Background: Good laboratory practices in terms of biosecurity and biosafety are essential for the protection of laboratory personnel, the populations and the environment. If not implemented, the risk of pathogen spreading from the laboratory is conceivable. In addition, in current African geopolitical context, the reliable practices such as the protection, control and tracking of valuable biological material will prevent their loss, theft, uncontrolled access and potential misuse. WAHO, with the support of KfW is implementing trainings of trainers on biosecurity in the ECOWAS region in a project called PROALAB. PROALAB team realized that many partners were aiming at implementing in parallel quite similar non internationally certified programs in the ECOWAS region. Therefore, PROALAB decided to organize a meeting of key stakeholders in order to pool efforts, avoid duplication, and harmonize strategies and procedures. Objective: Develop a harmonized strategy and harmonized procedures for the training of health professionals in biosecurity/biosafety and biohazard risk management in ECOWAS member countries to pass internationally certified biosafety and biosecurity examinations. Methods: Programs of each of the institutions were presented and discussed. A regional program on biosecurity/biosafety training was elaborated during group working and a regional program of certification of trainers was discussed and validated. A work plan and timeline were developed through group working. Results: In the ECOWAS region, some countries are more advanced in biosecurity training. Training is generally organized by national biosecurity associations with the support of the IFBA or other partners (WHO, FMx, PHE, etc.). Côte d’Ivoire, Nigeria and Mali seem more advanced and could be solicited to organize training for less-advanced countries. The IFBA certification covers five domains: (1) Biorisk management, (2) biosecurity, (3) biosafety cabinet, (4) biological waste management and (5) biocontainment facility. The technical working group decided consensually that basic training for biosecurity managers in the laboratories should cover the first two domains. More advanced experts advising governments should be trained in the other domains progressively. Progressive training could be built on the 3 levels Nigerian/Ivoirian system of “basic-advanced-consolidated (or expert)” system. Existing module from WHO, FMx, IPD, IPCI and other ECOWAS countries should be adapted to match the international certification needs. Practical training should also be organized in the laboratory by the same institutions that will be allowed to deliver a practical certificate on behalf of WAHO. Conclusion: With the extension of laboratory activities in the context of strengthening the surveillance programs in the ECOWAS region, more expertise on biosecurity is needed. With the support of national biosecurity associations, WAHO could play a role in harmonizing reinforcing training on biosecurity and legislation.

Objetivos:caracterizar os acidentes de trabalho com material biológico de acordo com o tipo de exposição e descrever o perfil dos profissionais dos serviços de saúde que sofreram esses acidentes. Método:estudo epidemiológico, transversal e retrospectivo.Fizeram parte da pesquisa 1.269 notificações de acidentes de trabalho com material biológico, os quais foram coletados através do Sistema de Informação de Agravos de Notificação (SINAN) e analisados por meio de estatística descritiva e inferencial. Para a análise de associação entre as variáveis selecionadas, utilizou-se o teste qui quadrado e a Correlação de Pearson.Resultados:identificou-se que o tipo de exposição tem relação direta com a circunstância do acidente, com o tempo de trabalho e com o agente causador do acidente, com predominância do sexo feminino, em adultos jovens, com escolaridade em nível médio, atuando principalmente na área da enfermagem, com experiência profissional igual ou inferior a 5 anos. Conclusão:ressalta-se a importância das boas práticas, como o uso de equipamentos de proteção individual e esquema vacinal, e o acompanhamento do profissional pós-exposição a acidentes de trabalho com material biológico, com intuito primordial de promover a saúde do trabalhador e a gestão de riscos nas instituições.

Objective: The aim of the present study was to evaluate the knowledge, prevalence and seroconversion to hepatitis B in health professionals. Methods: A descriptive and quantitative research was conducted through the application of questionnaires, examination analysis and immunization records of employees of a private health clinic in Cascavel, Paraná. Results: Of the 96 employees, 87 (90.6%) had anti-HBs reagent verified by the exam. Of these, the majority of employees were female (91.6%), had knowledge of the hepatitis B virus (HBV) (96.3%) and were between 31 and 40 years old (47.4%). Also, they had not had an accident with biological material (86.5%), had no HBV carrier (87.4%), or had had contact with carriers of the infection (90.6%). Conclusion: Although the most effective prevention method against HBV is vaccination, approximately 10% of the employees did not have antibodies detected by the test. In addition, some individuals showed little knowledge about hepatitis B, which is of concern, as they are health professionals who are at risk of acquiring infections due to contact with patients and biological materials.

