class="Corpo"><strong>Keywords</strong></p><p class="Corpo">Chitosan; Dental Adhesives; Failure Mode; Microtensile Bond Strength.</p>

Background. Contamination of dentin surface is one of the common problems in restorative dentistry. The aim was to investigate the effects of different surface contaminators on the dentin shear bond strength (SBS) of universal adhesive system (UAS) applied in etch-and-rinse (ER) and self-etch (SE) strategies. Methods. One hundred forty-four maxillary anterior sound human teeth were divided into six groups based on the types of surface contaminators: no surface contaminator (control) and experimental groups contaminated with blood, saliva, aluminium chloride (ALC), ferric sulphate (FS), and caries disclosing agent (CDA). Then, each group was further subdivided into two, based on the application strategy of UAS (ER and SE). After applying the adhesive according to the manufacturer’s instructions, and bonding cylindrical composite samples, the SBS was measured. The data were analysed using two-way ANOVA, Tukey’s HSD test and t test (P<0.05). Results. The SBS in all contaminated groups, except for CDA, was significantly lower in both ER and SE strategies compared to control group (P<0.05). A comparison between the application strategies revealed that ER and SE were only significantly different in the FS contaminated group (P<0.05). Conclusion. All tested contaminators, except CDA, significantly decreased SBS of UAS in both ER and SE strategies.