Color contrasting of 1 to 2 micron sections of plastic embedded biological material is an important adjunct to electron microscopy. The procedures in general use today are simple and rapid giving monochromatic results, e.g., toluidine blue. Although many di- and polychromatic histologic staining techniques have been modified to obtain a counterstaining effect with plasticembedded tissue, the methods are usually undesirable for routine work because they are time consuming, complicated and often defy good reproducibility.

During the last years it was shown that also by means of cryo-ultra-microtomy a good preservation of substructural details of biological material was possible. However the specimen generally was prefixed in these cases with aldehydes.Preparing ultrathin frozen sections of chemically non-prefixed material commonly was linked up to considerable technical and manual expense and the results were not always satisfying. Furthermore, it seems to be impossible to carry out cytochemical investigations by means of treating sections of unfixed biological material with aqueous solutions.We therefore tried to overcome these difficulties by preparing yeast cells (S. cerevisiae) in the following manner:

For electron microscope techniques such as ferritin-labeled antibody staining it would be advantageous to have available a simple means of thin sectioning biological material without subjecting it to lipid solvents, impregnation with plastic monomers and their subsequent polymerization. With this aim in view we have re-examined the use of protein as an embedding medium. Gelatin which has been used in the past is not very satisfactory both because of its fibrous nature and the high temperature necessary to keep its solutions fluid. We have found that globular proteins such as the serum and egg albumins can be cross-linked so as to yield blocks which are suitable for ultrathin sectioning.

The successful application of ferritin labeled antibodies (F-A) to ultrathin sections of biological material has been hampered by two main difficulties. Firstly the normally used procedures for the preparation of material for thin sectioning often result in a loss of antigenicity. Secondly the polymers employed for embedding may non-specifically absorb the F-A. Our earlier use of cross-linked polyampholytes as embedding media partially overcame these problems. However the water-soluble monomers used for this method still extract many lipids from the material.

Zusammenfassung. Hintergrund: Die Corona-Pandemie führte zu einer starken Beanspruchung von Health Professionals, deren allgemeine berufliche Situation mit einem hohem Belastungserleben verbunden ist. Quantitative Daten zum Belastungserleben der im Gesundheitswesen Tätigen in der Corona-Pandemie und mögliche Auswirkungen auf das Arbeitsengagement fehlen bis dato für Deutschland. Methode: Mittels einer Querschnittsbefragung wurden das Stresserleben, die Sorge um die Gesundheit und das Arbeitsengagement von Health Professionals deutschlandweit erhoben. Das „snapshot survey“ nutzte neben selbst entwickelten Fragen Items eines validierten Instrumentes zur Erfassung des Arbeitsengagements. Über soziale Medien wurden in Form einer Gelegenheitsstichprobe Health Professionals zur Teilnahme eingeladen. Es konnten 1168 gültige Fälle ausgewertet werden. Die Teilnehmenden waren mehrheitlich Pflegende (80 %, n = 855). Ergebnisse: Es zeigt sich, dass Health Professionals mit Direktkontakt zu COVID-19-Erkrankten im Gegensatz zu Befragten ohne Kontakt ein höheres Stresslevel angeben (MW = 3,81, SD = 1,09 vs. MW = 3,44, SD = 1,12, t(1062) = 5,40, p < 0,001; ε = 0,33), sich am meisten Sorgen um die Gesundheit ihrer Angehörigen sowie von Freundinnen und Freunden machen und dass diese Sorgen größer als bei Health Professionals ohne Kontakt sind (MW = 4,45, SD = 0,84 vs. MW = 4,19, SD = 0,94, t(1062) = 4,74, p < 0,001; ε = 0,29). Die Korrelationsanalysen (r = –0,182, p < 0,001) und die multiple Regressionsanalyse (β = –0,182, p < 0,001) ergaben, dass das Arbeitsengagement mit steigendem, pandemiebedingtem Stress abnimmt. Diskussion: Es zeigen sich theoriekonforme Zusammenhänge zwischen Anforderungen, Stresserleben und Arbeitsengagement. Im Vergleich zu früheren Studien ist das Arbeitsengagement in der Stichprobe gering. Zur Reduktion psychischer Belastungsfaktoren in einer Pandemie liegen vielfältige Empfehlungen vor, die nun auch in der Breite für Deutschland zur Anwendung kommen sollten. Hierzu zählen beispielsweise pandemiespezifische Präventionspläne oder die Schaffung eines sanktionsfreien Arbeitsumfeldes.
